    Anionic Host Defence Peptides from the Plant Kingdom: Their Anticancer Activity and Mechanisms of Action

    It is becoming increasingly clear that plants, ranging from across the plant kingdom produce anionic host defence peptides (AHDPs) with potent activity against a wide variety of human cancers cells. In general, this activity involves membrane partitioning by AHDPs, which leads to membranolysis and / or internalization to attack intracellular targets such as DNA. Several models have been proposed to describe these events including: the toroidal pore and Shai-Matsuzaki-Huang mechanisms but, in general, the mechanisms underpinning the membrane interactions and anticancer activity of these peptides are poorly understood. Plant AHDPs with anticancer activity can be conveniently discussed with reference to two groups: cyclotides, which possess cyclic molecules stabilized by cysteine knot motifs, and other ADHPs that adopt extended and α-helical conformations. Here, we review research into the anticancer action of these two groups of peptides along with current understanding of the mechanisms underpinning this action

    In Search of Differential Inhibitors of Aldose Reductase

    Aldose reductase, classified within the aldo-keto reductase family as AKR1B1, is an NADPH dependent enzyme that catalyzes the reduction of hydrophilic as well as hydrophobic aldehydes. AKR1B1 is the first enzyme of the so-called polyol pathway that allows the conversion of glucose into sorbitol, which in turn is oxidized to fructose by sorbitol dehydrogenase. The activation of the polyol pathway in hyperglycemic conditions is generally accepted as the event that is responsible for a series of long-term complications of diabetes such as retinopathy, cataract, nephropathy and neuropathy. The role of AKR1B1 in the onset of diabetic complications has made this enzyme the target for the development of molecules capable of inhibiting its activity. Virtually all synthesized compounds have so far failed as drugs for the treatment of diabetic complications. This failure may be partly due to the ability of AKR1B1 to reduce alkenals and alkanals, produced in oxidative stress conditions, thus acting as a detoxifying agent. In recent years we have proposed an alternative approach to the inhibition of AKR1B1, suggesting the possibility of a differential inhibition of the enzyme through molecules able to preferentially inhibit the reduction of either hydrophilic or hydrophobic substrates.     Correlated electron-hole plasma in organometal perovskites

    Organic-inorganic perovskites are a class of solution-processed semiconductors holding promise for the realization of low-cost efficient solar cells and on-chip lasers. Despite the recent attention they have attracted, fundamental aspects of the photophysics underlying device operation still remain elusive. Here we use photoluminescence and transmission spectroscopy to show that photoexcitations give rise to a conducting plasma of unbound but Coulomb-correlated electron-hole pairs at all excitations of interest for light-energy conversion and stimulated optical amplification. The conductive nature of the photoexcited plasma has crucial consequences for perovskite-based devices: in solar cells, it ensures efficient charge separation and ambipolar transport while, concerning lasing, it provides a low threshold for light amplification and justifies a favourable outlook for the demonstration of an electrically driven laser. We find a significant trap density, whose cross-section for carrier capture is however low, yielding a minor impact on device performance

    An Egyptian green schist palette and an amazonite gemstone from the “Palace of the Copper Axes” at Batrawy, Jordan

    The exploration of the “Palace of the Copper Axes”, the Early Bronze Age III palace of the easternmost city of Southern Levant during the 3rd millennium BC, was resumed in 2018, during the 14th season of excavation at Khirbet al-Batrawy in north-central Jordan, and completed in 2019. In the entrance hall of the palace, a square space with ceilings supported by four pillars, an Egyptian green schist palette, a cyan gemstone of amazonite and a pierced bead of fluorapatite were found buried under the destruction layer, while a barrel-shaped carnelian bead from Mesopotamia was found in the destruction layer just outside the Eastern Pavilion of the palace. These finds again testify to the inclusion of the palace and the city into a wide international trade network and its special connections with Pharaonic Egypt

    Photo-electro-chemical properties of TiO2 mediated by the enzyme glucose oxidase

    Electrochemical measurements show that the enzyme Glucose oxidase (GO) is adsorbed on the surface of TiO2 without apparently changing the flat band potential of the semiconductor, indicating that it does not cause a change of the energy of conduction band electrons. On the other hand, it is observed that GO markedly increases the efficiency of the two electron reduction of O2 to H2O2 which is accumulated in the solution phase. ESR spin trapping investigations indicate that GO favors the formation of OH . radicals, due to either the inhibition of charge recombination processes or to H2O2 reduction by conduction band electrons. Accordingly, photo-oxidation of different alcohols to the corresponding radical species is also enhanced in the presence of GO. The photo-oxidation of 1,2-propandiol on TiO2/GO is regioselective in that i) partial oxidation to hydroxyacetone is observed and ii) no mineralization (full combustion to CO2) of the substrate occurs. These facts are of particular interest in the field of studies concerning the design of new photocatalytic systems with enhanced activity and controllable oxidative power

    Mitochondrial DNA Regionalism and Historical Demography in the Extant Populations of Chirocephalus kerkyrensis (Branchiopoda: Anostraca)

    Background: Mediterranean temporary water bodies are important reservoirs of biodiversity and host a unique assemblage of diapausing aquatic invertebrates. These environments are currently vanishing because of increasing human pressure. Chirocephalus kerkyrensis is a fairy shrimp typical of temporary water bodies in Mediterranean plain forests and has undergone a substantial decline in number of populations in recent years due to habitat loss. We assessed patterns of genetic connectivity and phylogeographic history in the seven extant populations of the species from Albania, Corfu Is. (Greece), Southern and Central Italy. Methodology/Principal Findings: We analyzed sequence variation at two mitochondrial DNA genes (Cytochrome Oxidase I and 16s rRNA) in all the known populations of C. kerkyrensis. We used multiple phylogenetic, phylogeographic and coalescence-based approaches to assess connectivity and historical demography across the whole distribution range of the species. C. kerkyrensis is genetically subdivided into three main mitochondrial lineages; two of them are geographically localized (Corfu Is. and Central Italy) and one encompasses a wide geographic area (Albania and Southern Italy). Most of the detected genetic variation (<81%) is apportioned among the aforementioned lineages. Conclusions/Significance: Multiple analyses of mismatch distributions consistently supported both past demographic and spatial expansions with the former predating the latter; demographic expansions were consistently placed during interglacial warm phases of the Pleistocene while spatial expansions were restricted to cold periods. Coalescence methods revealed a scenario of past isolation with low levels of gene flow in line with what is already known for other co-distributed fairy shrimps and suggest drift as the prevailing force in promoting local divergence. We recommend that these evolutionary trajectories should be taken in proper consideration in any effort aimed at protecting Mediterranean temporary water bodies

    Analyzing children's expectations from robotic companions in educational settings

    The use of robots as educational partners has been extensively explored, but less is known about the required characteristics these robots should have to meet children's expectations. Thus the purpose of this study is to analyze children's assumptions regarding morphology, functionality, and body features, among others, that robots should have to interact with them. To do so, we analyzed 142 drawings from 9 to 10 years old children and their answers to a survey provided after interacting with different robotic platforms.
