    An Investigation of the Effects of Public Policy on Spatial Concentration and Company Linkages in the Martitime Sector

    Industry clusters (Porter, 1990, 1998b) have been the focus of numerous studies, and public policy programmes in recent decades (Ketels, 2013b). The maritime sector in particular has seen a number of maritime cluster organisations established in recent years as policy tools for regional development. Cluster analysis has focused on identifying and mapping clusters, yet comparatively little research has been undertaken on the optimal structure for cluster organisations (or initiatives) as additionality policy tools, to achieve the aim of strengthening cluster linkages and boosting the innovation capacity of cluster firms. Therefore this research addresses the following questions: 1. To what extent is there a functioning maritime cluster in the Cork region, what model best describes its structure, and its development to date? 2. How effective is the IMERC cluster organisation in its role of bridging `innovation gaps\u27 and facilitating linkages between the cluster actors? A nested methodology incorporating an analysis of cluster firms\u27 linkages utilising the V-LINC methodology; participatory evaluation conducted in the form of a Delphi study; and case studies; is used to analyse the effect the structure of a cluster organisation has on linkages within clusters. The analysis concludes that there is a functioning nascent maritime cluster in the Cork region. It also find that the IMERC cluster organisation is playing an effective role in bridging `innovation gaps\u27 between cluster actors.     The role of mitochondrial functionality in brewing yeast

    Within yeast cell cultures typically 1-3% of the total cells contain damaged mitochondrial DNA (mtDNA). Damaged mtDNA manifests in the production of respiratory deficient ‘petite’ yeast cells. In certain circumstances such as in response to stress-inducing conditions associated with the modern brewing process the number of petites can exceed 10-20% of the total cell count. In order to provide insight into the formation of petites, this study aimed to analyse the mitochondria of several brewing yeast. The mitochondrial genotype was examined through sequencing and mtDNA fingerprints while the mitochondrial phenotype was examined using high resolution microscopy and flow cytometry. Interestingly, mtDNA fingerprinting revealed variation across S. cerevisiae strains, while conversely revealing little variation across S. pastorianus strains. Microscopy revealed some insight into the morphological variations of mitochondria, but flow cytometry yielded the greater insight into mitochondrial quantity and functionality. Additionally, changes in mitochondrial content throughout propagation, fermentation and storage were examined. With an understanding of the mitochondrial phenotype and genotype of respiratory competent ‘grande’, a comparative examination of the petite phenotype was undertaken. Genotypically and phenotypically the petites were shown to greatly differ to their grande counterparts, with less diverse mtDNA fingerprints and stunted mitochondrial morphology.     Click Pt(IV)-Carbohydrates Pro-Drugs for Treatment of Osteosarcoma

    The selectivity vs. cancer cells has always been a major challenge for chemotherapeutic agents and in particular for cisplatin, one of the most important anticancer drugs for the treatment of several types of tumors. One strategy to overtake this challenge is to modify the coordination sphere of the metallic center with specific vectors whose receptors are overexpressed in the tumoral cell membrane, such as monosaccharides. In this paper, we report the synthesis of four novel glyco-modified Pt(IV) pro-drugs, based on cisplatin scaffold, and their biological activity against osteosarcoma (OS), a malignant tumor affecting in particular adolescents and young adults.     Multiple unbiased approaches identify oxidosqualene cyclase as the molecular target of a promising anti-leishmanial

    Phenotypic screening identified a benzothiophene compound with activity against Leishmania donovani, the causative agent of visceral leishmaniasis. Using multiple orthogonal approaches, oxidosqualene cyclase (OSC), a key enzyme of sterol biosynthesis, was identified as the target of this racemic compound and its enantiomers. Whole genome sequencing and screening of a genome-wide overexpression library confirmed that OSC gene amplification is associated with resistance to compound 1. Introduction of an ectopic copy of the OSC gene into wild-type cells reduced susceptibility to these compounds confirming the role of this enzyme in resistance. Biochemical analyses demonstrated the accumulation of the substrate of OSC and depletion of its product in compound (S)-1-treated-promastigotes and cell-free membrane preparations, respectively. Thermal proteome profiling confirmed that compound (S)-1 binds directly to OSC. Finally, modeling and docking studies identified key interactions between compound (S)-1 and the LdOSC active site.     2D and 3D anticancer properties of C2-functionalised glucosamine-Pt (IV) prodrugs based on cisplatin scaffold

    A series of C2-functionalied Pt (IV) glycoconjugates based on glucosamine have been synthesised, characterised and tested as anticancer agents on a series of different 2D and 3D cancer cell lines. The carbohydrate will act as a targeted delivery system to improve the selectivity, exploiting the Warburg Effect and the GLUTs receptors that are overexpressed in most of the cancer cells. The hydroxyl at C2 of the carbohydrates does not participate in hydrogen bonding with the GLUTs receptors, making C2 an attractive position for drug conjugation as seen in literature. In this study, we use the amino functionality at the C2 position in glucosamine and Copper-catalysed Azide-Alkyne Cycloaddition “click” (CuAAC) reaction to connect the prodrug Pt (IV) scaffold to the carbohydrate. We have investigated complexes with different linker lengths, as well as acetyl protected and free derivatives.     Toolkit of Approaches To Support Target-Focused Drug Discovery for Plasmodium falciparum Lysyl tRNA Synthetase

    There is a pressing need for new medicines to prevent and treat malaria. Most antimalarial drug discovery is reliant upon phenotypic screening. However, with the development of improved target validation strategies, target-focused approaches are now being utilized. Here, we describe the development of a toolkit to support the therapeutic exploitation of a promising target, lysyl tRNA synthetase (PfKRS). The toolkit includes resistant mutants to probe resistance mechanisms and on-target engagement for specific chemotypes; a hybrid KRS protein capable of producing crystals suitable for ligand soaking, thus providing high-resolution structural information to guide compound optimization; chemical probes to facilitate pulldown studies aimed at revealing the full range of specifically interacting proteins and thermal proteome profiling (TPP); as well as streamlined isothermal TPP methods to provide unbiased confirmation of on-target engagement within a biologically relevant milieu. This combination of tools and methodologies acts as a template for the development of future target-enabling packages

    Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage.     Effects of antiplatelet therapy after stroke due to intracerebral haemorrhage (RESTART): a randomised, open-label trial

    Background: Antiplatelet therapy reduces the risk of major vascular events for people with occlusive vascular disease, although it might increase the risk of intracranial haemorrhage. Patients surviving the commonest subtype of intracranial haemorrhage, intracerebral haemorrhage, are at risk of both haemorrhagic and occlusive vascular events, but whether antiplatelet therapy can be used safely is unclear. We aimed to estimate the relative and absolute effects of antiplatelet therapy on recurrent intracerebral haemorrhage and whether this risk might exceed any reduction of occlusive vascular events. Methods: The REstart or STop Antithrombotics Randomised Trial (RESTART) was a prospective, randomised, open-label, blinded endpoint, parallel-group trial at 122 hospitals in the UK. We recruited adults (≥18 years) who were taking antithrombotic (antiplatelet or anticoagulant) therapy for the prevention of occlusive vascular disease when they developed intracerebral haemorrhage, discontinued antithrombotic therapy, and survived for 24 h. Computerised randomisation incorporating minimisation allocated participants (1:1) to start or avoid antiplatelet therapy. We followed participants for the primary outcome (recurrent symptomatic intracerebral haemorrhage) for up to 5 years. We analysed data from all randomised participants using Cox proportional hazards regression, adjusted for minimisation covariates. This trial is registered with ISRCTN (number ISRCTN71907627). Findings: Between May 22, 2013, and May 31, 2018, 537 participants were recruited a median of 76 days (IQR 29–146) after intracerebral haemorrhage onset: 268 were assigned to start and 269 (one withdrew) to avoid antiplatelet therapy. Participants were followed for a median of 2·0 years (IQR [1·0– 3·0]; completeness 99·3%). 12 (4%) of 268 participants allocated to antiplatelet therapy had recurrence of intracerebral haemorrhage compared with 23 (9%) of 268 participants allocated to avoid antiplatelet therapy (adjusted hazard ratio 0·51 [95% CI 0·25–1·03]; p=0·060). 18 (7%) participants allocated to antiplatelet therapy experienced major haemorrhagic events compared with 25 (9%) participants allocated to avoid antiplatelet therapy (0·71 [0·39–1·30]; p=0·27), and 39 [15%] participants allocated to antiplatelet therapy had major occlusive vascular events compared with 38 [14%] allocated to avoid antiplatelet therapy (1·02 [0·65–1·60]; p=0·92). Interpretation: These results exclude all but a very modest increase in the risk of recurrent intracerebral haemorrhage with antiplatelet therapy for patients on antithrombotic therapy for the prevention of occlusive vascular disease when they developed intracerebral haemorrhage. The risk of recurrent intracerebral haemorrhage is probably too small to exceed the established benefits of antiplatelet therapy for secondary prevention
