    Formation of keratinocyte multilayers on filters under airlifted or submerged culture conditions in medium containing calcium, ascorbic acid, and keratinocyte growth factor.

    Three-dimensional (3D) cell culture is a powerful in vitro technique to study the stratification and differentiation of keratinocytes. However, culture conditions, including culture media, supplements, and scaffolds (e.g., collagen gels with or without fibroblasts), can vary considerably. Here, we evaluated the roles of calcium, L-ascorbic acid phosphate magnesium salt n-hydrate (APM), and keratinocyte growth factor (KGF) in a chemically defined medium, EpiLife, in 3D cultures of primary human epidermal keratinocytes directly plated on polycarbonate filter inserts under airlifted or submerged conditions. Eight culture media containing various combinations of these three supplements were examined. Calcium was necessary for the stratification and differentiation of keratinocytes based on the localization of keratins and involucrin. However, the localization patterns of keratins and integrin β4 were partially disrupted and Ki67-positive basal cells almost disappeared 3 weeks after airlift. The addition of KGF, but not APM, prevented these changes. Further addition of APM markedly improved the tissue architecture, including basal cell morphology and the appearance of keratohyalin granules and localized involucrin in the upper suprabasal cells, even after 1 week. Although the submerged culture also formed cornified epithelium-like multilayers, involucrin was localized in the cornified layer, where nuclei were often found. Based on these results, it is most effective to culture keratinocytes at the air-liquid interface in EpiLife medium supplemented with calcium, APM, and KGF to form well-organized and orthokeratinized multilayers as skin analogues.福岡歯科大学2016年

    Functional shoulder radiography with use of a dynamic flat panel detector

    Our purpose in this study was to develop a functional form of radiography and to perform a quantitative analysis for the shoulder joint using a dynamic flat panel detector (FPD) system. We obtained dynamic images at a rate of 3.75 frames per second (fps) using an FPD system. Three patients and 5 healthy controls were studied with a clinically established frontal projection, with abduction of the arms. The arm angle, glenohumeral angle (G-angle), and scapulothoracic angle (S-angle) were measured on dynamic images. The ratio of the G-angle to the S-angle (GSR) was also evaluated quantitatively. In normal subjects, the G-angle and S-angle changed gradually along with the arm angle. The G-angle was approximately twice as large as the S-angle, resulting in a GSR of 2 throughout the abduction of the shoulder. Changes in G-angle and S-angle tended to be irregular in patients with shoulder disorders. The GSR of the thoracic outlet syndrome, recurrent dislocation of the shoulder joint, and anterior serratus muscle paralysis were 3-7.5, 4-9.5, and 3.5-7.5, respectively. The GSR of the anterior serratus muscle paralysis improved to approximately 2 after orthopedic treatment. Our preliminary results indicated that functional radiography by FPD and computer-aided quantitative analysis is useful for diagnosis of some shoulder disorders, such as the thoracic outlet syndrome, recurrent dislocation of the shoulder joint, and anterior serratus muscle paralysis. The technique and procedures described comprise a simple, functional shoulder radiographic method for evaluation of the therapeutic effects of surgery and/or rehabilitation. © 2014 The Author(s)

    The Outcome of Cochlear Implantation for Mitochondrial Disease Patients With Syndromic Hearing Loss.

    【Objectives】To evaluate the outcome and to confirm the validity of cochlear implantation for syndromic deafness in patients with mitochondrial disease. Study Design: Retrospective case review. 【Setting】Tertiary referral center. 【Patients】We reviewed medical charts of 367 cochlear implantation cases at Kyoto University Hospital between 1987 and 2012. We identified five patients with syndromic mitochondrial disease who underwent cochlear implantation surgery. The mean age of the patients (four women and one man) when they underwent surgeries was 44.4 years (range, 30–64 yr; median, 41 yr). 【Interventions】Therapeutic and rehabilitative. 【Main Outcome Measure】In four of five patients, speech perception performance was measured using Japanese vowels, consonant-vowel syllables, and short sentences. 【Results】Only 1.4% (5 of 367) of cochlear implantation cases at Kyoto University Hospital underwent cochlear implantation surgery because of syndromic mitochondrial diseases. Four of those patients showed significantly improved speech perception outcomes, and the beneficial effects of the intervention continued long after surgery. One patient could not perform speech perception test presumably because of poor cognitive function. 【Conclusion】Mitochondrial disease patients who underwent cochlear implantation surgery sustained gains in hearing performance even long after surgery. A single patient showed poor postoperative speech perception associated with cognitive problems. Cochlear implantation for mitochondrial disease patients seems to be a viable treatment option in the absence of significant cognitive impairment

    Keratinocytes take up serum-derived retinol (vitamin A) and metabolize it to all-trans-retinoic acid (atRA), which binds to the nuclear retinoic acid receptor (RAR). We previously reported that serum-affected keratinocyte differentiation and function; namely, it inhibited keratinization, decreased loricrin (LOR) and claudin (CLDN) 1 expression, increased keratin (K) 4 and CLDN4 levels, and reduced paracellular permeability in three-dimensional (3D) cultures of mouse keratinocytes (COCA). Contrarily, RAR inhibition reversed these changes. Here, we aimed to examine whether atRA exerted the same effects as serum, and whether it was involved in the differential oral mucosa keratinization among animal species. Porcine oral mucosal keratinocytes, which form non-keratinized epithelium in vivo, established keratinized epithelium in 3D cultures. Both mouse and porcine sera induced non-keratinized epithelium at 0.1% in COCA 3D cultures. Although atRA caused the same changes as serum, its effective concentration differed. atRA inhibited keratinization at 0.1 nM and 1 nM in porcine or human keratinocytes and COCA, respectively. Furthermore, atRA upregulated CLDN7 in the cytoplasm but not in cell-cell contacts. These atRA-induced changes were reverted by RAR inhibition. The results indicate that serum-induced changes are probably due to the effect of serum-derived atRA, and that mouse keratinocytes require higher atRA concentrations to suppress keratinization than porcine and human keratinocytes. We propose that the lower susceptibility of mouse keratinocytes to atRA, rather than a lower retinol concentration, is a possible reason for the keratinization of mouse oral mucosal epithelium.福岡歯科大学2019年

    The AKARI far-infrared all-sky survey maps

    We present a far-infrared all-sky atlas from a sensitive all-sky survey using the Japanese AKARI satellite. The survey covers >99% of the sky in four photometric bands centred at 65 μm, 90 μm, 140 μm, and 160 μm, with spatial resolutions ranging from 1' to 1".5. These data provide crucial information on the investigation and characterisation of the properties of dusty material in the interstellar medium (ISM), since a significant portion of itsenergy is emitted between ∼ 50 and 200μm. The large-scale distribution of interstellar clouds, their thermal dust temperatures, and their column densities can be investigated with the improved spatial resolution compared to earlier all-sky survey observations. In addition to the point source distribution, the large-scale distribution of ISM cirrus emission, and its filamentary structure, are well traced. We have made the first public release of the full-sky data to provide a legacy data set for use in the astronomical communit

    本研究は, 「21世紀型の教科学力」の新たな観点としての「協同的創造力」の育成をめざして, 自分たちで新たな文化を創造する子どもを育てる協同的創造学習のあり方について実証的に研究を進め, 単元モデルと評価方法を開発することを目的としている。そこで, 教科学習を「協同的創造学習」としてとらえ直すとともに, 中学校での従来の選択教科の時間に加えて, 小学校第5・6学年合同の選択教科の時間を新設して「協同的創造力」を特化して育むことにし, 本年度は, 選択教科の単元モデルの充実・改善と評価方法の確立に取り組んだ。その結果, 選択教科において, これまで開発した単元モデルをより充実させたり, 新たな単元モデルを開発したりすることができた。また, 評価の観点を整理し, 子どもの意識調査やカリキュラム評価に継続して取り組むことによって, 子どもの思いを汲み取り単元を見直していくことができた。今後も必修教科と選択教科のつながりや関連性, 各学年の系統性を整理するとともに, 協同的創造力育成の手だてを整理し, 来年度に向けて, これまで培ったものを生かす新たな学習開発を模索していきたいと考えている
