    Plume mapping and isotopic characterisation of anthropogenic methane sources

    Methane stable isotope analysis, coupled with mole fraction measurement, has been used to link isotopic signature to methane emissions from landfill sites, coal mines and gas leaks in the United Kingdom. A mobile Picarro G2301 CRDS (Cavity Ring-Down Spectroscopy) analyser was installed on a vehicle, together with an anemometer and GPS receiver, to measure atmospheric methane mole fractions and their relative location while driving at speeds up to 80 kph. In targeted areas, when the methane plume was intercepted, air samples were collected in Tedlar bags, for delta C-13-CH4 isotopic analysis by CF-GC-IRMS (Continuous Flow Gas Chromatography-Isotope Ratio Mass Spectrometry). This method provides high precision isotopic values, determining delta C-13-CH4 to +/- 0.05 per mil. The bulk signature of the methane plume into the atmosphere from the whole source area was obtained by Keeling plot analysis, and a delta C-13 -CH4 signature, with the relative uncertainty, allocated to each methane source investigated. Both landfill and natural gas emissions in SE England have tightly constrained isotopic signatures. The averaged delta C-13-CH4 for landfill sites is -58 +/- 3%o. The delta C-13-CH4 signature for gas leaks is also fairly constant around -36 +/- 2 parts per thousand, a value characteristic of homogenised North Sea supply. In contrast, signatures for coal mines in N. England and Wales fall in a range of -51.2 +/- 0.3 parts per thousand to 30.9 +/- 1.4 parts per thousand, but can be tightly constrained by region. The study demonstrates that CRDS-based mobile methane measurement coupled with off-line high precision isotopic analysis of plume samples is an efficient way of characterising methane sources. It shows that iiotopic measurements allow type identification, and possible location of previously unknown methane sources. In modelling studies this measurement provides an independent constraint to determine the contributions of different sources to the regional methane budget and in the verification of inventory source distribution.     Investigating the intrinsic noise limit of Dayem bridge NanoSQUIDs

    NanoSQUIDs made from Nb thin films have been produced with nanometre loop sizes down to 200 nm, using weak-link junctions with dimensions less than 60 nm. These composite (W/Nb) single layer thin film devices, patterned by FIB milling, show extremely good low-noise performance ∼170 nΦ0 at temperatures between 5 and 8.5 K and can operate in rather high magnetic fields (at least up to 1 T). The devices produced so far have a limited operating temperature range, typically only 1–2 K. We have the goal of achieving operation at 4.2 K, to be compatible with the best SQUID series array (SSA) preamplifier available. Using the SSA to readout the nanoSQUIDs provides us with a means of investigating the intrinsic noise of the former. In this paper we report improved white noise levels of these nanoSQUIDs, enabling potential detection of a single electronic spin flip in a 1-Hz bandwidth. At low frequencies the noise performance is already limited by SSA preamplifier noise

    Two major ruminant acute phase proteins, haptoglobin and serum amyloid A, as serum biomarkers during active sheep scab infestation

    Two ruminant acute phase proteins (APPs), haptoglobin (Hp) and serum amyloid A (SAA), were evaluated as serum biomarkers (BMs) for sheep scab–a highly contagious ectoparasitic disease caused by the mite Psoroptes ovis, which is a major welfare and production threat worldwide. The levels of both APPs increased in serum following experimental infestation of sheep with P. ovis, becoming statistically significantly elevated from pre-infestation levels at 4 weeks post-infestation. Following successful treatment of infested sheep with an endectocide, Hp and SAA serum levels declined rapidly, with half lives of less than 3 days. In contrast, serum IgG levels which specifically bound the P. ovis-derived diagnostic antigen Pso o 2 had a half-life of 56 days. Taking into account pre-infestation serum levels, rapidity of response to infestation and test sensitivity at the estimated optimum cut-off values, SAA was the more discriminatory marker. These studies illustrated the potential of SAA and Hp to indicate current sheep scab infestation status and to augment the existing Pso o 2 serological assay to give disease-specific indications of both infestation and successful treatment

    Study of the single body yawed-wing aircraft concept

    Areas relating to the development and improvement of the single-fuselage, yawed-wing transonic transport concept were investigated. These included: (1) developing an alternate configuration with a simplified engine installation;(2) determining a structural design speed placard that would allow the engine-airframe match for optimum airplane performance; and (3) conducting an aeroelastic stability and control analysis of the yawed-wing configuration with a flexible wing. A two-engine, single-fuselage, yawed-wing configuration was developed that achieved the Mach 1.2 design mission at 5560 km (3000 nmi) and payload of 18,140 kg (40,000 lb) with a gross weight of 217,700 kg (480,000 lb). This airplane was slightly heavier than the aft-integrated four-engine configuration that had been developed in a previous study. A modified structural design speed placard, which was determined, resulted in a 6% to 8% reduction in the gross weight of the yawed-wing configurations. The dynamic stability characteristics of the single-fuselage yawed-wing configuration were found to be very dependent on the magnitude of the pitch/roll coupling, the static longitudinal stability, and the dihedral effect

    Clostridium perfringens-associated ne- crotic enteritis causes significant economic losses. The objective of this study was to evaluate the effect of bismuth citrate, lactose, and organic acid on the devel- opment of necrotic enteritis in broilers. The first study was a dose response that evaluated bismuth citrate at 50, 100, or 200 ppm on bacterial intestinal coloniza- tion and lesion development associated with our C. perfringens challenge model. The second study evalu- ated bismuth citrate, lactose, and citric acid on intes- tinal pH and lesion development. For the third study, we determined if lactose would enhance the efficacy of bismuth citrate against intestinal colonization and lesion development associated with C. perfringens. In study 1, intestinal lesion scores at the 50, 100, and 200 ppm bismuth citrate treatment level were reduced (P ≤ 0.05) when compared with the birds fed 0 ppm bismuth citrate. Intestinal C. perfringens colonization of the 100 and 200 ppm bismuth citrate treatment group was sig- nificantly reduced when compared with birds fed 0 ppm bismuth citrate. In study 2, we found no significant differences in lesion development, after C. perfringens challenge, between birds fed 100 ppm bismuth citrate or fed a combination of 100 ppm bismuth citrate with dietary lactose or citric acid relative to the controls. The intestinal pH of birds fed 100 ppm bismuth cit- rate or fed a combination of 100 ppm bismuth citrate with dietary lactose or citric acid was not significantly reduced when compared with the controls. However, a significant reduction in pH was observed in birds fed a combination of 100 ppm bismuth citrate and lactose relative to the negative controls. In study 3, a decrease (P ≤ 0.05) in intestinal lesion scores occurred in birds fed lactose with 100 ppm bismuth citrate, compared with the positive controls. There were no significant differences in intestinal bacterial colonization. These preliminary data suggest that bismuth citrate may re- duce intestinal lesion development and C. perfringens colonization in broilers infected with necrotic enteritis

    The stellar mass function of star-forming galaxies and the mass-dependent SFR function since z = 2.23 from HiZELS

    We explore a large uniformly selected sample of Hα selected star-forming galaxies (SFGs) at z = 0.40, 0.84, 1.47, 2.23 to unveil the evolution of the star formation rate (SFR) function and the stellar mass function. We find strong evolution in the SFR function, with the typical SFR of SFGs declining exponentially in the last 11 Gyr as SFR*(T[Gyr]) = 104.23/T + 0.37 M⊙ yr−1, but with no evolution in the faint-end slope, α ≈ −1.6. The stellar mass function of SFGs, however, reveals little evolution: α ≈ −1.4, M* ∼ 1011.2 ± 0.2 M⊙ and just a slight increase of ∼2.3× in Φ* from z = 2.23 to z = 0.4. The stellar mass density within SFGs has been roughly constant since z = 2.23 at ∼107.65 ± 0.08 M⊙ Mpc−3, comprising ≈100 per cent of the stellar mass density in all galaxies at z = 2.23, and declining to ≈20 per cent by z = 0.40, driven by the rise of the passive population. We find that SFGs with ∼1010.0 ± 0.2 M⊙ contribute most to the SFR density (ρSFR) per d log10M, and that there is no significant evolution in the fractional contribution from SFGs of different masses to ρSFR or ρSFR(d log10M)−1 since z = 2.23. Instead, we show that the decline of SFR* and of ρSFR is primarily driven by an exponential decline in SFRs at all masses. Our results have important implications not only on how SFGs need to be quenched across cosmic time, but also on the driver(s) of the exponential decline in SFR* from ∼66 M⊙ yr−1 to 5 M⊙ yr−1 since z ∼ 2.23

    Mixed culture of commensal bacteria reduces E. coli in nursery pigs

    The purpose of the present study was to use field trials to evaluate the efficacy of a porcine-derived, defined culture (RPCF) of commensal bacteria for prevention of clinical disease from enterotoxigenic strains of Escherichia coli in weaned pigs. Neonates (\u3c 24 h old) were orally administered RPCF and were monitored throughout the post-weaning nursery period on five geographically separated farms. The farms had a history of high mortality from F-18 strains of E. coli. RPCF-treated pigs had reduced mortality, morbidity, and medication costs from E. coli compared to untreated pigs. Although experimental, RPCF may become an effective control procedure for enterotoxigenic E. coli
