    Screening of a HUVEC cDNA library with transplant-associated coronary artery disease sera identifies RPL7 as a candidate autoantigen associated with this disease.

    A HUVEC cDNA library was screened with sera from two patients who had developed transplant-associated coronary artery disease (TxCAD) following cardiac transplantation. A total of six positive clones were isolated from a primary screen of 40 000 genes. Subsequent DNA sequence analysis identified these to be lysyl tRNA synthetase, ribosomal protein L7, ribosomal protein L9, beta transducin and TANK. Another gene whose product could not be identified showed homology to a human cDNA clone (DKFZp566M063) derived from fetal kidney. Full-length constructs of selected genes were expressed as his-tag recombinant fusion proteins and used to screen a wider patient base by ELISA to determine prevalence and association with TxCAD.     Interoceptive attention and mood in daily life: an experience-sampling study

    Theories of emotion ascribe a fundamental role to the processing of bodily signals (interoception) in emotional experience. Despite evidence consistent with this, current knowledge is limited by a focus on interoceptive accuracy and laboratory-based interoception measures. This experience-sampling study examines how state interoceptive attention and state emotional experience are related in everyday life, providing the first data to our knowledge examining: (1) within-subject fluctuations in interoceptive attention across domains, and (2) the relationship between trait and state interoception. Compared with rates of exteroceptive attention (auditory attention: engaged 83% of the time), interoceptive signals captured attention approximately 20% of the time, with substantial within- and between-person variability across domains. There were relationships between interoceptive attention and emotion in daily life (greater attention being associated with more negative valence and fatigue) that were specific to interoceptive attention (different patterns were observed with exteroceptive attention). State measures of interoceptive (but not exteroceptive) attention were correlated with the trait interoceptive attention, but not accuracy. Results underscore the relationship between interoceptive attention and emotion, providing new insights into interoceptive attention and the structure of interoceptive ability. Future research should examine the source(s) of within- and between-person variability in interoceptive and exteroceptive attention and its relationship with emotional experience.     Search for domain wall dark matter with atomic clocks on board global positioning system satellites

    Cosmological observations indicate that 85% of all matter in the Universe is dark matter (DM), yet its microscopic composition remains a mystery. One hypothesis is that DM arises from ultralight quantum fields that form macroscopic objects such as topological defects. Here we use GPS as a ~ 50,000 km aperture DM detector to search for such defects in the form of domain walls. GPS navigation relies on precision timing signals furnished by atomic clocks hosted on board GPS satellites. As the Earth moves through the galactic DM halo, interactions with topological defects could cause atomic clock glitches that propagate through the GPS satellite constellation at galactic velocities ~ 300 km/s. Mining 16 years of archival GPS data, we find no evidence for DM in the form of domain walls at our current sensitivity level.     Understanding the contribution of neural and physiological signal variation to the low repeatability of emotion-induced BOLD responses

    Previous studies have reported low repeatability of BOLD activation measures during emotion processing tasks. It is not clear, however, whether low repeatability is a result of changes in the underlying neural signal over time, or due to insufficient reliability of the acquired BOLD signal caused by noise contamination. The aim of this study was to investigate the influence of “cleaning” the BOLD signal, by correcting for physiological noise and for differences in BOLD responsiveness, on measures of repeatability. Fifteen healthy volunteers were scanned on two different occasions, performing an emotion provocation task with faces (neutral, 50% fearful, 100% fearful) followed by a breath-hold paradigm to provide a marker of BOLD responsiveness. Repeatability of signal distribution (spatial repeatability) and repeatability of signal amplitude within two regions of interest (amygdala and fusiform gyrus) were estimated by calculating the intraclass correlation coefficient (ICC). Significant repeatability of signal amplitude was only found within the right amygdala during the perception of 50% fearful faces, but disappeared when physiological noise correction was performed. Spatial repeatability was higher within the fusiform gyrus than within the amygdala, and better at the group level than at the participant level. Neither physiological noise correction, nor consideration of BOLD responsiveness, assessed through the breath-holding, increased repeatability. The findings lead to the conclusion that low repeatability of BOLD response amplitude to emotional faces is more likely to be explained by the lack of stability in the underlying neural signal than by physiological noise contamination. Furthermore, reported repeatability might be a result of repeatability of task-correlated physiological variation rather than neural activity. This means that the emotion paradigm used in this study might not be useful for studies that require the BOLD response to be a stable measure of emotional processing, for example in the context of biomarkers

    Is alexithymia characterised by impaired interoception?:Further evidence, the importance of control variables, and the problems with the Heartbeat Counting Task

    Interoception, the perception of one’s internal state, is commonly quantified using the heartbeat counting task (HCT) – which is thought to be a measure of cardiac interoceptive sensitivity (accuracy). Interoceptive sensitivity has been associated with a number of clinical traits and aspects of higher order cognition, including emotion processing and decision-making. It has been proposed that alexithymia (difficulties identifying and describing one’s own emotions) is associated with impaired interoceptive sensitivity, but new research questions this association. Problematically, much evidence attesting to the absence of this association has been conducted using the HCT, a measure affected by various physiological and psychological factors. Here, we present novel data (N = 287) examining the relationship between alexithymia and HCT performance, controlling for a number of potential confounds. Inclusion of these control measures reveals the predicted negative relationship between alexithymia and HCT performance. Results are discussed with regard to difficulties quantifying interoceptive sensitivity using the HCT

    The relationship between heartbeat counting and heartbeat discrimination:a meta-analysis

    Interoception concerns the perception of the body’s internal state. Despite the importance of this ability for health and aspects of higher-order cognition, its measurement remains problematic. Most studies of interoception employ one of two tasks: the heartbeat counting or heartbeat discrimination task. These tasks are thought to index common abilities, an assertion often used to justify the use of a single measure of cardiac interoception. However, mixed findings regarding the relationship between performance on these tasks raises the question of whether they can be used interchangeably to assess interoceptive accuracy, confidence and awareness (‘metacognition’). The present study employed a meta-analytical approach to assess the association between these tasks. Pooled findings from 22 studies revealed a small relationship between accuracy scores on the measures. Additional analyses demonstrated a moderate relationship between confidence ratings but no association between measures of interoceptive awareness. These findings question the interchangeable use of the two tasks
