Combined immunosuppressive therapy with low dose FK506 and antimetabolites in rat allogeneic heart transplantation
Following rat heterotopic heart allotransplantation, low to lethal doses of the antimetabolites mizoribine (MIZ), RS-61443 (RS), and AZA were given alone or in combination with subtherapeutic doses of FK506 (0.04 mg/kg/day) for 14 days after transplantation. With the median effect analysis of Chou and Kahan for quantitative drug interactions, substantial therapeutic synergism was demonstrated between FK506 and nontoxic doses of MĪZ (2.5, 5, and 10 mg/kg/day) or AZA (5, 30, and 45 mg/kg/day), which was particularly evident with the lowest dose MIZ (2.5 mg/kg/day). When FK506 was used in combination with MIZ or AZA but not with RS, the maximum effect (peak median graft survival) was enhanced significantly from 15 days (MIZ alone) to 26 days (P<0.05), and from 19 days (AZA alone) to 32 days (P<0.0l). In contrast, RS interacted with FK506 no more than additively. Although RS was the most powerful single antimetabolite, the best overall survival was obtained by combining AZA and FK506. The addition of FK506 did not significantly increase the percent mortality and LD50 of the antimetabolites. © 1994 by Williams and Wilkins
Virulence of Escherichia coli Isolates Obtained from Layer Chickens with Colibacillosis Associated with Pericarditis, Perihepatitis, and Salpingitis in Experimentally Infected Chicks and Embryonated Eggs
To evaluate the virulence of avian pathogenic Escherichia coli (APEC) isolates obtained from colibacillosis cases associated with pericarditis, perihepatitis, and salpingitis, the embryo lethality assay and experimental infection model in chicks were used in this study. According to the established criteria based on mortality in the embryo lethality assay for evaluating the virulence of E. coli isolates, 23 of the 26 APEC isolates associated with pericarditis and perihepatitis and 8 of the 20 isolates associated with salpingitis were found to be virulent. Isolate D137, which had been obtained from a case with pericarditis and perihepatitis and had an embryo mortality of 92%, and isolate D445, which had been obtained from a case with pericarditis and perihepatitis and had an embryo mortality of 17%, were used for the experimental infection. Four of the five 11-day-old chickens inoculated through the air sac with isolate D137 died 1 day postinoculation, and the challenge strain was recovered from the air sac, pericardial sac, or liver; however, colibacillosis lesions were found in only one of the five birds postmortem. All five chicks inoculated with isolate D445 survived for 7 days postinoculation and exhibited airsacculitis or pericarditis lesions at 7 days postinoculation; the challenge strain was not recovered from the lesions postmortem. The results obtained in this study suggest that the different APEC isolates tested cause illness in chickens through distinct pathogenesis
Neutrino Background Flux from Sources of Ultrahigh-Energy Cosmic-Ray Nuclei
Motivated by Pierre Auger Observatory results favoring a heavy nuclear
composition for ultrahigh-energy (UHE) cosmic rays, we investigate implications
for the cumulative neutrino background. The requirement that nuclei not be
photodisintegrated constrains their interactions in sources, therefore limiting
neutrino production via photomeson interactions. Assuming a injection spectrum and
photodisintegration via the giant dipole resonance, the background flux of
neutrinos is lower than if UHE nuclei ubiquitously survive in
their sources. This is smaller than the analogous Waxman-Bahcall flux for UHE
protons by about one order of magnitude, and is below the projected IceCube
sensitivity. If IceCube detects a neutrino background, it could be due to other
sources, e.g., hadronuclear interactions of lower-energy cosmic rays; if it
does not, this supports our strong restrictions on the properties of sources of
UHE nuclei.Comment: 7 pages, 3 figure
Intestinal transplantation in children under FK 506 immunosuppression
Intestinal transplantation, solitary (n = 3) or in combination with the liver (n = 7), was performed in 10 pediatric patients with intestinal failure. The liver was only replaced if there was liver failure and portal hypertension. Immunosuppression was based on FK 506. Two patients died, one of graft-versus-host disease and one of lymphoproliferative disease. One patient was still in the intensive care unit 1 month posttransplantation due to perioperative complications. The function of the intestinal grafts in the remaining patients is normal. All nutrition and medications including immunosuppression are being administered enterally. This series indicates that small bowel transplantation, alone or in combination with the liver, is feasible in pediatric patients. © 1993
Phase transition of compartmentalized surface models
Two types of surface models have been investigated by Monte Carlo simulations
on triangulated spheres with compartmentalized domains. Both models are found
to undergo a first-order collapsing transition and a first-order surface
fluctuation transition. The first model is a fluid surface one. The vertices
can freely diffuse only inside the compartments, and they are prohibited from
the free diffusion over the surface due to the domain boundaries. The second is
a skeleton model. The surface shape of the skeleton model is maintained only by
the domain boundaries, which are linear chains with rigid junctions. Therefore,
we can conclude that the first-order transitions occur independent of whether
the shape of surface is mechanically maintained by the skeleton (= the domain
