    Genetic Deletion of Vasohibin-2 Exacerbates Ischemia-Reperfusion-Induced Acute Kidney Injury

    Acute kidney injury (AKI) has been increasingly recognized as a risk factor for transition to chronic kidney disease. Recent evidence suggests that endothelial damage in peritubular capillaries can accelerate the progression of renal injury. Vasohibin-2 (VASH2) is a novel proangiogenic factor that promotes tumor angiogenesis. However, the pathophysiological roles of VASH2 in kidney diseases remain unknown. In the present study, we examined the effects of VASH2 deficiency on the progression of ischemia-reperfusion (I/R) injury-induced AKI. I/R injury was induced by bilaterally clamping renal pedicles for 25 min in male wild-type (WT) andVash2homozygous knockout mice. Twenty-four hours later, I/R injury-induced renal dysfunction and tubular damage were more severe in VASH2-deficient mice than in WT mice, with more prominent neutrophil infiltration and peritubular capillary loss. After induction of I/R injury, VASH2 expression was markedly increased in injured renal tubules. These results suggest that VASH2 expression in renal tubular epithelial cells might be essential for alleviating I/R injury-induced AKI, probably through protecting peritubular capillaries and preventing inflammatory infiltration

    Esophageal metastasis of renal cell carcinoma resected by endoscopic submucosal dissection: a case report

    Background: Esophageal metastasis of renal cell carcinoma (RCC) is extremely rare. We have described herein a case of a 59-year-old man with esophageal metastasis of RCC that was endoscopically resected.Case presentation: The case was a 59-year-old man who had undergone left nephrectomy for renal clear cell carcinoma 17 years ago and splenectomy for splenic metastasis 3 years ago. Esophagogastroduodenoscopy (EGD) performed 9 years ago revealed a small reddish elevated lesion with a smooth surface in the middle esophagus; this lesion increased in size 4 years ago. However, no biopsy was performed. The lesion continued to grow in size and was found to have become nodular during the present observation. Biopsy revealed clear cell carcinoma. Endoscopic ultrasound (EUS) revealed that the lesion had not invaded the submucosa, and contrast-enhanced computed tomog-raphy did not reveal any other metastasis. The lesion was successfully removed en bloc via endoscopic submucosal dissection (ESD). Pathologically, the tumor was detected in the subepithelium with focal infiltration of the muscularis mucosa. It consisted of monotonous cells with small nuclei and a clear cytoplasm. Immunohistological findings indi-cated that the tumor was a metastasis of RCC. The lateral and vertical margins were noted to be free.Conclusions: We have presented herein a case of esophageal metastasis of RCC that had progressed over 9 years and was then resected en bloc through endoscopic submucosal dissection

    The Blimp1–Bcl6 axis is critical to regulate osteoclast differentiation and bone homeostasis

    Controlling osteoclastogenesis is critical to maintain physiological bone homeostasis and prevent skeletal disorders. Although signaling activating nuclear factor of activated T cells 1 (NFATc1), a transcription factor essential for osteoclastogenesis, has been intensively investigated, factors antagonistic to NFATc1 in osteoclasts have not been characterized. Here, we describe a novel pathway that maintains bone homeostasis via two transcriptional repressors, B cell lymphoma 6 (Bcl6) and B lymphocyte–induced maturation protein-1 (Blimp1). We show that Bcl6 directly targets ‘osteoclastic’ molecules such as NFATc1, cathepsin K, and dendritic cell-specific transmembrane protein (DC-STAMP), all of which are targets of NFATc1. Bcl6-overexpression inhibited osteoclastogenesis in vitro, whereas Bcl6-deficient mice showed accelerated osteoclast differentiation and severe osteoporosis. We report that Bcl6 is a direct target of Blimp1 and that mice lacking Blimp1 in osteoclasts exhibit osteopetrosis caused by impaired osteoclastogenesis resulting from Bcl6 up-regulation. Indeed, mice doubly mutant in Blimp1 and Bcl6 in osteoclasts exhibited decreased bone mass with increased osteoclastogenesis relative to osteoclast-specific Blimp1-deficient mice. These results reveal a Blimp1–Bcl6–osteoclastic molecule axis, which critically regulates bone homeostasis by controlling osteoclastogenesis and may provide a molecular basis for novel therapeutic strategies

    Characteristics of the chiral stationary phase with poly(l-phenylalanine) peptide selector, which was in α-helical state, was reported. Since environmental factors affect peptide conformation, the changes in enantioselectivity were examined depending on column temperature and mobile phase conditions (ionic strength, pH, mobile phase composition). Column temperature and pH drastically affected the enantioselectivity. Based on these changes, the relation between chiral recognition and secondary structure of the peptide selector was discussed. The column stability during sequential analysis under different separation conditions was also evaluated

    育児参加は父親にどのような影響を与えるか : 多胎児の父親と単胎児の父親との比較

    近年、男女共同参画社会の実現、少子化問題、子どもの社会性の発達や父親自身の人格的成長、QOLやワーク・ライフ・バランスなどとの関連において、父親の育児参加が注目されている。多胎児の父親は単胎児の父親より積極的に育児参加するものが多いとの報告があるが、育児参加が父親に与える影響について多胎児と単胎児の父親を比較した調査はない。そこで、本研究では、多胎児と単胎児の父親の「子育て観・次世代育成観」「母性神話」「仕事観」「子ども観」を調査した。ツインマザースクラブの会員1016名に自己記入式アンケートを郵送、211名の父親から回答を得た(回収率20.8%)。また、比較群として、小中一貫校の児童・生徒および大学生の単胎児の父親300名にアンケート調査を実施し、101名から回答を得た(回収率33.7%)。結果、「子どもが3歳になるまで、母親は育児に専念するほうがよい」「子どもを出産した後は、母親は仕事をやめたほうがよい」といった、いわゆる「三歳児神話」や「母性神話」については、多胎児の父親は単胎児の父親と比較して反対側が有意に多かった。「子育ては自分の自由な時間を奪う」「仕事は自分の自由な時間を奪う」「子育て中は、勤務時間を自分で調整できる方がよい」は多胎児の父親で賛成側が有意に多く、「育児休暇をとると昇進にひびく」は、多胎児の父親で反対側が有意に多かった。ふたごの父親は、単胎児の父親と比較して積極的に育児参加している者が多く、育児の担い手として実質的に育児に関わる経験が母性神話に対する価値観や子育て観、仕事観に影響している可能性が示唆された

    Abstract Introduction Blackcurrant (Ribs nigrum L.) is a classical fruit that has long been used to prepare juice, jam, liqueur, and sometimes medicines in Europe. Previously, we reported a genome defense effect by the antioxidative activity of several types of blackcurrant extracts (BCEs) in yeast and human cell gene mutation assays. In this study, we determined if BCE exerted radioprotective activity against DNA damage, chromosomal aberration, and gene mutations in the TK6 human lymphoblastoid cell line. We prepared aqueous BCE extracted from mature fruits cultivated in the Aomori Prefecture, Japan. Findings In the micronucleus test and TK gene mutation assay, TK6 cells were irradiated with 0, 0.125, 0.250, 0.500, and 1.000 Gy with or without 1.0 mg/mL BCE. Intracellular hydrogen peroxide (H2O2) was measured using the fluorescent probe BES-H2O2-Ac. Induction of micronuclei and gene mutations by γ-irradiation exposure was suppressed in combination with BCE. In addition, BCE reduced intracellular H2O2 levels caused by γ-irradiation. Conclusions Our findings clearly support the genome defense potential of blackcurrant against γ-induced DNA damage. We postulate that these genome defense activities are related to the antioxidant compounds in blackcurrant

    Preparation and characterization of poly(l-phenylalanine) chiral stationary phases with varying peptide length.

    Three new chiral stationary phases with different lengths of l-phenylalanine peptide were prepared by solid-phase synthesis with tert-butoxycarbonyl (Boc)-l-phenylalanine on silica. The effect of phenylalanine peptide length on enantioselectivity was studied. The best separation of R/S-warfarin was achieved by the chiral stationary phase with intermediate peptide length. These stationary phases were found to exist mainly in alpha-helical conformation by using FT-IR spectra. The end-capping reagents for the N-terminus of the peptide were also evaluated
