    Morphological re-description and molecular identification of Tabanidae (Diptera) in East Africa

    Biting flies of the family Tabanidae are important vectors of human and animal diseases across continents. However, records of Africa tabanids are fragmentary and mostly cursory. To improve identification, documentation and description of Tabanidae in East Africa, a baseline survey for the identification and description of Tabanidae in three eastern African countries was conducted. Tabanids from various locations in Uganda (Wakiso District), Tanzania (Tarangire National Park) and Kenya (Shimba Hills National Reserve, Muhaka, Nguruman) were collected. In Uganda, octenol baited F-traps were used to target tabanids, while NG2G traps baited with cow urine and acetone were employed in Kenya and Tanzania. The tabanids were identified using morphological and molecular methods. Morphologically, five genera (Ancala, Tabanus, Atylotus, Chrysops and Haematopota) and fourteen species of the Tabanidae were identified. Among the 14 species identified, six belonged to the genus Tabanus of which two (T. donaldsoni and T. guineensis) had not been described before in East Africa. The greatest diversity of tabanid species were collected from the Shimba Hills National Reserve, while collections from Uganda (around the shores of Lake Victoria) had the fewest number of species. However, the Ancala genus was found in Uganda, but not in Kenya or Tanzania. Maximum likelihood phylogenies of mitochondrial cytochrome c oxidase 1 (COI) genes sequenced in this study show definite concordance with morphological species identifications, except for Atylotus. This survey will be critical to building a complete checklist of Tabanidae prevalent in the region, expanding knowledge of these important vectors of human and animal diseases

    Effect of HIV infection on the acute antibody response to malaria antigens in children: an observational study

    <p>Abstract</p> <p>Background</p> <p>In sub-Saharan Africa, the distributions of malaria and HIV widely overlap. Among pregnant and non-pregnant adults, HIV affects susceptibility to malaria, its clinical course and impairs antibody responses to malaria antigens. However, the relationship between the two diseases in childhood, when most deaths from malaria occur, is less clear. It was previously reported that HIV is associated with admission to hospital in rural Kenya with severe malaria among children, except in infancy. HIV-infected children with severe malaria were older, had higher parasite density and increased mortality, raising a hypothesis that HIV interferes with naturally acquired immunity to malaria, hence with little effect at younger ages (a shorter history of exposure). To test this hypothesis, levels of anti-merozoite and schizont extract antibodies were compared between HIV-infected and uninfected children who participated in the original study.</p> <p>Methods</p> <p>IgG responses to malaria antigens that are potential targets for immunity to malaria (AMA1, MSP2, MSP3 and schizont extract) were compared between 115 HIV-infected and 115 age-matched, HIV-uninfected children who presented with severe malaria. The children were classified as high and low responders for each antigen and assigned antibody-response breadth scores according to the number of antigens to which they were responsive. A predictive logistic regression model was used to test if HIV was an effect modifier on the age-related acquisition of antibody responses, with age as a continuous variable.</p> <p>Results</p> <p>Point estimates of the responses to all antigens were lower amongst HIV-infected children, but this was only statistically significant for AMA1 (P = 0.028). HIV-infected children were less likely to be high responders to AMA1 [OR 0.44 (95%CI, 0.2-0.90) P = 0.024]. HIV was associated with a reduced breadth of responses to individual merozoite antigens (P = 0.02). HIV strongly modified the acquisition of antibodies against schizont extract with increasing age (P < 0.0001), but did not modify the rate of age-related acquisition of responses to individual merozoite antigens.</p> <p>Conclusions</p> <p>In children with severe malaria, HIV infection is associated with a lower magnitude and narrower breadth of IgG responses to merozoite antigens and stunting of age-related acquisition of the IgG antibody response to schizont extract.</p

    New antitrypanosomal tetranotriterpenoids from Azadirachta Indica

    Organic extracts of the leaves of Azadirachta indica A. Juss. yielded ten antitrypanosomal terpenoids. Three of these (1 – 3), are novel and are derivatives of nimbolide and nimbin. They were extracted from chloroformfraction of methanol extract. These compounds were found to exhibit strong antitrypanosomal activities against Trypanosoma brucei rhodesiense with MIC values ranging of 6.9, 15.6 and 7.8 μg/ml respectively and were more active than Cymerlarsan ( a standard drug), which had an MIC value of 187.5 μg/ml when tested against T. b. rhodesiense The structures were elucidated by spectroscopic methods including; NMR, MS, UV and IR.Key words: Meliaceae, limonoids, Trypanosoma brucei rhodesiense, Azadirachta indica, antitrypanosomal activity

    Teen Dating Violence among Italian High School Students: A Quantitative Study on Gender Differences

    Healthy relationships involve trust, integrity, respect, and cooperation. Unfortunately, teen dating violence is a serious problem and there has been a consensus that it has severe consequences on the victims’ physical and psychological health. It can negatively influence the development of healthy sexuality, intimacy, and identity of adolescents as they transition into adulthood. This study examined the gender differences in teen dating violence among 336 Italian adolescent students from four secondary schools in Reggio Emilia, aged 14 to 20 years. The study used questionnaires for data collection and the results revealed that both genders have a significantly greater acceptance of control behavior when compared to their acceptance of aggressive behavior. In addition, boys accepted interpersonal violence more than girls. A Significant high percentage of girls reported victimization, with physical, emotional, and threatening violence perpetrated more against them. Most of the teens were aware of TDV among peers, and an experience of TDV was among the causal attributions mentioned. Others reported the fear of losing a partner and reaction to a provoking behavior. The recommendations drawn included the importance of addressing masculinity models that see aggressiveness as part of their gender identity and the relevance of raising awareness of control behaviors as antecedents of teen dating violence

    Hydrogen Peroxide Inhibition of Bicupin Oxalate Oxidase

    Oxalate oxidase is a manganese containing enzyme that catalyzes the oxidation of oxalate to carbon dioxide in a reaction that is coupled with the reduction of oxygen to hydrogen peroxide. Oxalate oxidase from Ceriporiopsis subvermispora (CsOxOx) is the first fungal and bicupin enzyme identified that catalyzes this reaction. Potential applications of oxalate oxidase for use in pancreatic cancer treatment, to prevent scaling in paper pulping, and in biofuel cells have highlighted the need to understand the extent of the hydrogen peroxide inhibition of the CsOxOx catalyzed oxidation of oxalate. We apply a membrane inlet mass spectrometry (MIMS) assay to directly measure initial rates of carbon dioxide formation and oxygen consumption in the presence and absence of hydrogen peroxide. This work demonstrates that hydrogen peroxide is both a reversible noncompetitive inhibitor of the CsOxOx catalyzed oxidation of oxalate and an irreversible inactivator. The build-up of the turnover-generated hydrogen peroxide product leads to the inactivation of the enzyme. The introduction of catalase to reaction mixtures protects the enzyme from inactivation allowing reactions to proceed to completion. Circular dichroism spectra indicate that no changes in global protein structure take place in the presence of hydrogen peroxide. Additionally, we show that the CsOxOx catalyzed reaction with the three carbon substrate mesoxalate consumes oxygen which is in contrast to previous proposals that it catalyzed a non-oxidative decarboxylation with this substrate

    Global summary of baseline household survey results

    This report summarises the results of a baseline household-level survey, designed by the CCAFS team and implemented in late 2010/early 2011 in 3 regions: East Africa, West Africa and South Asia. This survey was designed with the intent of developing simple, comparable cross-site household level indicators, for which changes can be evaluated over time, of food security, households assets, agricultural production diversity, agricultural sales diversity, changes being made in farming practices for adaptation, innovation, and/or to help reduce emissions or store greenhouse gases (mitigation), and gender indicators (e.g. men’s versus women’s reception of weather-related information). A standardised survey, guidelines and training materials were implemented across 12 countries in West Africa (Senegal, Mali, Burkina Faso, Niger, and Ghana), East Africa (Kenya, Uganda, Ethiopia, and Tanzania), and South Asia (India, Nepal and Bangladesh). Thus we have a total of 15 core sites covering 108 villages and 2095 households. This summary report gives an overview of the farming practices, and asset, livelihoods and food security status of rural households in these sites. It describes what changes farmers have been making in recent years with respect to crop management, livestock practices, use of inputs and other agricultural practices. We explore what kinds of weather/climate and associated information these households are receiving, how and by whom. This information provides important baseline information, as these households will be revisited in 5-10 years’ time in order to evaluate the changes in these indicators. This will give us important information as to if, how, and which households are adapting to a changing climate while improving their food security status

Decreases in environmental Pb contamination corresponding with the near-global phase-out of leaded automobile gasoline beginning in the 1970s have since been observed. Measurements of Pb concentration in snow and ice core samples from Antarctica show that recent fluxes of industrial Pb to Antarctica have similarly declined. Here, we present measurements of Pb concentrations and isotopic compositions in seawater and surface sediments from the Amundsen Sea continental shelf including the Amundsen Sea Polynya. Both sets of measurements show that most (∼60–95%) of the Pb at our sites, at the time of sampling, is natural in source: that is, derived from the weathering of Antarctic continental rocks. These fluxes of natural Pb then become entrained into polynya waters either from sediment resuspension or from the transport of sediment-laden glacial melt waters to the polynya.publishedVersio

    The impact of malaria parasites on dendritic cell–T cell interaction

    Malaria is caused by apicomplexan parasites of the genus Plasmodium. While infection continues to pose a risk for the majority of the global population, the burden of disease mainly resides in Sub-Saharan Africa. Although immunity develops against disease, this requires years of persistent exposure and is not associated with protection against infection. Repeat infections occur due to the parasite's ability to disrupt or evade the host immune responses. However, despite many years of study, the mechanisms of this disruption remain unclear. Previous studies have demonstrated a parasite-induced failure in dendritic cell (DCs) function affecting the generation of helper T cell responses. These T cells fail to help B cell responses, reducing the production of antibodies that are necessary to control malaria infection. This review focuses on our current understanding of the effect of Plasmodium parasite on DC function, DC-T cell interaction, and T cell activation. A better understanding of how parasites disrupt DC-T cell interactions will lead to new targets and approaches to reinstate adaptive immune responses and enhance parasite immunity
