On the Buckling of Elastic Rings by External Confinement
We report the results of an experimental and numerical investigation into the buckling of thin elastic rings confined within containers of circular or regular polygonal cross section. The rings float on the surface of water held in the container and controlled removal of the fluid increases the confinement of the ring. The increased compressive forces can cause the ring to buckle into a variety of shapes. For the circular container, finite perturbations are required to induce buckling, whereas in polygonal containers the buckling occurs through a linear instability that is closely related to the canonical Euler column buckling. A model based on Kirchhoff–Love beam theory is developed and solved numerically, showing good agreement with the experiments and revealing that in polygons increasing the number of sides means that buckling occurs at reduced levels of confinement. Intestinal barrier tightening by a cell-penetrating antibody to Bin1, a candidate target for immunotherapy of ulcerative colitis.
Patients afflicted with ulcerative colitis (UC) are at increased risk of colorectal cancer. While its causes are not fully understood, UC is associated with defects in colonic epithelial barriers that sustain inflammation of the colon mucosa caused by recruitment of lymphocytes and neutrophils into the lamina propria. Based on genetic evidence that attenuation of the bridging integrator 1 (Bin1) gene can limit UC pathogenicity in animals, we have explored Bin1 targeting as a therapeutic option. Early feasibility studies in the dextran sodium sulfate mouse model of experimental colitis showed that administration of a cell-penetrating Bin1 monoclonal antibody (Bin1 mAb 99D) could prevent lesion formation in the colon mucosa in part by preventing rupture of lymphoid follicles. Reliability of functional outcome measures in adults with neurofibromatosis 1
Objectives: To determine intra-rater and inter-rater reliability of functional outcome measures in adults with neurofibromatosis 1 (NF1) and to ascertain how closely objective and subjective measures align.
Methods: Forty-nine ambulant adults with NF1 aged 16 years and over were included in this observational study. Median age 31 years (range 16-66), 29 females, 20 males. Participants were video-recorded or photographed performing four functional outcome measures. Four raters from the Neurofibromatosis centre multi-disciplinary team independently scored the measures to determine inter-rater reliability. One rater scored the measures a second time on a separate occasion to determine intra-rater reliability. The measures evaluated were the functional reach, timed up and go, ten metre walk and a modified nine-hole peg tests. Participants also completed a disease specific quality of life questionnaire (INF1-QOL).
Results: Inter-rater reliability and intra-rater reliability scores (intra-class coefficient, ICC) were similar for each outcome measure. Excellent rater agreement (ICC r ≥ 0.9) was found for the functional reach, timed up and go and the 10 metre walk tests. Rater agreement was good for the modified 9-hole peg test; ICC r= 0.75 for intra-rater reliability and 0.76 for inter-rater reliability. The timed up and go and the 10 metre walk tests correlated highly with perceived mobility challenges in the quality of life questionnaire (INF1-QOL).
Conclusions: The functional reach, timed up and go and 10 metre walk tests are potentially useful outcome measures for monitoring NF1 treatment and will be assessed for validity and reliability in future multi-centre studies
An 18S rDNA Perspective on the Classification of Criconematoidea
In the nematode family Criconematidae, a taxonomy primarily based on cuticle characters has created classifications that are notoriously volatile. Molecular characters may lead to their stabilization. A phylogenetic tree of Criconematoidea was constructed using 166 new near full-length 18S rDNA sequences and 58 sequences from GenBank. Bayesian and maximum likelihood (ML) analyses produced trees with similar topologies. Major features include a strongly supported clade that includes Criconematidae and Hemicycliophoridae, excluding Paratylenchidae and Tylenchulidae. Another well-supported clade groups Criconema, Ogma, Crossonema, and Hemicriconemoides plus Xenocriconemella, combining nematodes with cuticular scales with those without scales at any life stage. Mesocriconema, Discocriconemella limitanea, Hemicaloosia, and Lobocriconema are recognized as monophyletic groups, but Criconemoides is paraphyletic. Both trees support an unexpected sister relationship between Bakernema and Hemicycliophora. The 18S rDNA dataset was insufficient for distinguishing genus boundaries between Criconema, Ogma, and Crossonema. The relationships depicted by the 18S rDNA phylogeny suggest that key morphological characters used in the classification of Criconematidae are not homologous
A global database of soil nematode abundance and functional group composition
As the most abundant animals on earth, nematodes are a dominant component of the soil community. They play critical roles in regulating biogeochemical cycles and vegetation dynamics within and across landscapes and are an indicator of soil biological activity. Here, we present a comprehensive global dataset of soil nematode abundance and functional group composition. This dataset includes 6,825 georeferenced soil samples from all continents and biomes. For geospatial mapping purposes these samples are aggregated into 1,933 unique 1-km pixels, each of which is linked to 73 global environmental covariate data layers. Altogether, this dataset can help to gain insight into the spatial distribution patterns of soil nematode abundance and community composition, and the environmental drivers shaping these patterns
Statistics of velocity gradients in wall-bounded shear flow turbulence
The statistical properties of velocity gradients in a wall-bounded turbulent
channel flow are discussed on the basis of three-dimensional direct numerical
simulations. Our analysis is concentrated on the trend of the statistical
properties of the local enstrophy and the energy
dissipation rate with increasing distance from the wall.
We detect a sensitive dependence of the largest amplitudes of both fields
(which correspond with the tail of the distribution) on the spectral
resolution. The probability density functions of each single field as well as
their joint distribution vary significantly with increasing distance from the
wall. The largest fluctuations of the velocity gradients are found in the
logarithmic layer. This is in agreement with recent experiments which observe a
bursting of hairpin vortex packets into the logarithmic region.Comment: 13 pages, 5 figure
