    Folic acid-functionalized human serum albumin nanocapsules for targeted drug delivery to chronically activated macrophages

    Activated synovial macrophages play a key role in Rheumatoid Arthritis (RA). Recent studies have shown that folate receptor beta (FRβ) is specifically expressed by activated macrophages. Therefore a folate-based nanodevice would provide the possibility of delivering therapeutic agents to activated macrophages without affecting normal cells and tissues. This study shows for the first time the sonochemical preparation of HSA nanocapsules avoiding toxic cross linking chemicals and emulsifiers used in other methods. Production of HSA nanocapsules was optimized leading to a diameter of 443.5 ± 9.0 nm and a narrow size distribution indicated by a polydispersity index (PDI) of 0.066 ± 0.080. Nanocapsules were surface modified with folic acid (FA) and the FA content was determined to be 0.38 and 6.42 molecules FA per molecule HSA, depending on the surplus of FA employed. Dynamic light scattering was used to determine size, PDI and zetapotential of the produced nanocapsules before and after surface modification. FA distribution on the surface of HSA nanocapsules was localized three-dimensionally after fluorescence labeling using confocal laser scanning microscopy (CLSM). Furthermore, specific binding and internalization of HSA nanocapsules by FRβ-positive and FRβ-negative macrophages, obtained from human peripheral blood mononuclear cells, was demonstrated by flow cytometry.     Assessment of liposome disruption to quantify drug delivery in vitro

    Efficient liposome disruption inside the cells is a key for success with any type of drug delivery system. The efficacy of drug delivery is currently evaluated by direct visualization of labeled liposomes internalized by cells, not addressing objectively the release and distribution of the drug. Here, we propose a novel method to easily assess liposome disruption and drug release into the cytoplasm. We propose the encapsulation of the cationic dye Hoechst 34,580 to detect an increase in blue fluorescence due to its specific binding to negatively charged DNA. For that, the dye needs to be released inside the cell and translocated to the nucleus. The present approach correlates the intensity of detected fluorescent dye with liposome disruption and consequently assesses drug delivery within the cells.Eugénia Nogueira (SFRH/BD/81269/2011), Célia F.     O PROGRAMA CIÊNCIA SEM FRONTEIRAS SOB O OLHAR DOS ESTUDANTES DO IFPB: EXPERIÊNCIA E PERSPECTIVAS

    Esta pesquisa teve por objetivo analisar o Programa Ciências sem Fronteiras enquanto política pública de internacionalização da educação superior, a partir da experiência de estudantes do Instituto Federal da Paraíba. Para isso, realizou-se uma pesquisa mista, em que se realizou, primeiramente, uma revisão bibliográfica sobre o contexto do Programa na Instituição. Posteriormente foi desenvolvida a pesquisa empírica, através da aplicação de questionário e de entrevista com os estudantes. O tratamento dos dados se deu através da análise descritiva e da análise de conteúdo. A partir das falas dos sujeitos referenciadas, identificou-se que a principal dificuldade dos alunos foi com relação ao conteúdo curricular das disciplinas e ao domínio do segundo idioma. Apesar disso, a maior parte dos entrevistados teve um aproveitamento considerável no Programa, que contribuiu em suas trajetórias acadêmicas e profissionais

    Recovery of rare earth elements from wastewater towards a circular economy

    The use of rare earth elements is a growing trend in diverse industrial activities, leading to the need for eco-friendly approaches to their efficient recovery and reuse. The aim of this work is the development of an environmentally friendly and competitive technology for the recovery of those elements from wastewater. Kinetic and equilibria batch assays were performed with zeolite, with and without bacterial biofilm, to entrap rare earth ions from aqueous solution. Continuous assays were also performed in column setups. Over 90% removal of lanthanum and cerium was achieved using zeolite as sorbent, with and without biofilm, decreasing to 70% and 80%, respectively, when suspended Bacillus cereus was used. Desorption from the zeolite reached over 60%, regardless of the tested conditions. When in continuous flow in columns, the removal yield was similar for all of the rare earth elements tested. Lanthanum and cerium were the elements most easily removed by all tested sorbents when tested in single- or multi-solute solutions, in batch and column assays.     Obesidade juvenil com enfoque na promoção da saúde: revisão integrativa

    Foi realizada uma revisão integrativa com o objetivo de sintetizar as contribuições das pesquisas em Enfermagemsobre obesidade juvenil com enfoque na promoção da saúde. Através do levantamento bibliográfico foram identificados20 artigos considerando os critérios: data, idioma e os descritores Enfermagem, adolescente, obesidade epromoção da saúde. O periódico Pediatric Nursing Journal apresentou 7 (35%) artigos. As publicações indexadasocorreram, nos últimos dez anos, nos idiomas português e inglês. Os resultados evidenciaram a construção do conhecimentocientífico da Enfermagem para o desenvolvimento de estratégias com enfoque na promoção da saúdena obesidade juvenil e, assim, contribuir para o desenvolvimento da profissão. Consideramos que a visualizaçãodo risco cumulativo que a obesidade juvenil apresenta em tornar o sujeito um adulto obeso é um dado precioso paraque o planejamento das ações de enfermagem direcionadas a essa população seja implementado e alcance resultadosefetivos

    Intervalos de referência de hemograma da população adulta brasileira: Pesquisa Nacional de Saúde

    Objective: to estimate the reference intervals (RIs) of complete blood count parameters in the Brazilian adult population. Methods: Cross-sectional study, with data from the National Health Survey (PNS), between 2014-2015. The final sample consisted of 2,803 adults. The final sample consisted of 2,803 adults. To establish the RI, exclusion criteria were applied, outliers were removed and partitions were made by sex, age and race/skin color. The non-parametric method was adopted. Differences were assessed using the Mann Withney and Kruskal Wallis tests (p≤0.05). Results: There were statistically significant differences for the following hematological parameters based on sex, red blood cells, hemoglobin, hematocrit, MCH, MCHC, eosinophils and absolute monocytes, neutrophils and platelets (p≤0.05). When analyzed by age, the RIs were statistically different in females for hematocrit, MCV, white blood cells and RDW and in males for red blood cells, white blood cells, eosinophils, mean platelet volume, MCV, RDW and MCH(p≤0.05). For race/color there were differences in the RIs for parameters of hemoglobin, MCH, MCHC, white blood cells and mean platelet volume, neutrophils and absolute eosinophils (p ≤ 0.05). Conclusion: The differences found in the RIs of some in blood count parameters in Brazilian adults reaffirm the importance of having their own laboratory reference standards. The results can support a more accurate interpretation of tests, adequate identification and disease prevention in Brazil.Objetivo: estimar os intervalos de referência (IR) de parâmetros de hemograma completo na população adulta brasileira. Métodos: Estudo transversal, com dados da Pesquisa Nacional de Saúde (PNS), entre 2014-2015. A amostra final constitui-se de 2.803 adultos. Para estabelecer os IR, aplicou-se critérios de exclusão, removeram-se outliers e feito particionamentos por sexo, idade e raça/cor da pele. Adotou-se o método não paramétrico.  As diferenças foram avaliadas pelos testes Mann Withney e Kruskal Wallis (p≤0,05). Resultados: houve diferenças estatisticamente significativas nos IR segundo sexo para glóbulos vermelhos, hemoglobina, hematócrito, HCM, CHCM, eosinófilos, monócitos, neutrófilos absolutos e plaquetas (p≤0,05). Quando analisados por idade,  houve diferenças nos IR de mulheres para hematócrito, VCM, glóbulos brancos e RDW e nos homens de glóbulos vermelhos, glóbulos brancos, eosinófilos, volume plaquetário médios, VCM, RDW e HCM (p≤0,05). Para raça/cor houve diferenças nos IR de hemoglobina, HCM, CHMC, glóbulos brancos e volume plaquetário médio, neutrófilos e eosinófilos absolutos (p ≤ 0,05). Conclusão: As diferenças encontradas nos IR de alguns em parâmetros de hemograma em adultos brasileiros  reafirmam a importância de se ter padrões de referência laborarias próprios. Os resultados podem subsidiar a interpretação mais precisa dos exames, identificação adequada e a prevenção de doenças no Brasil

    Panorama da infecção pelo SARS-COV-2 no Centro de Progressão Penitenciária do Butantan

    OBJECTIVE: To estimate the prevalence of exposure to the SARS-CoV-2 virus among individuals living in restricted freedom. METHODS: A seroprevalence survey was carried out with the population of the female penitentiary of the Centro de Progressão Penitenciária (CPP) in Butantan (municipality of São Paulo), between June 24 and August 20, 2020. During this period, according to the Secretariat of Penitentiary Administration (SAP), the positivity of rapid tests among inmates ranged from 65% to 78%. The evaluation method used in the study was the “One Step COVID-19” rapid test (chromatography), from the company Wondfo, also using the RT-PCR method in symptomatic participants to confirm the viral condition. The study population consisted of 879 female inmates and 170 employees of the institution. RESULTS: The prevalence of total antibodies (IgG/IgM) against the SARS-CoV-2 virus in the total population of 1049 study participants was 6.1%; among the population of 879 inmates, a prevalence of 5.8% was observed, and among the institution’s employees, 7.5%. CONCLUSIONS: The prevalence of covid-19 at the Butantan CPP was low, which is due to the implementation of simple prevention measures at the institution, such as the use of masks (with appropriate changes), emphasis on hygiene, hand washing and social distancing, in addition to other strategies, such as suspending inmates’ visits from relatives and friends and cutting back on elective medical appointments and outside work.OBJETIVO: Estimar a prevalência da exposição ao vírus SARS-CoV-2 entre indivíduos vivendo em restrição de liberdade. MÉTODOS: Foi realizado inquérito de soroprevalência com a população da penitenciária feminina do Centro de Progressão Penitenciária (CPP) do Butantan (município de São Paulo), entre 24 de junho e 20 de agosto de 2020. Nesse período, segundo a Secretaria de Administração Penitenciária (SAP), a positividade dos testes rápidos entre detentos variou de 65 a 78%. O método de avaliação utilizado no estudo foi o teste rápido “One Step COVID-19” (cromatografia), da empresa Wondfo, empregando-se também o método RT-PCR em participantes sintomáticos para confirmação do quadro viral. A população do estudo foi constituída por 879 reeducandas e 170 funcionários da instituição. RESULTADOS: A prevalência de anticorpos totais (IgG/IgM) contra o vírus SARS-CoV-2 na população total de 1.049 participantes do estudo foi de 6,1%; entre a população de 879 reeducandas foi observada prevalência de 5,8% e entre os servidores da instituição, 7,5%. CONCLUSÃO: Houve baixa prevalência de covid-19 no CPP do Butantan, o que se deve à implementação de medidas de prevenção simples na instituição, como o uso de máscaras (com trocas adequadas), ênfase na higiene, lavagem das mãos e distanciamento social, além de outras estratégias, como suspensão de visitas de familiares e amigos das reeducandas, cortes de consultas médicas eletivas e do trabalho externo

    Low-level laser therapy and Calendula officinalis in repairing diabetic foot ulcers

    Abstract OBJECTIVE To evaluate the effects of low-level laser therapy isolated and associated with Calendula officinalis oil in treating diabetic foot ulcers. METHOD An experimental, randomized, controlled, prospective, interventional clinical case study using a quantitative approach. The sample consisted of 32 diabetic patients of both genders. Participants were randomly divided into four groups. Doppler Ultrasound evaluation of the Ankle-Brachial Index, brief pain inventory and analog pain scale were performed at baseline and after 30 days. RESULTS Reduced pain was observed in the Low-level laser therapy and Low-level laser therapy associated with Essential Fatty Acids groups (p<0.01). Regarding the Ankle-Brachial Index and Doppler Ultrasound, all groups remained stable. By analyzing lesion area reduction, Low-level laser therapy associated with Essential fatty acids group showed a significance of p=0.0032, and the Low-level laser therapy group showed p=0.0428. CONCLUSION Low-level laser therapy, performed alone or associated with the Calendula officinalis oil was effective in relieving pain and accelerating the tissue repair process of diabetic foot
