The HSV-1 Latency-Associated Transcript Functions to Repress Latent Phase Lytic Gene Expression and Suppress Virus Reactivation from Latently Infected Neurons
open access articleHerpes simplex virus 1 (HSV-1) establishes life-long latent infection within sensory neurons, during which viral lytic gene expression is silenced. The only highly expressed viral gene product during latent infection is the latency-associated transcript (LAT), a non-protein coding RNA that has been strongly implicated in the epigenetic regulation of HSV-1 gene expression. We have investigated LAT-mediated control of latent gene expression using chromatin immunoprecipitation analyses and LAT-negative viruses engineered to express firefly luciferase or β-galactosidase from a heterologous lytic promoter. Whilst we were unable to determine a significant effect of LAT expression upon heterochromatin enrichment on latent HSV-1 genomes, we show that reporter gene expression from latent HSV-1 genomes occurs at a greater frequency in the absence of LAT. Furthermore, using luciferase reporter viruses we have observed that HSV-1 gene expression decreases during long-term latent infection, with a most marked effect during LAT-negative virus infection. Finally, using a fluorescent mouse model of infection to isolate and culture single latently infected neurons, we also show that reactivation occurs at a greater frequency from cultures harbouring LAT-negative HSV-1. Together, our data suggest that the HSV-1 LAT RNA represses HSV-1 gene expression in small populations of neurons within the mouse TG, a phenomenon that directly impacts upon the frequency of reactivation and the maintenance of the transcriptionally active latent reservoir

Concepciones alternativas de los estudiantes de la Escuela Normal Superior Distrital María Montessori del grado 902 en torno al concepto de ecosistema
Alternative concepts are important for the teaching-learning process since they should be taken as a basis for the construction of knowledge in the classroom. The present study was developed with the objective of identifying and categorizing the alternative conceptions of 30 students of grade 902 of the Escuela Normal Superior Distrital María Montessori de Bogotá around the concept of ecosystem from the application of a diagnostic instrument; the alternative conceptions of the students were classified in levels 0, 1, 2 and 3 of complexity. Fifty percent of the definitions were located in level 1 of complexity (additive level) and only 10% of the definitions were identified in level 3 of complexity (complex organizational level). The usefulness of images for the detection of alternative conceptions is also identified.Las concepciones alternativas son importantes para el proceso de enseñanza-aprendizaje ya que deben tomarse como base para la construcción de conocimientos en el aula. El presente estudio se desarrolló con el objetivo de identificar y categorizar las concepciones alternativas de 30 estudiantes del grado 902 de la Escuela Normal Superior Distrital María Montessori de Bogotá alrededor del concepto de ecosistema a partir de la aplicación de un instrumento diagnóstico; las concepciones alternativas de los estudiantes fueron clasificadas en los niveles 0, 1, 2 y 3 de complejidad. El 50% de las definiciones se ubicaron en el nivel 1 de complejidad (nivel aditivo) y sólo el 10% de las definiciones se identificaron en el nivel 3 de complejidad (nivel de organización compleja). Impact of ancestral sequence reconstruction on mechanistic and structural enzymology
Ancestral sequence reconstruction (ASR) provides insight into the changes within a protein sequence across evolution. More specifically, it can illustrate how specific amino acid changes give rise to different phenotypes within a protein family. Over the last few decades it has established itself as a powerful technique for revealing molecular common denominators that govern enzyme function. Here, we describe the strength of ASR in unveiling catalytic mechanisms and emerging phenotypes for a range of different proteins, also highlighting biotechnological applications the methodology can provide.</p
Ancestral reconstruction of mammalian FMO1 enables structural determination, revealing unique features that explain its catalytic properties
Mammals rely on the oxidative flavin-containing monooxygenases (FMOs) to detoxify numerous and potentially deleterious xenobiotics; this activity extends to many drugs, giving FMOs high pharmacological relevance. However, our knowledge regarding these membrane-bound enzymes has been greatly impeded by the lack of structural information. We anticipated that ancestral-sequence reconstruction could help us identify protein sequences that are more amenable to structural analysis. As such, we hereby reconstructed the mammalian ancestral protein sequences of both FMO1 and FMO4, denoted as ancestral flavin-containing monooxygenase (AncFMO)1 and AncFMO4, respectively. AncFMO1, sharing 89.5% sequence identity with human FMO1, was successfully expressed as a functional enzyme. It displayed typical FMO activities as demonstrated by oxygenating benzydamine, tamoxifen, and thioanisole, drug-related compounds known to be also accepted by human FMO1, and both NADH and NADPH cofactors could act as electron donors, a feature only described for the FMO1 paralogs. AncFMO1 crystallized as a dimer and was structurally resolved at 3.0 Å resolution. The structure harbors typical FMO aspects with the flavin adenine dinucleotide and NAD(P)H binding domains and a C-terminal transmembrane helix. Intriguingly, AncFMO1 also contains some unique features, including a significantly porous and exposed active site, and NADPH adopting a new conformation with the 2'-phosphate being pushed inside the NADP+ binding domain instead of being stretched out in the solvent. Overall, the ancestrally reconstructed mammalian AncFMO1 serves as the first structural model to corroborate and rationalize the catalytic properties of FMO1
Teaching Ourselves: A Model to Improve, Assess and Spread the Word
This paper presents a model for small, interdisciplinary groups of faculty to work together to improve their teaching while engaging in research that provides evidence of improved student learning. In doing so, we have developed a four-step process of faculty-driven scholarship of teaching and learning: Genesis, Organization, Implementation and Dissemination. We illustrate this model by describing our use of Fink’s (2003) concepts of course design to reshape our courses and assess the effectiveness of these changes through examination of student learning. We describe how others may follow this approach with a variety of applications
Aplicación de ingeniería de métodos para incrementar la productividad en el proceso de envasado en GENESIS E.I.R.L. – Chimbote, 2021
La presente investigación tuvo como objetivo general aplicar la ingeniería de
métodos para incrementar la productividad en el proceso de envasado en GENESIS
E.I.R.L. Este estudio fue del tipo aplicada, teniendo un diseño de investigación pre
experimental. La población estuvo conformada por la productividad de los procesos
productivos para la elaboración del filete de bonito en aceite vegetal, cuya muestra
fue la productividad del proceso de envasado. Algunos de los instrumentos
importantes fueron cursograma analítico del operario, diagrama de recorrido,
diagrama bimanual y formato de estudio de tiempos. Se obtuvo como resultado que
el porcentaje de actividades improductivas en el método propuesto fue de 30%, que
significó una disminución de 10% respecto al método actual. Por lo que se redujo
el tiempo estándar de 1864.02 segundos/caja a 1309.43 segundos/caja, generando
una disminución de 30%. Finalmente, las productividades de materia prima, mano
de obra y costo de mano de obra se incrementaron teniendo una variación de
2.24%, 32.64% y 32.31% respectivamente. Llegando a la conclusión que, gracias
a la aplicación de la ingeniería de métodos, se logró el incremento de la
productividad conllevando al beneficio de la empresa y a los trabajadores del
proceso de envasado
