© 2020 by the authors. This study aimed to evaluate the performance of a commercial protease (Novo-Pro D (NPD)), both in soluble and immobilized forms, in the hydrolysis of proteins (using casein as model protein). Immobilization of the protease NPD on 6% agarose activated with glyoxyl groups for 24 h at 20 °C and pH 10.0 allowed preparing immobilized biocatalyst with around 90% immobilization yield, 92% recovered activity versus small substrate, and a thermal stability 5.3-fold higher than the dialyzed soluble enzyme at 50 °C and pH 8.0. Immobilization times longer than 24 h lead to a decrease in the recovered activity and did not improve the biocatalyst stability. At 50 °C and pH 6.5, the immobilized NPD was around 20-fold more stable than the dialyzed soluble protease. Versus casein, the immobilized NDP presented a 10% level of activity, but it allowed hydrolyzing casein (26 g/L) at 50 °C and pH 6.5 up to a 40% degree of hydrolysis (DH) after 2 h reaction, while under the same conditions, only a 34% DH was achieved with soluble NPD. In addition, the immobilized NPD showed good reusability, maintaining the DH of casein for at least ten 2h-reaction batches. This research was funded by National Council for Scientific and Technology Development (CNPq), grant numbers 141487/2016-0, 422131/2016-4; Coordenação de Aperfeiçoamento de Pessoal de Nível Superior – Brazil (CAPES), Finance Code 001 and Spanish Ministerio de Ciencia e Innovación, grant number CTQ2017-86170-R.Peer reviewe

This study was carried out for screening the phytase production using agroindustrial residues as substrate for solid state fermentation (SSF) and several fungal strains. High phytase production was observed for most of the microorganisms studied, as well as very different biochemical characteristics, including activities at specific pH values. The phytases showed very distinct behavior of optimum and stability pH, optimal values of temperature and stability. A. niger showed the higher values of enzyme activity. Lyophilized enzyme produced by A. niger reached 8,575 U/g and yield of 826,096.50 U/Kg of substrate when fermented in wheat bran. Therefore, the substrate as well as the microorganism strain can affect the biochemical character of the enzyme produced. The high phytase activity and very distinct characteristics established, plus the low cost of substrates, make these fungal phytases potential alternatives for biotechnological industry.

Fungal strains were screened for lipase producing activities and 10 strains were classified as good producers. Aspergillus sp., Fusarium sp., and Penicillium sp. exhibited the highest activities when fermented in wheat bran (WB) and soybean bran (SB). No fungal growth was observed using sugarcane bagasse (CB). An experimental design was applied to incorporate CB into the fermentation process for lipase production by Aspergillus sp. and Penicillium sp., and to evaluate the best moisture content for the substrate. Strains studied achieved maximum lipase activities with 25% CB combined with 75% WB or SB at 40% moisture content. The highest lipase activities were observed for WB and SB, and for SB combined with CB using Aspergillus sp. Fermentation of 96 h was the optimum period for enzyme production.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP
