    Evaluación de la alteración de biochars utilizados como enmienda de un cultivo de girasol bajo condiciones de clima mediterráneo

El biochar puede mejorar las propiedades físicas y químicas del suelo, y por tanto incrementar la calidad agronómica del mismo. Además se considera una estrategia eficaz para mitigar la emisión de dióxido de carbono a la atmósfera, ya que es rico en carbono recalcitrante funcionando como sumidero de carbono. Diversos factores como el tipo de materia prima, así como las condiciones de pirólisis determinarán la composición, estructura y propiedades de los biochars, lo queocasiona una gran heterogeneidad de los productos. Aún no se conoce suficientemente como afecta la degradación al biochar en condiciones de campo. Es necesario conocer cómo se modifica la composición de esta materia orgánica pirogénica una vez es incorporada al suelo, con este objetivo principal se sometió a cinco tipos diferentes de biochars a un envejecimiento en condiciones de clima Mediterráneo. Para ello, se mezclaron las muestras de biochar con un cambisol de la finca experimental “La Hampa” a modo de enmienda. Posteriormente se sembraron plantas de girasol, que fueron cosechadas a los 6 meses de su germinación. A los 6, 12 y 24 meses de iniciado el experimento se tomaron muestras de suelo y de biochar para su estudio. Con el fin de poder conocer las alteraciones en la composición y propiedades del biochar, se han realizado análisis de C y N, de pH, conductividad eléctrica y la determinación de la fragmentación. Además, el uso de la Resonancia Magnética Nuclear de 13C en estado sólido nos permitió observar los cambios en la funcionalidad de este material.M.     Transcriptomic profiling of skeletal muscle reveals candidate genes influencing muscle growth and associated lipid composition in Portuguese local pig breeds

    Gene expression is one of the main factors to influence meat quality by modulating fatty acid metabolism, composition, and deposition rates in muscle tissue. This study aimed to explore the transcriptomics of the Longissimus lumborum muscle in two local pig breeds with distinct genetic background using next-generation sequencing technology and Real-Time qPCR. RNA-seq yielded 49 differentially expressed genes between breeds, 34 overexpressed in the Alentejano (AL) and 15 in the Bísaro (BI) breed. Specific slow type myosin heavy chain components were associated with AL (MYH7) and BI (MYH3) pigs, while an overexpression of MAP3K14 in AL may be associated with their lower loin proportion, induced insulin resistance, and increased inflammatory response via NFkB activation. Overexpression of RUFY1 in AL pigs may explain the higher intramuscular (IMF) content via higher GLUT4 recruitment and consequently higher glucose uptake that can be stored as fat. Several candidate genes for lipid metabolism, excluded in the RNA-seq analysis due to low counts, such as ACLY, ADIPOQ, ELOVL6, LEP and ME1 were identified by qPCR as main gene factors defining the processes that influence meat composition and quality. These results agree with the fatter profile of the AL pig breed and adiponectin resistance can be postulated as responsible for the overexpression of MAP3K140s coding product NIK, failing to restore insulin sensitivity

    Responses of a native and a recent invader snail to warming and dry conditions: the case of the lower Ebro River

    Aquatic habitats have been highly modified by human actions that reduce their native diversity and create conditions suitable for tolerant alien species. Pomacea maculata was detected in 2009 in both the alluvial plain and the final stretch of the Ebro River. Since then, a permanent population has stabilized in the littoral area of the river where the water level fluctuates according to the river discharge. Melanopsis tricarinata is an endemic snail species highly affected by the reduction in its natural habitat. Currently, the two species do not share the same reaches in the river, but the possibility exists, as the distribution of the P. maculata is constantly increasing. This study aims to analyse the diets and to assess the responses of both snails to global change. The diet of both species was analysed in the field and their responses to water warming and dryness compared under laboratory conditions. This study includes the calculation of future river water temperatures based on air temperature projections. In addition, based on water discharge management scenarios, the study estimated the increase in dry river bed area. The diet of both snail species was similar and based on Cladophora. P. maculata better resisted high temperatures and dry conditions than M. tricarinata. The projections of water temperatures showed an increase in daily temperatures, especially in summer. The hydraulic model suggested that a relevant increase in dry river bed areas will occur. Overall, these results provide insight into the global change factors that could favour P. maculata spread in the river and the reduction in suitable habitat for M. tricarinata and will be useful for future decisions of water discharge management

    Comparative Transcriptomic Analysis of Subcutaneous Adipose Tissue from Local Pig Breeds

    When compared to modern lean-type breeds, Portuguese local Alentejano (AL) and Bísaro (BI) pig breeds present a high potential for subcutaneous and intramuscular fat (IMF) deposition which contributes for better meat quality. The aim of this work was to explore the genome function to better understand the underlying physiological mechanisms associated with body fat accretion. Dorsal subcutaneous fat samples were collected at slaughter from adult animals (n = 4 for each breed) with ~150 kg body weight. Total RNA was obtained and sequenced for transcriptome analysis using DESeq2. A total of 458 di erentially expressed (DE) genes (q-value < 0.05) were identified, with 263 overexpressed in AL and 195 in BI. Key genes involved in de novo fatty acid biosynthesis, elongation and desaturation were upregulated in AL such as ACLY, FASN, ME1, ELOVL6 and SCD. A functional enrichment analysis of the DE genes was performed using Ingenuity Pathway Analysis. Cholesterol synthesis is suggested to be higher in AL via SREBF2, SCAP and PPARG, while lipolytic activity may be more active in BI through GH and AMPK signalling. Increased signalling of CD40 together with the predicted activation of INSIG1 and INSIG2 in BI suggests that this breed is more sensitive to insulin whereas the AL is less sensitive like the Iberian breed

    Substituting a copper atom modifies the melting of aluminum clusters

    Producción CientíficaHeat capacities have been measured for Al(n−1)Cu− clusters (n = 49–62) and compared with results for pure Aln+ clusters. Al(n−1)Cu− and Aln+ have the same number of atoms and the same number of valence electrons (excluding the copper d electrons). Both clusters show peaks in their heat capacities that can be attributed to melting transitions; however, substitution of an aluminum atom by a copper atom causes significant changes in the melting behavior. The sharp drop in the melting temperature that occurs between n = 55 and 56 for pure aluminum clusters does not occur for the Al(n−1)Cu− analogs. First-principles density-functional theory has been used to locate the global minimum energy structures of the doped clusters. The results show that the copper atom substitutes for an interior aluminum atom, preferably one with a local face-centered-cubic environment. Substitution does not substantially change the electronic or geometric structures of the host cluster unless there are several Aln+ isomers close to the ground state. The main structural effect is a contraction of the bond lengths around the copper impurity, which induces both a contraction of the whole cluster and a stress redistribution between the Al–Al bonds. The size dependence of the substitution energy is correlated with the change in the latent heat of melting on substitution

    Identification of IL-6 Signalling Components as Predictors of Severity and Outcome in COVID-19

    IL-6 is one of the major mediators of the hyper-inflammatory responses with complex biological functions as it can signal via different modes of action. IL-6 by classical signalling has anti-inflammatory and antibacterial activities, while trans-signalling mediates pro-inflammatory effects. The net biological effect of IL-6 is established by multiple factors beyond its absolute concentration. Here, we assess the relationship between IL-6 signalling variables [IL-6, soluble IL-6R (sIL-6R) and soluble gp130 (sgp130)] and outcomes in a cohort of 366 COVID-19 patients. The potential trans-signalling was evaluated by a ratio between the pro-inflammatory binary IL-6:sIL-6R complex and the inactive ternary IL-6:sIL-6R:sgp130 complex (binary/ternary complex) and the fold molar excess of sgp130 over sIL-6R (FME). Our data provide new evidence that high levels of IL-6, sIL-6R, sgp130, binary/ternary complex ratio, and low FME are independent predictors of COVID-19 severity in survivor patients (without death), and the combination of IL-6 + sIL-6R + sgp130 exhibited the most robust classification capacity. Conversely, in a subgroup of patients with a very poor prognosis, we found that high levels of IL-6 and low levels of sIL-6R, sgp130, and binary/ternary complex ratio were predictors of death. In this context, the highest predictive capacity corresponded to the combined analysis of IL-6 + FME + lymphopenia + creatinine.     Anticoagulant therapy for splanchnic vein thrombosis: an individual patient data meta-analysis

    Robust evidence on the optimal management of splanchnic vein thrombosis (SVT) is lacking. We conducted an individual-patient meta-analysis to evaluate the effectiveness and safety of anticoagulation for SVT. Medline, Embase, and clincaltrials.gov were searched up to June 2021 for prospective cohorts or randomized clinical trials including patients with SVT. Data from individual datasets were merged, and any discrepancy with published data was resolved by contacting study authors. Three studies of a total of 1635 patients were included. Eighty-five percent of patients received anticoagulation for a median duration of 316 days (range, 1-730 days). Overall, incidence rates for recurrent venous thromboembolism (VTE), major bleeding, and mortality were 5.3 per 100 patient-years (p-y; 95% confidence interval [CI], 5.1-5.5), 4.4 per 100 p-y (95% CI, 4.2-4.6), and 13.0 per 100 p-y (95% CI, 12.4-13.6), respectively. The incidence rates of all outcomes were lower during anticoagulation and higher after treatment discontinuation or when anticoagulation was not administered. In multivariable analysis, anticoagulant treatment appeared to be associated with a lower risk of recurrent VTE (hazard ratio [HR], 0.42; 95% CI, 0.27-0.64), major bleeding (HR, 0.47; 95% CI, 0.30-0.74), and mortality (HR, 0.23; 95% CI, 0.17-0.31). Results were consistent in patients with cirrhosis, solid cancers, myeloproliferative neoplasms, unprovoked SVT, and SVT associated with transient or persistent nonmalignant risk factors. In patients with SVT, the risk of recurrent VTE and major bleeding is substantial. Anticoagulant treatment is associated with reduced risk of both outcomes. © 2022 by The American Society of Hematology

    Heart failure (HF) is characterized by deterioration of the electrical and contractile function of the heart due to structural and functional remodelling, leading to development of arrhythmias and increased sudden cardiac death risk. SK channels are a type of calcium-activated potassium channels that do not play a relevant role in normal ventricular electrophysiology. However, it has been hypothesized that these channels become more relevant in pathologies such as HF. Nontheless, their role in human ventricular electrophysiology is not fully characterized

    Effects of amphetamine on development of oral candidiasis in rats

    [Abstract] Experiments were conducted to evaluate the effects of amphetamine (0.4 mg/kg of body weight/day) on the development of oral candidiasis in Sprague-Dawley rats. Animals were submitted to surgical hyposalivation in order to facilitate the establishment and persistence of Candida albicans infection. Treatment with drugs (placebo or amphetamine) was initiated 7 days before C. albicans inoculation and lasted until the end of the experiments, day 15 postinoculation. Establishment of C. albicans infection was evaluated by swabbing the inoculated oral cavity with a sterile cotton applicator on days 2 and 15 after inoculation, followed by plating on YEPD (yeast extract-peptone-dextrose) agar. Tissue injury was determined by the quantification of the number and type (normal or abnormal) of papillae on the dorsal tongue per microscopic field. A semiquan- titative scale was devised to assess the degree of colonization of the epithelium by fungal hyphae. Our results show that amphetamine exacerbates C. albicans infection of the tongues of rats. Significant increases in Candida counts, the percentage of the tongue’s surface covered with clinical lesions, the percentage of abnormal papillae, and the colonization of the epithelium by fungal hyphae were found in amphetamine-treated rats compared to those found in the rats injected with a placebo. The last two parameters increased in rats treated with the placebo compared to the parameters of the untreated control rats
