    Human ribosomal protein S13 regulates expression of its own gene at the splicing step by a feedback mechanism

    The expression of ribosomal protein (rp) genes is regulated at multiple levels. In yeast, two genes are autoregulated by feedback effects of the protein on pre-mRNA splicing. Here, we have investigated whether similar mechanisms occur in eukaryotes with more complicated and highly regulated splicing patterns. Comparisons of the sequences of ribosomal protein S13 gene (RPS13) among mammals and birds revealed that intron 1 is more conserved than the other introns. Transfection of HEK 293 cells with a minigene-expressing ribosomal protein S13 showed that the presence of intron 1 reduced expression by a factor of four. Ribosomal protein S13 was found to inhibit excision of intron 1 from rpS13 pre-mRNA fragment in vitro. This protein was shown to be able to specifically bind the fragment and to confer protection against ribonuclease cleavage at sequences near the 5′ and 3′ splice sites. The results suggest that overproduction of rpS13 in mammalian cells interferes with splicing of its own pre-mRNA by a feedback mechanism

    Outcomes of the multicenter monitoring of the causative agent of invasive listeriosis in the metropolis

    Introduction. Invasive listeriosis is a rare disease posing a threat to high-risk groups and often leading to a fatal outcome. Its causative agent is Listeria monocytogenes, a ubiquitous saprophyte that has turned into an important foodborne pathogen with the growing industry of semi-cooked and ready-to-eat products. The aim of the study is the characterization of L. monocytogenes isolates in the Moscow region and identification of possible causes of susceptibility to infection Materials and methods. The multicenter monitoring of L. monocytogenes was conducted in the Moscow metropolitan area, using bacteriological and genomic methods for description of the pathogen, medical history collection and detailed analysis of patient case summaries. Results. In the cohorts of patients with perinatal listeriosis (PL) and meningitis-septicemia (MS), invasive listeriosis had a year-round occurrence with slight upswings in MarchApril and JulyNovember. During the COVID-19 pandemic, in the MS group, the minimum age of patients decreased to 31 years and the proportion of deaths increased 1.57-fold compared to 20182019. During the pandemic, an increase in the diversity of L. monocytogenes genotypes was observed, along with changes in the spectrum of pathogen genotypes throughout the pandemic stages. During the monitoring, a total of 73 L. monocytogenes clinical isolates belonging to 24 genotypes were described. Seven genotypes belonged to the first phylogenetic lineage (PLI); 14 genotypes belonged to PLII. The PL cohort had the highest proportion of PLI genotypes (52%). In the MS cohort, the group of men had the widest diversity of genotypes, 6 of which were identical to genotypes of food isolates. In the analysed set of isolates, 12 new profiles of internalin genes were identified and described. The whole genome sequencing detected the presence of plasmids in 9 of 58 genomes of clinical isolates. The comparison of core genomes revealed an epidemic relationship between isolates of the same genotype for ST4, ST21, and ST425. Conclusion. The performed study presents a detailed description of the diversity and virulence of L. monocytogenes circulating in the Moscow metropolitan area, thus providing information for timely diagnosis and treatment of invasive listeriosis

    Atrasentan and renal events in patients with type 2 diabetes and chronic kidney disease (SONAR): a double-blind, randomised, placebo-controlled trial

    Background: Short-term treatment for people with type 2 diabetes using a low dose of the selective endothelin A receptor antagonist atrasentan reduces albuminuria without causing significant sodium retention. We report the long-term effects of treatment with atrasentan on major renal outcomes. Methods: We did this double-blind, randomised, placebo-controlled trial at 689 sites in 41 countries. We enrolled adults aged 18–85 years with type 2 diabetes, estimated glomerular filtration rate (eGFR)25–75 mL/min per 1·73 m 2 of body surface area, and a urine albumin-to-creatinine ratio (UACR)of 300–5000 mg/g who had received maximum labelled or tolerated renin–angiotensin system inhibition for at least 4 weeks. Participants were given atrasentan 0·75 mg orally daily during an enrichment period before random group assignment. Those with a UACR decrease of at least 30% with no substantial fluid retention during the enrichment period (responders)were included in the double-blind treatment period. Responders were randomly assigned to receive either atrasentan 0·75 mg orally daily or placebo. All patients and investigators were masked to treatment assignment. The primary endpoint was a composite of doubling of serum creatinine (sustained for ≥30 days)or end-stage kidney disease (eGFR <15 mL/min per 1·73 m 2 sustained for ≥90 days, chronic dialysis for ≥90 days, kidney transplantation, or death from kidney failure)in the intention-to-treat population of all responders. Safety was assessed in all patients who received at least one dose of their assigned study treatment. The study is registered with ClinicalTrials.gov, number NCT01858532. Findings: Between May 17, 2013, and July 13, 2017, 11 087 patients were screened; 5117 entered the enrichment period, and 4711 completed the enrichment period. Of these, 2648 patients were responders and were randomly assigned to the atrasentan group (n=1325)or placebo group (n=1323). Median follow-up was 2·2 years (IQR 1·4–2·9). 79 (6·0%)of 1325 patients in the atrasentan group and 105 (7·9%)of 1323 in the placebo group had a primary composite renal endpoint event (hazard ratio [HR]0·65 [95% CI 0·49–0·88]; p=0·0047). Fluid retention and anaemia adverse events, which have been previously attributed to endothelin receptor antagonists, were more frequent in the atrasentan group than in the placebo group. Hospital admission for heart failure occurred in 47 (3·5%)of 1325 patients in the atrasentan group and 34 (2·6%)of 1323 patients in the placebo group (HR 1·33 [95% CI 0·85–2·07]; p=0·208). 58 (4·4%)patients in the atrasentan group and 52 (3·9%)in the placebo group died (HR 1·09 [95% CI 0·75–1·59]; p=0·65). Interpretation: Atrasentan reduced the risk of renal events in patients with diabetes and chronic kidney disease who were selected to optimise efficacy and safety. These data support a potential role for selective endothelin receptor antagonists in protecting renal function in patients with type 2 diabetes at high risk of developing end-stage kidney disease.     Problems and forecast of the development of the Russian fuel and energy complex: the factorial approach

    Existing forecasts of Russian energy sector development have a number of general provisions and differ in approaches to their construction. The most realistic forecast is made within the framework of the Energy Strategy of Russia for the period up to 2035. However, the dynamism and high uncertainty of economic processes, international relations, domestic policy and the rapid growth of scientific and technical progress need a systematic correction of any forecast. The purpose of the article is to develop theoretical provisions and practical recommendations for making a forecast for the development of the Russian fuel and energy complex based on the application of the factor approach. The forecasting results are used in the development of management decisions, therefore, the authors of the article take into account the influence of various factors while making the forecast, but they (factors) are all quantitatively commensurable. To make a reliable forecast, the internal environment of the research object is studied in detail, organizational and economic problems in the development of the Russian fuel and energy complex are revealed, and methods for their elimination are suggested. The external environment is also analyzed and trends in the development of world energy markets are revealed. To forecast the state of the fuel and energy complex, an integral indicator is chosen reflecting the level of its technical and economic development

    Study of the effect of the acid-base surface properties of ZnO, Fe2O3 and ZnFe2O4 oxides on their gas sensitivity to ethanol vapor

    Binary (ZnO, Fe2O3) and ternary (ZnFe2O4) gas-sensitive oxide materials are synthesized, and the correlation between their sensitivity to ethanol vapor and the functional chemical composition of the surface is studied by X-ray photoelectron spectroscopy and by the technique of the adsorption of acid-base indicators. It is found that the sensitivity to ethanol increases with increasing content of Bronsted acid sites with the acidity index pK (a) a parts per thousand 2.5 and with increasing percentage of surface oxygen involved in OH/CO3/C-O groups. This interrelation is attributed to the specific features of interaction between ethanol molecules and hydroxyl groups on the surface of the oxides

    The relevance of the research is due to the global trends of development of the information society that are associated with the rapid advancement of civilization (IT penetration, increased computer availability, variability) and innovation processes in the sphere of education (competency-based approach, humanization and humanitarization). These development trends of the information society influence both the development of personality as a subject of activity and of its relevant features, such as information competency and creative initiative. With regard to this, the paper is aimed at revealing the essential meanings of the phenomena "information competency" and "creative initiative of personality" via the dialectics of their development, self-development and mutual influence. The leading method for studying this problem is modeling that allows considering in an integrated manner the existing and the other as factors detailing the mutual influence of the information competency and creative initiative of personality within the structure of its individuality, as well as detailing the dialectics of development of the information competency and creative initiative of personality through the manifestation of subjective reality in the structure of individuality. In the paper, the essence of the human information competency and its role in the development of the information society is identified and approaches to understanding the personality's creative initiative and its manifestations in the activity. The paper also details the mechanisms of transformation and integration of the processes of development of information competency and creative activity of personality as of a subject of activity. The authors refer to them the following: competence, reflexivity, "inner dialectics", subjectness, "intersubjectivity", ability of dialogue, and manifestation of individuality. The materials of the paper are of practical value for master degree students, postgraduate students, teachers of higher educational institutions and other educational establishments pursuing the research in this sphere who set themselves the objectives of building the forecast models of dynamics of the development of the personality's information competency and creative initiative
