    A Generative Product-of-Filters Model of Audio

    We propose the product-of-filters (PoF) model, a generative model that decomposes audio spectra as sparse linear combinations of "filters" in the log-spectral domain. PoF makes similar assumptions to those used in the classic homomorphic filtering approach to signal processing, but replaces hand-designed decompositions built of basic signal processing operations with a learned decomposition based on statistical inference. This paper formulates the PoF model and derives a mean-field method for posterior inference and a variational EM algorithm to estimate the model's free parameters. We demonstrate PoF's potential for audio processing on a bandwidth expansion task, and show that PoF can serve as an effective unsupervised feature extractor for a speaker identification task.Comment: ICLR 2014 conference-track submission.     A DNA-binding Molecule Targeting the Adaptive Hypoxic Response in Multiple Myeloma has Potent Anti-tumor Activity

    Multiple myeloma is incurable and invariably becomes resistant to chemotherapy. Although the mechanisms remain unclear, hypoxic conditions in the bone marrow have been implicated in contributing to multiple myeloma progression, angiogenesis, and resistance to chemotherapy. These effects occur via adaptive cellular responses mediated by hypoxia-inducible transcription factors (HIF), and targeting HIFs can have anticancer effects in both solid and hematologic malignancies. Here, it was found that in most myeloma cell lines tested, HIF1α, but not HIF2α expression was oxygen dependent, and this could be explained by the differential expression of the regulatory prolyl hydroxylase isoforms. The anti–multiple myeloma effects of a sequence-specific DNA-binding pyrrole-imidazole (Py-Im) polyamide (HIF-PA), which disrupts the HIF heterodimer from binding to its cognate DNA sequences, were also investigated. HIF-PA is cell permeable, localizes to the nuclei, and binds specific regions of DNA with an affinity comparable with that of HIFs. Most of the multiple myeloma cells were resistant to hypoxia-mediated apoptosis, and HIF-PA treatment could overcome this resistance in vitro. Using xenograft models, it was determined that HIF-PA significantly decreased tumor volume and increased hypoxic and apoptotic regions within solid tumor nodules and the growth of myeloma cells engrafted in the bone marrow.     F0-consistent many-to-many non-parallel voice conversion via conditional autoencoder

    Non-parallel many-to-many voice conversion remains an interesting but challenging speech processing task. Many style-transfer-inspired methods such as generative adversarial networks (GANs) and variational autoencoders (VAEs) have been proposed. Recently, AutoVC, a conditional autoencoders (CAEs) based method achieved state-of-the-art results by disentangling the speaker identity and speech content using information-constraining bottlenecks, and it achieves zero-shot conversion by swapping in a different speaker's identity embedding to synthesize a new voice. However, we found that while speaker identity is disentangled from speech content, a significant amount of prosodic information, such as source F0, leaks through the bottleneck, causing target F0 to fluctuate unnaturally. Furthermore, AutoVC has no control of the converted F0 and thus unsuitable for many applications. In the paper, we modified and improved autoencoder-based voice conversion to disentangle content, F0, and speaker identity at the same time. Therefore, we can control the F0 contour, generate speech with F0 consistent with the target speaker, and significantly improve quality and similarity. We support our improvement through quantitative and qualitative analysis

    Preparation and Use of a Yeast shRNA Delivery System for Gene Silencing in Mosquito Larvae

    The mosquito genome projects facilitated research in new facets of mosquito biology, including functional genetic studies in the dengue and Zika virus vector Aedes aegypti and the primary African malaria vector Anopheles gambiae. RNA interference (RNAi) has facilitated gene silencing experiments in both of these disease vector mosquito species and could one day be applied as a new method of vector control. Here, we describe a procedure for the genetic engineering of Saccharomyces cerevisiae (baker’s yeast) that express short hairpin RNA (shRNA) corresponding to mosquito target genes of interest. Following cultivation, which facilitates inexpensive propagation of shRNA, the yeast is inactivated and prepared in a ready-to-use dry tablet formulation that is fed to mosquito larvae. Ingestion of the yeast tablets results in effective larval target gene silencing. This technically straightforward and affordable technique may be applicable to a wide variety of mosquito species and potentially to other arthropods that feed on yeast

    The visual microphone: Passive recovery of sound from video

    When sound hits an object, it causes small vibrations of the object's surface. We show how, using only high-speed video of the object, we can extract those minute vibrations and partially recover the sound that produced them, allowing us to turn everyday objects---a glass of water, a potted plant, a box of tissues, or a bag of chips---into visual microphones. We recover sounds from high-speed footage of a variety of objects with different properties, and use both real and simulated data to examine some of the factors that affect our ability to visually recover sound. We evaluate the quality of recovered sounds using intelligibility and SNR metrics and provide input and recovered audio samples for direct comparison. We also explore how to leverage the rolling shutter in regular consumer cameras to recover audio from standard frame-rate videos, and use the spatial resolution of our method to visualize how sound-related vibrations vary over an object's surface, which we can use to recover the vibration modes of an object.Qatar Computing Research InstituteNational Science Foundation (U.S.) (CGV-1111415)National Science Foundation (U.S.).     Spectroscopic signatures of magnetospheric accretion in Herbig Ae/Be stars. I. The case of HD101412

    Models of magnetically-driven accretion and outflows reproduce many observational properties of T Tauri stars. This concept is not well established for the more massive Herbig Ae/Be stars. We intend to examine the magnetospheric accretion in Herbig Ae/Be stars and search for rotational modulation using spectroscopic signatures, in this first paper concentrating on the well-studied Herbig Ae star HD101412. We used near-infrared spectroscopic observations of the magnetic Herbig Ae star HD101412 to test the magnetospheric character of its accretion disk/star interaction. We reduced and analyzed 30 spectra of HD101412, acquired with the CRIRES and X-shooter spectrographs installed at the VLT (ESO, Chile). The spectroscopic analysis was based on the He I lambda 10,830 and Pa gamma lines, formed in the accretion region. We found that the temporal behavior of these diagnostic lines in the near-infrared spectra of HD101412 can be explained by rotational modulation of line profiles generated by accreting gas with a period P = 20.53+-1.68 d. The discovery of this period, about half of the magnetic rotation period P_m = 42.076 d previously determined from measurements of the mean longitudinal magnetic field, indicates that the accreted matter falls onto the star in regions close to the magnetic poles intersecting the line-of-sight two times during the rotation cycle.
