    Antitumoral activity of the mithralog EC-8042 in triple negative breast cancer linked to cell cycle arrest in G2

    This is an open-access article distributed under the terms of the Creative Commons Attribution License.Triple negative breast cancer (TNBC) is an aggressive form of breast cancer. Despite response to chemotherapy, relapses are frequent and resistance to available treatments is often observed in the metastatic setting. Therefore, identification of new therapeutic strategies is required. Here we have investigated the effect of the mithramycin analog EC-8042 (demycarosil-3D-β-D-digitoxosyl mithramycin SK) on TNBC. The drug caused a dose-dependent inhibition of proliferation of a set of TNBC cell lines in vitro, and decreased tumor growth in mice xenografted with TNBC cells. Mechanistically, EC-8042 caused an arrest in the G2 phase of the cell cycle, coincident with an increase in pCDK1 and Wee1 levels in cells treated with the drug. In addition, prolonged treatment with the drug also causes apoptosis, mainly through caspaseindependent routes. Importantly, EC-8042 synergized with drugs commonly used in the therapy of TNBC in vitro, and potentiated the antitumoral effect of docetaxel in vivo. Together, these data suggest that the mithralog EC-8042 exerts an antitumoral action on TNBC cells and reinforces the action of standard of care drugs used in the therapy of this disease. These characteristics, together with a better toxicology profile of EC-8042 with respect to mithramycin, open the possibility of its clinical evaluation.The Cancer Research Institute receives support from the European Community through the regional development funding program (FEDER), and from the Fundación Ramón Areces.     Europium-doped NaGd(WO4)2 nanophosphors: synthesis, luminescence and their coating with fluorescein for pH sensing

    Uniform Eu-doped NaGd(WO4)2 nanophosphors with a spherical shape have been synthesized for the first time by using a wet chemistry method based on a homogeneous precipitation process at low temperature (120 °C) in ethylene glycol/water mixtures. The obtained nanoparticles crystallized into the tetragonal structure and presented polycrystalline character. The europium content in such phosphors has been optimized through the analysis of the luminescence dynamics (lifetime measurements).     Synthesis and Functionalization of Monodisperse Near-ultraviolet and Visible Excitable Multifunctional Eu3+, Bi3+:REVO4 Nanophosphors for Bioimaging and Biosensing Applications

    Near-ultraviolet and visible excitable Eu- and Bi-doped NPs based on rare earth vanadates (REVO4, RE = Y, Gd) have been synthesized by a facile route from appropriate RE precursors, europium and bismuth nitrate, and sodium orthovanadate, by homogeneous precipitation in an ethylene glycol/water mixture at 120 °C. The NPs can be functionalized either by a one-pot synthesis with polyacrylic acid (PAA) or by a Layer-by-Layer approach with poly(allylamine hydrochloride) (PAH) and PAA. In the first case, the particle size can also be tuned by adjusting the amount of PAA. The Eu- Bi-doped REVO4 based nanophosphors show the typical red luminescence of Eu(III), which can be excited through an energy transfer process from the vanadate anions, resulting in a much higher luminescence intensity in comparison to the direct excitation of the europium cations. The incorporation of Bi into the REVO4 structure shifts the original absorption band of the vanadate anions towards longer wavelengths, giving rise to nanophosphors with an excitation maximum at 342 nm, which can also be excited in the visible range. The suitability of such nanophosphors for bioimaging and biosensing applications, as well as their colloidal stability in different buffer media of biological interest, their cytotoxicity, their degradability at low pH, and their uptake by HeLa cells have been evaluated.     Rare earth based nanostructured materials: Synthesis, functionalization, properties and bioimaging and biosensing applications

    Rare earth based nanostructures constitute a type of functional materials widely used and studied in the recent literature. The purpose of this review is to provide a general and comprehensive overview of the current state of the art, with special focus on the commonly employed synthesis methods and functionalization strategies of rare earth based nanoparticles and on their different bioimaging and biosensing applications. The luminescent (including downconversion, upconversion and permanent luminescence) and magnetic properties of rare earth based nanoparticles, as well as their ability to absorb X-rays, will also be explained and connected with their luminescent, magnetic resonance and X-ray computed tomography bioimaging applications, respectively. This review is not only restricted to nanoparticles, and recent advances reported for in other nanostructures containing rare earths, such as metal organic frameworks and lanthanide complexes conjugated with biological structures, will also be commented on.European Union 267226Ministerio de Economía y Competitividad MAT2014-54852-

    The current work began with the observation of the needs of high school students and their constant concern for the level of their development in communication skills according to the social standards of their context. Two instruments were used: The Sternberg (adapted) questionnaire (STAT), which evaluates the successful intelligence of students, where systematized quantitative information is collected as well as the teacher’s observation card to the student, for the qualitative information of the three categories of communication skills. The design is of a mixed type. Several reference works were considered in order to contrast and triangulate according to experts’ opinions

    Improvements in the Academic Satisfaction of University Students Through the Effective Use of Learning Management Systems and Pedagogical Innovations

    The fast speed of technology has meant that the educational environment has altered characteristics of its environment, making it difficult for various colleges and professional schools to understand the need to incorporate these changes into their educational offerings. In today's interconnected world, a curriculum based on the development of professional competencies and scientific research cannot prepare oblivious students for professional success whether nationally or internationally. Students participating in diverse programs require a flexible curriculum that helps them identify and develop the knowledge, skills, and mindset they will need to succeed in the global markets in which they wish to work. To better prepare students to compete in global markets, it is recommended that a conceptual framework be built to align education with changes in technology and the global environment. The overall objective of this study was to investigate how learning management and innovation affect students' perceptions of their educational experience. Quantitative, fundamental-substantive, explanatory, cross-sectional, and non-experimental methods were used. Random sampling was chosen for practical reasons, and135 university students were selected. The findings showed very high reliability (measured by Cronbach's alpha). Also, multinomial logistic regression examined the data collected after service delivery. Since the tests showed no correlation between the variables, the null hypothesis was accepted and ruled out the significance of the impact of the variable

    Estrés laboral y escolar en la vida familiar durante la epidemia de COVID-19 (caso de estudio)

    The International Labour Organization (2016) defines stress as the psychological and physical reaction of the individual to a situation that threatens their balance. The objective of the research is to demonstrate the incidence of work and academic stress in the family dynamics of the Nursing Rotation Intern of a Public University of Ecuador during the pandemic, 2023. A quantitative approach was used, descriptive, correlational and explanatory, with a positivist philosophical stance. A population of 180 students who are in their final phase of the Nursing Degree was approached, to whom three pre-established instruments were applied with adaptations made and validated by the judgment of 7 experts, surveys that have allowed the appropriate collection of quantitative data with emphasis on reliability, construct validity and objectivity in the treatment of information and understanding of the data. A descriptive analysis was obtained, characterizing the study population and demonstrating the hypotheses raised through logistic regression, weighting the value of the variables and their indicators of the instruments. The variables studied have significant associations, proving the researcher's hypothesis. It is concluded that there is an impact of work and academic stress on the family dynamics of the student who is taking their pre-professional nursing internships.La Organización Internacional del Trabajo (2016), define el estrés como la reacción psicológica y física del individuo ante una situación que atenta contra su equilibrio. El objetivo de la investigación de demostrar la incidencia del estrés laboral y académico en la dinámica familiar del Interno Rotativo de Enfermería de una Universidad Pública del Ecuador durante la pandemia, 2023. Se utilizó un enfoque cuantitativo, nivel descriptivo, correlacional y explicativo, con una postura filosófica positivista. Se abordó a una población de 180 estudiantes que cursan su fase final de la Carrera de Enfermería, a quienes se les aplicó tres instrumentos preestablecidos con adaptaciones efectuadas y validadas por el juicio de 7 expertos, encuestas que han permitido recolectar apropiadamente datos cuantitativos con énfasis en la fiabilidad, validez de constructo y objetividad en el tratamiento de información y comprensión de los datos. Se ha obtenido el análisis descriptivo, caracterizando a la población de estudio y demostrándose las hipótesis planteadas a través de la regresión logística, ponderando el valor de las variables y sus indicadores de los instrumentos. Las variables estudiadas tienen asociaciones significativas, comprobándose la hipótesis del investigador. Se concluye que existe impacto del estrés laboral y académico en la dinámica familiar del estudiante que cursa sus prácticas preprofesionales de Enfermería

    Predictive Power of the "Trigger Tool" for the detection of adverse events in general surgery: a multicenter observational validation study

    Background In spite of the global implementation of standardized surgical safety checklists and evidence-based practices, general surgery remains associated with a high residual risk of preventable perioperative complications and adverse events. This study was designed to validate the hypothesis that a new “Trigger Tool” represents a sensitive predictor of adverse events in general surgery. Methods An observational multicenter validation study was performed among 31 hospitals in Spain. The previously described “Trigger Tool” based on 40 specific triggers was applied to validate the predictive power of predicting adverse events in the perioperative care of surgical patients. A prediction model was used by means of a binary logistic regression analysis. Results The prevalence of adverse events among a total of 1,132 surgical cases included in this study was 31.53%. The “Trigger Tool” had a sensitivity and specificity of 86.27% and 79.55% respectively for predicting these adverse events. A total of 12 selected triggers of overall 40 triggers were identified for optimizing the predictive power of the “Trigger Tool”. Conclusions The “Trigger Tool” has a high predictive capacity for predicting adverse events in surgical procedures. We recommend a revision of the original 40 triggers to 12 selected triggers to optimize the predictive power of this tool, which will have to be validated in future studies

    Telemedicine and medical education in the COVID-19 pandemic: Achievements, challenges and prospects

    Due to the global urgency produced by the Covid-19 pandemic, the development of alternative, viable processes, accessible through low-cost and technological resources, became more than necessary. Such is the case of telemedicine which, since its appearance towards the end of the 1960s through telephone consultations, has been transformed according to technological development. Information and communication technologies currently allow the provision of information and medical care services in diverse and distant locations, which is why it turns out to be relevant in the case of the current pandemic. This review work is aimed at examining the current state of the scope in the use of telemedicine that occurred during the current Covid-19 pandemic, its link in the case of medical education in various realities; as well as the analysis, evaluation and proposals on the benefits and concomitant challenges regarding telemedicine in the future
