    Prognostic Impact of Hypoxia-Inducible miRNA-210

    Objective. The aim of this study was to investigate the prognostic value of MicroRNA-210 (miR-210) expression in patients with non-small-cell lung cancer (NSCLC). Methods. We examined the miR-210 expression of samples of 80 patients, who underwent surgical resection at Fukushima Medical University from 2004 to 2007, by using quantitative RT-PCR. The relationship between miR-210 expression and clinicopathological factors as well as histological subtype was statistically analyzed. Results. miR-210 expression showed an inverse correlation with disease-free and overall survival in patients with NSCLC. Significant correlations were found between miR-210 expression and lymph node metastasis, late disease stages, and poor prognosis in patients with adenocarcinoma. Multivariate Cox analysis indicated that miR-210 expression was an independent prognostic factor for disease-free survival in patients with adenocarcinoma. Conclusions. We showed that miR-210 may be a prognostic biomarker for patients with NSCLC, especially for those with lung adenocarcinoma

    Novel quantitative immunohistochemical analysis for evaluating PD-L1 expression with phosphor-integrated dots for predicting the efficacy of patients with cancer treated with immune checkpoint inhibitors

    IntroductionProgrammed cell death ligand 1 (PD-L1) expression in tumor tissues is measured as a predictor of the therapeutic efficacy of immune checkpoint inhibitors (ICIs) in many cancer types. PD-L1 expression is evaluated by immunohistochemical staining using 3,3´-diaminobenzidine (DAB) chronogenesis (IHC-DAB); however, quantitative and reproducibility issues remain. We focused on a highly sensitive quantitative immunohistochemical method using phosphor-integrated dots (PIDs), which are fluorescent nanoparticles, and evaluated PD-L1 expression between the PID method and conventional DAB method.MethodsIn total, 155 patients with metastatic or recurrent cancer treated with ICIs were enrolled from four university hospitals. Tumor tissue specimens collected before treatment were subjected to immunohistochemical staining with both the PID and conventional DAB methods to evaluate PD-L1 protein expression.ResultsPD-L1 expression assessed using the PID and DAB methods was positively correlated. We quantified PD-L1 expression using the PID method and calculated PD-L1 PID scores. The PID score was significantly higher in the responder group than in the non-responder group. Survival analysis demonstrated that PD-L1 expression evaluated using the IHC-DAB method was not associated with progression-free survival (PFS) or overall survival (OS). Yet, PFS and OS were strikingly prolonged in the high PD-L1 PID score group.ConclusionQuantification of PD-L1 expression as a PID score was more effective in predicting the treatment efficacy and prognosis of patients with cancer treated with ICIs. The quantitative evaluation of PD-L1 expression using the PID method is a novel strategy for protein detection. It is highly significant that the PID method was able to identify a group of patients with a favorable prognosis who could not be identified by the conventional DAB method

    The Hyper Suprime-Cam SSP survey: Overview and survey design

    Hyper Suprime-Cam (HSC) is a wide-field imaging camera on the prime focus of the 8.2-m Subaru telescope on the summit of Mauna Kea in Hawaii. A team of scientists from Japan, Taiwan, and Princeton University is using HSC to carry out a 300-night multi-band imaging survey of the high-latitude sky. The survey includes three layers: the Wide layer will cover 1400 deg2 in five broad bands (grizy), with a 5 σ point-source depth of r ≈ 26. The Deep layer covers a total of 26 deg2 in four fields, going roughly a magnitude fainter, while the UltraDeep layer goes almost a magnitude fainter still in two pointings of HSC (a total of 3.5 deg2). Here we describe the instrument, the science goals of the survey, and the survey strategy and data processing. This paper serves as an introduction to a special issue of the Publications of the Astronomical Society of Japan, which includes a large number of technical and scientific papers describing results from the early phases of this survey

    Design and Implementation of an On-Demand Home Power Management System based on a Hierarchical Protocol

    2015 IEEE 39th Annual Computer Software and Applications Conference Workshop (COMPSACW): Taichung, Taiwan - July 1-5, 2015We have designed and implemented an on-demand home power management system for saving electric power consumption reliably and systematically without affecting our Quality of Life. The system is based on protocol hierarchy composed of the request/response layer, the path control layer, and the physical layer. Based on the layered protocol architecture, it becomes possible to treat power allocation, power flow setting, and power transmission separately. This makes the on-demand power management system flexibly extensible. In this paper, we describe a use case of our system for power management at a single home, and show how power is saved with least degradation of the Quality of Life in experiments done in a demonstration house

    The Online Graph Exploration Problem on Restricted Graphs

    The purpose of the online graph exploration problem is to visit all the nodes of a given graph and come back to the starting node with the minimum total traverse cost. However, unlike the classical Traveling Salesperson Problem, information of the graph is given online. When an online algorithm (called a searcher) visits a node ν, then it learns information on nodes and edges adjacent to ν. The searcher must decide which node to visit next depending on partial and incomplete information of the graph that it has gained in its searching process. The goodness of the algorithm is evaluated by the competitive analysis. If input graphs to be explored are restricted to trees, the depth-first search always returns an optimal tour. However, if graphs have cycles, the problem is non-trivial. In this paper we consider two simple cases. First, we treat the problem on simple cycles. Recently, Asahiro et al. proved that there is a 1.5-competitive online algorithm, while no online algorithm can be (1.25-ε)-competitive for any positive constant ε. In this paper, we give an optimal online algorithm for this problem; namely, we give a frac1+sqrt32(simeq1.366)frac{1+sqrt{3}}{2}(simeq1.366)-competitive algorithm, and prove that there is no (frac1+sqrt32epsilon)(frac{1+sqrt{3}}{2}-epsilon)-competitive algorithm for any positive constant ε. Furthermore, we consider the problem on unweighted graphs. We also give an optimal result; namely we give a 2-competitive algorithm and prove that there is no (2-ε)-competitive online algorithm for any positive constant ε

    Garcin syndrome caused by sphenoid bone metastasis of lung cancer: a case study

    Abstract Background Garcin syndrome, which consists of unilateral palsies of almost all cranial nerves without either sensory or motor long-tract disturbances or intracranial hypertension, can be caused by malignant tumors at the skull base. The case of a patient with lung cancer that metastasized to the sphenoid bone and resulted in Garcin syndrome is presented. Case presentation A 76-year-old woman was diagnosed as having non-small cell lung cancer with pericardial and diaphragmatic infiltration, cT4N1M0, stage 3A. The left lower lobectomy with concomitant resection of the pericardium and diaphragm was performed. The pathological diagnosis was pleomorphic carcinoma, pT2bN0M0, stage 1B. She was then followed in the surgery clinic, and 2 months after surgery, she visited an emergency room complaining of headache and diplopia. Neurological examination showed the left IV, V1, and VI cranial nerve palsies. Metastatic tumor with bone destruction was found in the left sphenoid sinus on head computed tomography (CT) and contrast magnetic resonance imaging (MRI), and she was diagnosed with Garcin syndrome caused by sphenoid bone metastasis of lung cancer. Irradiation was performed as palliative treatment, but her neurological findings did not improve. Her general condition gradually worsened, and she died 5 months after surgery. Conclusions Bone metastasis of lung cancer occurs frequently, but sphenoid bone metastasis is extremely rare. In this case report, Garcin syndrome caused by lung cancer is discussed in the context of the few previous reports

    Bactericidal and virucidal mechanisms in the alkaline disinfection of compost using calcium lime and ash

    In the present study, the bactericidal and virucidal mechanisms in the alkaline disinfection of compost with calcium lime and ash were investigated. Two indicator microorganisms, Escherichia coli and MS2 coliphage, were used as surrogates for enteric pathogens. The alkaline-treated compost with calcium oxide (CaO) or ash resulted primarily in damage to the outer membrane and enzyme activities of E. coli. The alkaline treatment of compost also led to the infectivity loss of the coliphage because of the partial capsid damage and RNA exteriorization due to a raised pH, which is proportional to the amount of alkaline agents added. These results indicate that the alkaline treatment of compost using calcium oxide and ash is effective and can contribute to the safe usage of compost from a mixing type dry toilet
