    Reproducibility of electrical caries measurements: A technical problem?

    The currently available instrument for electrical detection of occlusal caries lesions {[}Electronic Caries Monitor (ECM)] uses a site-specific measurement with co-axial air drying. The reproducibility of this method has been reported to be fair to good. It was noticed that the measurement variation of this technique appeared to be non-random. It was the aim of this study to analyse how such a non-random reproducibility pattern arises and whether it could be observed for other operators and ECM models. Analysis of hypothetical measurement pairs showed that the pattern was related to measurements at the high and low end of the measurement range for the instrument. Data sets supplied by other researchers to a varying degree showed signs of a similar non-random pattern. These data sets were acquired at different locations, by different operators and using 3 different ECM models. The frequency distribution of measurements in all cases showed a single or double end-peaked distribution shape. It was concluded that the pattern was a general feature of the measurement method. It was tentatively attributed to several characteristics such as a high value censoring, insufficient probe contact and unpredictable probe contact.     Diversification of myco-heterotrophic angiosperms: evidence from Burmanniaceae.

    Background - Myco-heterotrophy evolved independently several times during angiosperm evolution. Although many species of myco-heterotrophic plants are highly endemic and long-distance dispersal seems unlikely, some genera are widely dispersed and have pantropical distributions, often with large disjunctions. Traditionally this has been interpreted as evidence for an old age of these taxa. However, due to their scarcity and highly reduced plastid genomes our understanding about the evolutionary histories of the angiosperm myco-heterotrophic groups is poor. Results - We provide a hypothesis for the diversification of the myco-heterotrophic family Burmanniaceae. Phylogenetic inference, combined with biogeographical analyses, molecular divergence time estimates, and diversification analyses suggest that Burmanniaceae originated in West Gondwana and started to diversify during the Late Cretaceous. Diversification and migration of the species-rich pantropical genera Burmannia and Gymnosiphon display congruent patterns. Diversification began during the Eocene, when global temperatures peaked and tropical forests occurred at low latitudes. Simultaneous migration from the New to the Old World in Burmannia and Gymnosiphon occurred via boreotropical migration routes. Subsequent Oligocene cooling and breakup of boreotropical flora ended New-Old World migration and caused a gradual decrease in diversification rate in Burmanniaceae. Conclusion - Our results indicate that extant diversity and pantropical distribution of myco-heterotrophic Burmanniaceae is the result of diversification and boreotropical migration during the Eocene when tropical rain forest expanded dramaticall

    Lattice Boltzmann simulations of fluid flow in continental carbonate reservoir rocks and in upscaled rock models generated with multiple-point geostatistics

    Microcomputed tomography (mu CT) and Lattice Boltzmann Method (LBM) simulations were applied to continental carbonates to quantify fluid flow. Fluid flow characteristics in these complex carbonates with multiscale pore networks are unique and the applied method allows studying their heterogeneity and anisotropy. 3D pore network models were introduced to single-phase flow simulations in Palabos, a software tool for particle-based modelling of classic computational fluid dynamics. In addition, permeability simulations were also performed on rock models generated with multiple-point geostatistics (MPS). This allowed assessing the applicability of MPS in upscaling high-resolution porosity patterns into large rock models that exceed the volume limitations of the mu CT. Porosity and tortuosity control fluid flow in these porous media. Micro-and mesopores influence flow properties at larger scales in continental carbonates. Upscaling with MPS is therefore necessary to overcome volume-resolution problems of CT scanning equipment. The presented LBM-MPS workflow is applicable to other lithologies, comprising different pore types, shapes, and pore networks altogether. The lack of straightforward porosity-permeability relationships in complex carbonates highlights the necessity for a 3D approach. 3D fluid flow studies provide the best understanding of flow through porous media, which is of crucial importance in reservoir modelling

    Mononuclear but not polymorphonuclear phagocyte depletion increases circulation times and improves mammary tumor-homing efficiency of donor bone marrow-derived monocytes

    Tumor associated macrophages are an essential part of the tumor microenvironment. Consequently, bone marrow-derived monocytes (BMDMs) are continuously recruited to tumors and are therefore seen as ideal delivery vehicles with tumor-targeting properties. By using immune cell depleting agents and macroscopic in vivo fluorescence imaging, we demonstrated that removal of endogenous monocytes and macrophages (but not neutrophils) leads to an increased tumor accumulation of exogenously administered BMDMs. By means of intravital microscopy (IVM), we confirmed our macroscopic findings on a cellular level and visualized in real time the migration of the donor BMDMs in the tumors of living animals. Moreover, IVM also revealed that clodronate-mediated depletion drastically increases the circulation time of the exogenously administered BMDMs. In summary, these new insights illustrate that impairment of the mononuclear phagocyte system increases the circulation time and tumor accumulation of donor BMDMs

Gestreefd wordt naar een duurzaam en zo voorspelbaar mogelijk resultaat, waarbij zoveel mogelijk tandweefsel wordt gespaard. Aan het eind van dit literatuuroverzicht worden uitgangspunten geformuleerd voor een wortelstift die, voor standaardsituaties, het ideaalbeeld zo dicht mogelijk benadert
