    Simple Heuristics and the Modelling of Crowd Behaviours

    A crowd of pedestrians is a complex system that exhibits a rich variety of self-organized collective behaviors, such as lane formation, stop-and-go waves, or crowd turbulence. Understanding the mechanisms of crowd dynamics requires establishing a link between the local behavior of pedestrians during interactions, and the global dynamics of the crowd at high density. For this, the elaboration of a model is necessary. In this contribution, we will make a distinction between two kinds of modelling methods: outcome models that are often based on analogies with Newtonian mechanics, and process models based on concepts of cognitive science. While outcome models describe directly the movements of a pedestrian by means of repulsive forces or probabilities to move from one place to another, process models generate the movement from the bottom-up by describing the underlying cognitive process used by the pedestrian during navigation. Here, we will describe and compare two representatives of outcome and process models, namely the social force model on the one hand, and the heuristic model on the other hand. In particular, we will describe the strength and the limitations of each approach, and discuss possible future improvements for process models

    Interdisciplinary design - Forming and evaluating teams

    The College of Architecture and Environmental Design at California Polytechnic State University has offered an upper division, interdisciplinary experience for every student in the form of a project based, team oriented five unit studio laboratory. The course is now in its fifth year and requires small teams of architecture, engineering and construction students to complete the schematic level design of an actual building for a real client. The quality of the projects and student deliverables has been outstanding and students are clearly meeting the objective to prepare an integrated building design. The other course objective is to function as a member of an interdisciplinary team, which is more difficult to quantify. This paper focuses on the selection and assessment of teams in this course. Various personality and skills assessments are completed and used in the formations of teams. Assessment data on team performance are presented and future actions for this project are discussed

    UNLABELLED: Persistent human cytomegalovirus (HCMV) infection has been linked to several diseases, including atherosclerosis, transplant vascular sclerosis (TVS), restenosis, and glioblastoma. We have previously shown that factors secreted from HCMV-infected cells induce angiogenesis and that this process is due, at least in part, to increased secretion of interleukin-6 (IL-6). In order to identify the HCMV gene(s) responsible for angiogenesis promotion, we constructed a large panel of replication-competent HCMV recombinants. One HCMV recombinant deleted for UL1 to UL10 was unable to induce secretion of factors necessary for angiogenesis. Fine mapping using additional HCMV recombinants identified UL7 as a viral gene required for production of angiogenic factors from HCMV-infected cells. Transient expression of pUL7 induced phosphorylation of STAT3 and ERK1/2 MAP kinases and production of proangiogenic factors, including IL-6. Addition of recombinant pUL7 to cells was sufficient for angiogenesis and was again associated with increased IL-6 expression. Analysis of the UL7 structure revealed a conserved domain similar to the immunoglobulin superfamily domain and related to the N-terminal V-like domain of carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1). Our report therefore identifies UL7 as a novel HCMV-encoded molecule that is both structurally and functionally related to cellular CEACAM1, a proangiogenic factor highly expressed during vasculogenesis. IMPORTANCE: A hallmark of cytomegalovirus (CMV) infection is its ability to modulate the host cellular machinery, resulting in the secretion of factors associated with long-term diseases such as vascular disorders and cancer. We previously demonstrated that HCMV infection alters the types and quantities of bioactive proteins released from cells (designated the HCMV secretome) that are involved in the promotion of angiogenesis and wound healing. A key proangiogenic and antiapoptotic factor identified from a proteomic-based approach was IL-6. In the present report, we show for the first time that HCMV UL7 encodes a soluble molecule that is a structural and functional homologue of the CEACAM1 proangiogenic cellular factor. This report thereby identifies a critical component of the HCMV secretome that may be responsible, at least in part, for the vascular dysregulation associated with persistent HCMV infection

    Experimental and computational investigation of co production and dispersion in an automotive repair shop

    Carbon monoxide (CO), a highly toxic gas, is produced during the incomplete combustion of carbon-based fuels. In indoor environments, high CO concentrations constitute a serious occupational health hazard; this is especially true in the case of automotive repair shop (ARS) employees who are exposed on a daily basis to vehicle exhaust streams. The present study focuses on the experimental investigation and numerical simulation of CO production and dispersion inside an ARS facility. Detailed measurements of CO concentration, vehicle traffic and ventilation system velocities are performed; the obtained data are appropriately formulated to provide quantitative information for modelling purposes. A detailed Computational Fluid Dynamics simulation of the developing transient flow-field is performed. The numerical results are validated using the experimental data; an overall good qualitative and quantitative agreement is achieved. Aiming to improve the energy efficiency of the mechanical ventilation system, three alternative scenarios are investigated; it is shown that the utilization of a dynamic ventilation system may result in significant energy consumption benefits, while, at the same time, CO concentrations remain below the values suggested by current occupational health legislation. The obtained results may be utilized to assist the design of mechanical ventilation systems for ARS facilities

    Evaluation of Complex Whole-School Interventions: Methodological and Practical Considerations

    Evaluating the impact of complex whole-school interventions (CWSIs) is challenging. However, what evidence there is suggests that school leadership and other elements of whole-school contexts are important for pupils’ attainment (Leithwood et al., 2006), suggesting that interventions aimed at changing these have significant potential to improve pupil outcomes. Furthermore, strong leadership is likely important for the effective implementation of many interventions funded by the EEF since even class-level or targeted programmes are more likely to work best within supportive and effective settings. We therefore welcome the EEF’s commitment to exploring the issues inherent in evaluating CWSIs. Developing design and practice for evaluations of this type of intervention, focusing on the issues of complexity and managing change across a whole school, increases the scope of projects of which the EEF may confidently fund evaluations. In this document, we provide key messages for EEF evaluators on how to get the most out of evaluations of CWSIs, including considerations for both design and implementation. As far as possible, our suggestions aim to be practical steps that evaluators can implement immediately. A number of issues, and points 13 and 14 below in particular, require either further investigation or decisions from the EEF

    Relationship Between Cerebral Blood Flow and Blood Pressure in Long-Term Heart Transplant Recipients

    Heart transplant recipients are at an increased risk for cerebral hemorrhage and ischemic stroke, yet the exact mechanism for this derangement remains unclear. We hypothesized that alterations in cerebrovascular regulation is principally involved. To test this hypothesis, we studied cerebral pressure-flow dynamics in 8 clinically stable male heart transplant recipients (62 Β± 8 years of age and 9 Β± 7 years post-transplant, mean + SD), 9 male age-matched controls (63 Β± 8 years) and 10 male donor controls (27 Β± 5 years). To increase blood pressure variability and improve assessment of the pressure-flow dynamics, subjects performed squat-stand maneuvers at 0.05 and 0.10 Hz. Beat-to-beat blood pressure, middle cerebral artery velocity, and end-tidal carbon dioxide were continuously measured during five minutes of seated rest, and throughout the squat-stand maneuvers. Cardiac baroreceptor sensitivity gain and cerebral pressure-flow responses were assessed with linear transfer function analysis. Heart transplant recipients had reductions in R-R interval power and baroreceptor sensitivity low frequency gain (P<0.01) compared to both control groups; however, these changes were unrelated to transfer function metrics. Thus, in contrast to our hypothesis, the increased risk of cerebrovascular complication after heart transplantation does not appear to be related to alterations in cerebral pressure-flow dynamics. Future research is therefore warranted

    Steady-state modulation of voltage-gated K+ channels in rat arterial smooth muscle by cyclic AMP-dependent protein kinase and protein phosphatase 2B

    Voltage-gated potassium channels (Kv) are important regulators of membrane potential in vascular smooth muscle cells, which is integral to controlling intracellular Ca2+ concentration and regulating vascular tone. Previous work indicates that Kv channels can be modulated by receptor-driven alterations of cyclic AMP-dependent protein kinase (PKA) activity. Here, we demonstrate that Kv channel activity is maintained by tonic activity of PKA. Whole-cell recording was used to assess the effect of manipulating PKA signalling on Kv and ATP-dependent K+ channels of rat mesenteric artery smooth muscle cells. Application of PKA inhibitors, KT5720 or H89, caused a significant inhibition of Kv currents. Tonic PKA-mediated activation of Kv appears maximal as application of isoprenaline (a Ξ²-adrenoceptor agonist) or dibutyryl-cAMP failed to enhance Kv currents. We also show that this modulation of Kv by PKA can be reversed by protein phosphatase 2B/calcineurin (PP2B). PKA-dependent inhibition of Kv by KT5720 can be abrogated by pre-treatment with the PP2B inhibitor cyclosporin A, or inclusion of a PP2B auto-inhibitory peptide in the pipette solution. Finally, we demonstrate that tonic PKA-mediated modulation of Kv requires intact caveolae. Pre-treatment of the cells with methyl-Ξ²-cyclodextrin to deplete cellular cholesterol, or adding caveolin-scaffolding domain peptide to the pipette solution to disrupt caveolae-dependent signalling each attenuated PKA-mediated modulation of the Kv current. These findings highlight a novel, caveolae-dependent, tonic modulatory role of PKA on Kv channels providing new insight into mechanisms and the potential for pharmacological manipulation of vascular tone

    A success story: HIV prevention for injection drug users in Rhode Island

    BACKGROUND: New HIV diagnoses related to injection drug use (IDU) have declined in the United States. Access to clean syringes and decreasing HIV transmission among injection drug users have been HIV prevention priorities of the Rhode Island (RI) HIV community. To examine trends in IDU-related new HIV diagnoses in RI, we performed a retrospective analysis of new HIV diagnoses according to HIV risk factor from 1990–2003. RESULTS: There has been an 80% absolute reduction in IDU-related new HIV diagnoses in RI coincident with IDU-specific prevention efforts. CONCLUSION: There has been a greater decline in IDU-related new HIV diagnoses in Rhode Island compared to national data reported by the Centers for Disease Control and Prevention. We hypothesize that this dramatic decline in Rhode Island is related to extensive HIV prevention efforts targeting IDUs. Further research is needed to examine the impact of specific HIV prevention interventions for injection drug users
