    Neuroprotective and anti-oxidant effects of caffeic acid isolated from Erigeron annuus leaf

    <p>Abstract</p> <p>Background</p> <p>Since oxidative stress has been implicated in a neurodegenerative disease such as Alzheimer's disease (AD), natural antioxidants are promising candidates of chemopreventive agents. This study examines antioxidant and neuronal cell protective effects of various fractions of the methanolic extract of <it>Erigeron annuus </it>leaf and identifies active compounds of the extract.</p> <p>Methods</p> <p>Antioxidant activities of the fractions from <it>Erigeron annuus </it>leaf were examined with [2,2-azino-bis(3-ethylbenz thiazoline-6-sulfonic acid diammonium salt)] (ABTS) and ferric reducing antioxidant power (FRAP) assays. Neuroprotective effect of caffeic acid under oxidative stress induced by H<sub>2</sub>O<sub>2 </sub>was investigated with [3-(4,5-dimethythiazol-2-yl)-2,5-diphenyl tetrazolium bromide] (MTT) and lactate dehydrogenase (LDH) assays.</p> <p>Results</p> <p>This study demonstrated that butanol fraction had the highest antioxidant activity among all solvent fractions from methanolic extract <it>E. annuus </it>leaf. Butanol fraction had the highest total phenolic contents (396.49 mg of GAE/g). Caffeic acid, an isolated active compound from butanol fraction, showed dose-dependent <it>in vitro </it>antioxidant activity. Moreover, neuronal cell protection against oxidative stress induced cytotoxicity was also demonstrated.</p> <p>Conclusion</p> <p><it>Erigeron annuus </it>leaf extracts containing caffeic acid as an active compound have antioxidative and neuroprotective effects on neuronal cells.</p

    Gold in Peru: Present status and future challenge

    Total reported gold resources in Peru is about 192 Moz. Gold production in Peru was 4.9 Moz in 2013, which was ranked first in Latin America and sixth in the world. Historic cumulative gold production in Peru is 118 Moz, and production from main gold belts including Miocene epithermal belt, Carboniferous-Permian orogenic gold belt and Upper Cretaceous intrusion-related gold belt corresponds to 84%. Most of production areas are located in Northern part of Peru, which corresponds to 63.5% of the total domestic production. Annual production onces in Yanacocha mine and Alto Chicama were 1 Moz and 0.606 Moz in 2013, which were ranked first and second in Peru, respectively. Gold production in Peru is expected to be 6.5 Moz in 2017. To accomplish the expected production, ongoing 14 projects should be developed to the production stage in three years

    Silver in Peru: Present status and future challenge

    Silver production in Peru was 118 Moz in 2013, which was 3rd ranked in the world. Silver production mines which were ranked from 1st to 5th in Peru in 2013 were Antamina(16.7 Moz), Uchuchacua(12 Moz), Pallancata(7.6 Moz), Animón(7 Moz), and Arcata(5.4 Moz). Total reported silver resources in Peru is about 7,012 Moz, and resources from the belts of Miocene epithermal deposits and the belts of Miocene skarn, replacement and vein deposits are 4,812 Moz, which corresponds to 69% of total resources. There are 14 ongoing projects which will be developed to the production stage from 2014 to 2019. Through these projects, silver production in Peru is expected to be 148 Moz in 2017

    The landscape of PBMC methylome in canine mammary tumors reveals the epigenetic regulation of immune marker genes and its potential application in predicting tumor malignancy

    Background Genome-wide dysregulation of CpG methylation accompanies tumor progression and characteristic states of cancer cells, prompting a rationale for biomarker development. Understanding how the archetypic epige‑ netic modifcation determines systemic contributions of immune cell types is the key to further clinical benefts. Results In this study, we characterized the diferential DNA methylome landscapes of peripheral blood mononu‑ clear cells (PBMCs) from 76 canines using methylated CpG-binding domain sequencing (MBD-seq). Through gene set enrichment analysis, we discovered that genes involved in the growth and diferentiation of T- and B-cells are highly methylated in tumor PBMCs. We also revealed the increased methylation at single CpG resolution and reversed expression in representative marker genes regulating immune cell proliferation (BACH2, SH2D1A, TXK, UHRF1). Furthermore, we utilized the PBMC methylome to efectively diferentiate between benign and malignant tumors and the presence of mammary gland tumors through a machine-learning approach.     A sclerosing stromal tumor of the ovary with masculinization in a premenarchal girl

    A sclerosing stromal tumor of the ovary is an extremely rare benign tumor; it usually is found during the second and third decades of life. Patients present with pelvic pain or a palpable abdominal mass. Hormonal effects such as masculinization are uncommon. Here, an 11-year old premenarchal girl presented with deepening of the voice. In addition, clitoromegaly and hirsutism with a male suprapubic hair pattern were observed. The laboratory findings showed that the testosterone level was elevated to 3.67 ng/mL, andostenedione to above 10 ng/mL, dehydroepiandrosterone-sulfate to 346 µg/dL and 17-hydroxy progesterone (17-OHP) to 11.28 ng/mL. The chromosome evaluation revealed a 46,XX female karyotype. An adrenocorticotropic hormone stimulation test was performed. The 17-OHP to cortisol ratio in 30 minutes was 0.045, which suggested a heterozygote for the 21-hydroxylase deficiency. However, the CYP21A2 gene encoding steroid 21-hydroxylase showed normal. The pelvic ultrasound showed a heterogeneous mass consisting of predominantly solid tissue in the pelvic cavity. The pelvic magnetic resonance imaging revealed an 8.9×6.2×6.6 cm mass of the left ovary. A left oophrectomy was performed and microscopic examination confirmed a sclerosing stromal tumor. Immunohistochemical studies showed that the tumor was positive for smooth muscle actin and vimentin, but negative for S-100 protein and cytokeratin. Following surgery, the hormone levels returned to the normal range and the hirsutism resolved

    'Big Tigers, Big Data:' Learning Social Reactions to China’s Anticorruption Campaign through Online Feedback

    This study examines the effect of campaign-style anticorruption on political support using the case of China’s most recent anticorruption drive, which stands out for its harsh crackdown on high-ranking officials, or the “big tigers.” An exploratory text analysis of over 370,000 online comments on the downfall of the first 100 big tigers, from 2012 to 2015, reveals that public support for the top national leader who initiated the campaign significantly exceeds that afforded to anticorruption agencies and institutions. Further regression analyses show that support for the leaders vis-à-vis intuitions increases with the tigers’ party ranking. Findings suggest that while campaign-style enforcement can reinforce the central authority and magnify support for individual leaders, it may also marginalize the role of legal institutions crucial to long-term corruption control.published_or_final_versio

    The Effect of Apolipoprotein E Polymorphism on Lipid Levels in Korean Adults

    The aim of this study was to determine the effects of polymorphisms in the apolipoprotein E gene (APOE) on lipid levels in Korean adults and to investigate the interactions between these polymorphisms and environmental factors in determining lipid levels. We performed a cross-sectional study of 1,900 subjects (668 men and 1,232 women; 45-74 yr old) in Namwon, Korea, in 2004. APOE polymorphisms were determined by polymerase chain reaction and restriction enzyme analysis. Carriers of the APOE*E2 (E2) allele had significantly lower total cholesterol and low-density lipoprotein cholesterol (LDL-C) concentrations than did carriers of the APOE*E3 (E3) or APOE*E4 (E4) alleles, regardless of gender. The APOE allele type had significant effect on high-density lipoprotein cholesterol (HDL-C) and triglyceride levels in women, but not in men. The effect of APOE allele type on HDL-C levels was modified by age in women. In addition, in men, the effect of APOE allele type on triglyceride levels was modified by smoking. These findings highlight the important effect of gene-environment interactions on lipid levels

    NHERF2 specifically interacts with LPA(2) receptor and defines the specificity and efficiency of receptor-mediated phospholipase C-beta 3 activation

    Lysophosphatidic acid (LPA) activates a family of cognate G protein-coupled receptors and is involved in various pathophysiological processes. However, it is not clearly understood how these LPA receptors are specifically coupled to their downstream signaling molecules. This study found that LPA, but not the other LPA receptor isoforms, specifically interacts with Na+/H+ exchanger regulatory factor2 (NHERF2). In addition, the interaction between them requires the C-terminal PDZ domain-binding motif of LPA(2) and the second PDZ domain of NHERF2. Moreover, the stable expression of NHERF2 potentiated LPA-induced phospholipase C-beta (PLC-beta) activation, which was markedly attenuated by either a mutation in the PDZ-binding motif of LPA(2) or by the gene silencing of NHERF2. Using its second PDZ domain, NHERF2 was found to indirectly link LPA(2) to PLC-beta3 to form a complex, and the other PLC-beta isozymes were not included in the protein complex. Consistently, LPA(2)-mediated PLC-beta activation was specifically inhibited by the gene silencing of PLC-beta3. In addition, NHERF2 increases LPA-induced ERK activation, which is followed by cyclooxygenase-2 induction via a PLC-dependent pathway.     Quantitative Ultrasound of the Calcaneus in a Korean Population: Reference Data and Relationship to Bone Mineral Density Determined by Peripheral Dual X-ray Absorptiometry

    The aim of this study was to establish reference data for the quantitative ultrasound (QUS) of the calcaneus and for the bone mineral densities (BMD) of the calcaneus and distal forearm, and to evaluate the correlation between QUS parameters and BMD in a Korean population. We performed a cross-sectional study involving 3,053 subjects (1,225 men and 1,828 women). QUS was conducted on the calcaneus and was quantified as speed of sound (SOS, m/sec), broadband ultrasound attenuation (BUA, dB/MHz), and stiffness index. The BMD of the calcaneus and distal forearm were measured using dual X-ray absorptiometry. The peak mean values for the QUS parameters occurred in the 20 to 29-yr-old subjects of both sexes, with the exception of the BUA, which reached the highest values in women of 30-39 yr. For both sexes, the mean BMD of the calcaneus was highest in those 20-29 yr old and that of the distal forearm was highest in those 40-49 yr old. The correlations between the QUS and BMD results were found to be 0.41 to 0.73 in men and 0.51 to 0.76 in women. Theses data can serve as a reference values for both sexes in Korea
