    Violence, Care, Cure:(Self)perceptions Within the Medical Encounter

    Since the 1980s, scholars in the interdisciplinary field of the medical humanities have foregrounded how the distinction between ‘disease’, i.e. the medical classification of symptoms, and the individual experience of illness can be generative to unlock the tensions within the medical encounter. The doctor and the patient may tell two very different stories of illness, even contradictory ones. The mismatch that often arises between the medical gaze (Michel Foucault, 1963). and the individual’s gaze in the face of the same event can be productively addressed by paying attention to their visual and narrative representations. ​​In addition, the ways diseases have been portrayed with words and images have also affected the (self)perception of the body through history. If medicine is about stories (the patients’, doctors’, science’s, society’s), when and where do conflicts and misunderstandings arise that turn the cure into something perceived as violence? Literary and visual narratives contribute to sharing these stories and can offer a vantage point to address larger cultural scripts. How do these narratives represent perceptions of violence within medical settings and practices? The two-day symposium seeks to address the ambiguities of and tensions among perceptions at the cusp of internal (subjective) and external (social, cultural, political) ‘gazes’. What the individual experiences, at either end of the consultation room, is a complex interlacing of personal vicissitudes, global structures, and community practices: a prismatic network in which ‘care’ and ‘violence’ are reflected and refracted in a variety of oftentimes overlapping and divergent interpretative modes. Communities (whether concrete, virtual, or imagined) can be perceived as both providers of care and support, as well as instigators of violence. A case in point mirroring this ambiguity are for instance online pro-anorexia and pro-bulimia communities, where users create a space to share their experiences of eating disorders while at the same time promoting self-disruptive food behaviours. Another telling example is constituted by the outcomes of the ongoing pandemic; the alliance of extreme right-wing movements with anti-vaxers, and the protests against governments’ covid-related sanitary measures or the green pass (dubbed a ‘sanitary dictatorship’) have exposed the infiltration of radical ideologies and conspiracies into medical discourse, whereby care and violence lose their neat distinction. While a black and white opposition between internal/external, care/neglect, cure/violence may seem reductionist, engagement with these seemingly contrasting attitudes reveals the complex entanglements among possible scientific dogmatic drifts, social inequalities within healthcare systems, and idiosyncratic projections of individual and collective fears, which often lead to stigmatizing certain collectivities for the origin or transmission of contagious diseases (Zhao Xun and Sander L.     Impaired bone remodeling in children with osteogenesis imperfecta treated and untreated with bisphosphonates: the role of DKK1, RANKL, and TNF-α

    In this study, we investigated the bone cell activity in patients with osteogenesis imperfecta (OI) treated and untreated with neridronate. We demonstrated the key role of Dickkopf-1 (DKK1), receptor activator of nuclear factor-κB ligand (RANKL), and tumor necrosis factor alpha (TNF-α) in regulating bone cell of untreated and treated OI subjects. These cytokines could represent new pharmacological targets for OI. Introduction: Bisphosphonates are widely used in the treatment of children with osteogenesis imperfecta (OI) with the objective of reducing the risk of fractures. Although bisphosphonates increase bone mineral density in OI subjects, the effects on fracture incidence are conflicting. The aim of this study was to investigate the mechanisms underlying bone cell activity in subjects with mild untreated forms of OI and in a group of subjects with severe OI treated with cycles of intravenous neridronate. Methods: Sclerostin, DKK1, TNF-α, RANKL, osteoprotegerin (OPG), and bone turnover markers were quantified in serum of 18 OI patients (12 females, mean age 8.86 ± 3.90), 8 of which were receiving cyclic intravenous neridronate, and 21 sex- and age-matched controls. The effects on osteoblastogenesis and OPG expression of media conditioned by the serum of OI patients and anti-DKK1 neutralizing antibody were evaluated. Osteoclastogenesis was assessed in cultures from patients and controls. Results: DKK1 and RANKL levels were significantly increased both in untreated and in treated OI subjects with respect to controls. The serum from patients with high DKK1 levels inhibited both osteoblast differentiation and OPG expression in vitro. High RANKL and low OPG messenger RNA (mRNA) levels were found in lymphomonocytes from patients. High amounts of TNF-α were expressed by monocytes, and an elevated percentage of circulating CD11b-CD51/CD61+ osteoclast precursors was observed in patients. Conclusions: Our study demonstrated the key role of DKK1, RANKL, and TNF-α in regulating bone cell activity of subjects with OI untreated and treated with bisphosphonates. These cytokines could represent new pharmacological targets for OI patients

    The Masses of the Milky Way and Andromeda galaxies

    We present a family of robust tracer mass estimators to compute the enclosed mass of galaxy haloes from samples of discrete positional and kinematical data of tracers, such as halo stars, globular clusters and dwarf satellites. The data may be projected positions, distances, line of sight velocities or proper motions. Forms of the estimator tailored for the Milky Way galaxy and for M31 are given. Monte Carlo simulations are used to quantify the uncertainty as a function of sample size. For the Milky Way, the satellite sample consists of 26 galaxies with line-of-sight velocities. We find that the mass of the Milky Way within 300 kpc is ~ 0.9 x 10^12 solar masses assuming velocity isotropy. However, the mass estimate is sensitive to the anisotropy and could plausibly lie between 0.7 - 3.4 x 10^12 solar masses. Incorporating the proper motions of 6 Milky Way satellites into the dataset, we find ~ 1.4 x 10^12 solar masses. The range here if plausible anisotropies are used is still broader, from 1.2 - 2.7 x 10^12 solar masses. For M31, there are 23 satellite galaxies with measured line-of-sight velocities, but only M33 and IC 10 have proper motions. We use the line of sight velocities and distances of the satellite galaxies to estimate the mass of M31 within 300 kpc as ~ 1.4 x 10^12 solar masses assuming isotropy. There is only a modest dependence on anisotropy, with the mass varying between 1.3 -1.6 x 10^12 solar masses. Given the uncertainties, we conclude that the satellite data by themselves yield no reliable insights into which of the two galaxies is actually the more massive.Comment: 15 pages, submitted to MNRA

    A BLR origin for the iron Kα\alpha line in NGC7213

    The X-ray spectrum of NGC7213 is known to present no evidence for Compton reflection, a unique result among bright Seyfert 1s. The observed neutral iron Kα\alpha line, therefore, cannot be associated with a Compton-thick material, like the disc or the torus, but is due to Compton-thin gas, with the Broad Line Region (BLR) as the most likely candidate. To check this hypothesis, a long Chandra HETG observation, together with a quasi-simultaneous optical spectroscopic observation at the ESO NTT EMMI were performed. We found that the iron line is resolved with a FWHM=2400−600+11002 400^{+1 100}_{-600} km/s, in perfect agreement with the value measured for the broad component of the Hα\alpha, 2640−90+1102640^{+110}_{-90} km/s. Therefore, NGC7213 is the only Seyfert 1 galaxy whose iron Kα\alpha line is unambiguously produced in the BLR. We also confirmed the presence of two ionised iron lines and studied them in greater detail than before. The resonant line is the dominant component in the Fe XXV triplet, therefore suggesting an origin in collisionally ionised gas. If this is the case, the blueshift of around 1000 km/s of the two ionised iron lines could be the first measure of the velocity of a starburst wind from its X-ray emission.Comment: 5 pages, 4 figures, accepted for publication in MNRAS Letter

    Dietary patterns and associations with body mass index in low-income, ethnic minority youth in the United States according to baseline data from four randomized controlled trials

    Few studies have derived data-driven dietary patterns in youth in the United States (US). This study examined data-driven dietary patterns and their associations with BMI measures in predominantly low-income, racial/ethnic minority US youth. Data were from baseline assessments of the four Childhood Obesity Prevention and Treatment Research (COPTR) Consortium trials: NET-Works (N=534; 2–4-year-olds), GROW (N=610; 3–5-year-olds), GOALS (N=241; 7–11-year-olds), and IMPACT (N=360; 10–13-year-olds). Weight and height were measured. Children/adult proxies completed 3 24-hour dietary recalls. Dietary patterns were derived for each site from 24 food/beverage groups using k-means cluster analysis. Multivariable linear regression models examined associations of dietary patterns with BMI and percentage of the 95th BMI percentile. Healthy (produce and whole grains) and Unhealthy (fried food, savory snacks, and desserts) patterns were found in NET-Works and GROW. GROW additionally had a dairy and sugar-sweetened beverage based pattern. GOALS had a similar Healthy pattern and a pattern resembling a traditional Mexican diet. Associations between dietary patterns and BMI were only observed in IMPACT. In IMPACT, youth in the Sandwich (cold cuts, refined grains, cheese, and miscellaneous [e.g., condiments]) compared to Mixed (whole grains and desserts) cluster had significantly higher BMI [β=0.99 (95% CI: 0.01, 1.97)] and percentage of the 95th BMI percentile [β=4.17 (95% CI: 0.11, 8.24)]. Healthy and Unhealthy patterns were the most common dietary patterns in COPTR youth, but diets may differ according to age, race/ethnicity, or geographic location. Public health messages focused on healthy dietary substitutions may help youth mimic a dietary pattern associated with lower BMI

Each work place requires different set of soft skills. Thus, in addition to scientific/technical skills that are easier to access within a short time frame, several key soft skills are essential for the success of a researcher in today’s international work environment. In this paper, the trainees and trainers of the EU-CardioRNA COST Action CA17129 training school on soft skills present basic and advanced soft skills for early career researchers. Here, we particularly emphasize on the importance of transferable and presentation skills, ethics, literature reading and reviewing, research protocol and grant writing, networking, and career opportunities for researchers. All these skills are vital but are often overlooked by some scholars. We also provide tips to ace in aforementioned skills that are crucial in a day-to-day life of early and late career researchers in academia and industry.</ns4:p

    Multicentre Italian study of SARS-CoV-2 infection in children and adolescents, preliminary data as at 10 April 2020

    Data on features of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in children and adolescents are scarce. We report preliminary results of an Italian multicentre study comprising 168 laboratory-confirmed paediatric cases (median: 2.3 years, range: 1 day-17.7 years, 55.9% males), of which 67.9% were hospitalised and 19.6% had comorbidities. Fever was the most common symptom, gastrointestinal manifestations were frequent; two children required intensive care, five had seizures, 49 received experimental treatments and all recovered

    Visualization and 3D Reconstruction of Flame Cells of Taenia solium (Cestoda)

    BACKGROUND: Flame cells are the terminal cells of protonephridial systems, which are part of the excretory systems of invertebrates. Although the knowledge of their biological role is incomplete, there is a consensus that these cells perform excretion/secretion activities. It has been suggested that the flame cells participate in the maintenance of the osmotic environment that the cestodes require to live inside their hosts. In live Platyhelminthes, by light microscopy, the cells appear beating their flames rapidly and, at the ultrastructural, the cells have a large body enclosing a tuft of cilia. Few studies have been performed to define the localization of the cytoskeletal proteins of these cells, and it is unclear how these proteins are involved in cell function. METHODOLOGY/PRINCIPAL FINDINGS: Parasites of two different developmental stages of T. solium were used: cysticerci recovered from naturally infected pigs and intestinal adults obtained from immunosuppressed and experimentally infected golden hamsters. Hamsters were fed viable cysticerci to recover adult parasites after one month of infection. In the present studies focusing on flame cells of cysticerci tissues was performed. Using several methods such as video, confocal and electron microscopy, in addition to computational analysis for reconstruction and modeling, we have provided a 3D visual rendition of the cytoskeletal architecture of Taenia solium flame cells. CONCLUSIONS/SIGNIFICANCE: We consider that visual representations of cells open a new way for understanding the role of these cells in the excretory systems of Platyhelminths. After reconstruction, the observation of high resolution 3D images allowed for virtual observation of the interior composition of cells. A combination of microscopic images, computational reconstructions and 3D modeling of cells appears to be useful for inferring the cellular dynamics of the flame cell cytoskeleton
