    Granulosa Cell Conditioned Medium Enhances The Rate of Mouse Oocyte In Vitro Maturation and Embryo Formation

    Objective: In vitro maturation (IVM) and cryopreservation of oocytes are two important parts of assisted reproductive technology (ART), but their efficacy is low. This study aimed to improve the quality of in vitro vitrified-warmed maturated oocytes using granulosa cell conditioned medium (GCCM). Materials and Methods: In the experimental study, fresh/non-vitrified and vitrified-warmed mouse germinal vesicle (GV) oocytes (as F and V) were in vitro maturated using basal medium (BM) and also BM supplemented with 50% GCCM as treated groups (GM), and categorized as FBM, FGM, VBM and VGM groups, respectively. The rate of successful IVM (MII oocyte formation), mitochondrial membrane potential and the viability of MII oocytes were determined using inverted microscopy, JC-1 and trypan blue staining. Then, the rate of in vitro fertilization (IVF) and subsequent two-cell embryo formation was calculated. Finally, the expression levels of Oct4, Sox2, Cdk-2, Gdf9, Integrin beta1 and Igf2 were analyzed using real-time polymerase chain reaction (PCR) in MII oocytes and two-cell embryos. Results: These analyses showed that GCCM significantly increased the IVM rate, oocyte meiotic resumption and mitochondrial membrane potential (P<0.05). In addition, the rate of IVF and two-cell embryo formation was significantly higher in FGM and VGM compared to FBM and VBM (P<0.05). Interestingly, GCCM significantly affected the expression of the studied genes. Conclusion: Our findings suggest that GCCM might be useful for improving the efficiency of IVM and the subsequent IVF outcomes

    Fermi Surface and Quasiparticle Excitations of overdoped Tl2Ba2CuO6+d by ARPES

    The electronic structure of the high-T_c superconductor Tl2Ba2CuO6+d is studied by ARPES. For a very overdoped Tc=30K sample, the Fermi surface consists of a single large hole pocket centered at (pi,pi) and is approaching a topological transition. Although a superconducting gap with d_x^2-y^2 symmetry is tentatively identified, the quasiparticle evolution with momentum and binding energy exhibits a marked departure from the behavior observed in under and optimally doped cuprates. The relevance of these findings to scattering, many-body, and quantum-critical phenomena is discussed.Comment: Revised manuscript, in press on PRL.     A Computer Vision-Based Approach for Non-contact Modal Analysis and Finite Element Model Updating

    Computer vision-based techniques for modal analysis and system identification are rapidly becoming of great interest for both academic research and engineering practice in structural engineering. For instance, this is particularly relevant in fields such as bridge or tall building monitoring, where the large size of the structure would require an expensive sensor network, and for the characterisation of very slender, highly-flexible structural components, where physically-attached sensors cannot be deployed without altering the mass and stiffness of the system under investigation. This study concerns the latter case. Here, an algorithm for the full-field, non-contact extraction and processing of useful information from vibrational data is applied. Firstly, video acquisition is used to capture rapidly very spatially- and temporally-dense information regarding the vibrational behaviour of a high-aspect-ratio (HAR) prototype wing, with high image quality and high frame rate. Video processing is then applied to extract displacement time histories from the collected data; in turn, these are used to perform Modal Analysis (MA) and Finite Element Model Updating (FEMU). Results are benchmarked against the ones obtained from a single-point laser Doppler vibrometer (LDV). The study is performed on the beam-like spar of the wing prototype with and without the sensors attached to appreciate the disruptive effects of sensor loading. Promising results were achieved

    Strongly aligned coastal boulders on Ko Larn island (Thailand): a proxy for past typhoon-driven high-energy wave events in the Bay of Bangkok

    © 2019 Institute of Australian Geographers Recent studies of elevated carbonate boulder deposits on several rock islands near Bangkok have indicated that Thailand\u27s capital city may not be as protected from typhoon strikes as previously thought. Here, new evidence is presented for past high-energy wave (HEW) events in the form of statistically significant patterns of boulder alignment on exposed rocky shorelines of Ko Larn island. The long-axis orientations of 193 coastal sandstone boulders were analysed across four study sites. Several scenarios for the unimodal, bimodal, and polymodal patterns found can be envisaged. Either the most recent HEW event was the strongest—in which case most clasts were rearranged unimodally (one observation site), or the strongest HEW event was earlier and subsequent weaker ones realigned only smaller boulders to produce bimodal or polymodal patterns (three observation sites). Inferred northeastward or eastward onshore flow directions are consistent with palaeo-typhoons penetrating into the Bay of Bangkok on northwestward curving tracks. The calculated minimum flow velocities required to transport all sampled boulders are 5.5–7.8 m s−1, similar to other findings throughout the Asia-Pacific region. It was observed that the absence of a fitted boulder geomorphology lends credence to the earlier proposed time frame of 150–200 years between typhoon phases in the upper Gulf of Thailand. The current work has provided additional insights into the characteristics of past HEW events that have a possibility of reoccurring again at some time in the future. Our findings continue to raise awareness for a reassessment of the risks of coastal hazards for the Chao Phraya River delta and densely populated Bangkok, for which storm surge modelling should be an urgent priority, so as to give better perceptions on how typhoon-driven marine incursion would impact the city

    Full-field digital image correlation with Kriging regression

    A full-field Digital Image Correlation (DIC) method with integrated Kriging regression is presented in this article. The displacement field is formulated as a best linear unbiased model that includes the correlations between all the locations in the Region of Interest (RoI). A global error factor is employed to extend conventional Kriging interpolation to quantify displacement errors of the control points. An updating strategy for the self-adaptive control grid is developed on the basis of the Mean Squared Error (MSE) determined from the Kriging model. Kriging DIC is shown to outperform several other full-field DIC methods when using open-access experimental data. Numerical examples are used to demonstrate the robustness of Kriging DIC to different choices of initial control points and to speckle pattern variability. Finally Kriging DIC is tested on an experimental example

    Molecular basis of oocyte-paracrine signalling that promotes granulosa cell proliferation

    Copyright © 2006 Company of BiologistsOocytes regulate follicle growth by secreting paracrine growth factors that act on neighbouring granulosa cells (GCs). Those factors identified to date are mainly members of the transforming growth factor-ß (TGFß) superfamily, but little is known about which specific receptor/signalling system(s) they employ. This study was conducted to determine the requisite pathways utilised by oocytes to promote GC proliferation. We used an established oocyte-secreted mitogen bioassay, where denuded mouse oocytes are co-cultured with mural GCs. Oocytes, growth differentiation factor-9 (GDF9), TGFß1 and activin-A all promoted GC DNA synthesis, but bone-morphogenetic protein 6 (BMP6) did not. Subsequently, we tested the capacity of various TGFß superfamily receptor ectodomains (ECD) to neutralise oocyte- or specific growth factor-stimulated GC proliferation. The BMP type-II receptor (BMPR-II) ECD antagonised oocyte and GDF9 bioactivity dose-dependently, but had no or minimal effect on TGFß1 and activin-A bioactivity, demonstrating its specificity. The TGFßR-II, activinR-IIA and activinR-IIB ECDs all failed to neutralise oocyte- or GDF9-stimulated GC DNA synthesis, whereas they did antagonise the activity of their respective native ligands. An activin receptor-like kinase (ALK) 4/5/7 inhibitor, SB431542, also antagonised both oocyte and GDF9 bioactivity in a dose-dependent manner. Consistent with these findings, oocytes, GDF9 and TGFß1 all activated SMAD2/3 reporter constructs in transfected GC, and led to phosphorylation of SMAD2 proteins in treated cells. Surprisingly, oocytes did not activate the SMAD1/5/8 pathway in transfected GCs although exogenous BMP6 did. This study indicates that oocyte paracrine factors primarily utilise a similar signalling pathway first identified for GDF9 that employs an unusual combination of TGFß superfamily receptors, the BMPR-II and a SMAD2/3 stimulatory ALK (4, 5 or 7), for transmitting their mitogenic actions in GC. This cell-signalling pathway may also have relevance in the hypothalamic-pituitary axis and in germ-somatic cell interactions in the testis.Robert B. Gilchrist, Lesley J.     Investigating Online Fan Responses to the Rooney Rule in English Football

    Purpose This work critically investigates online fan responses towards the implementation of the affirmative action policy, the Rooney Rule, within English professional football. It explores systemic and structural racism and the history of the Rooney Rule, before analysing football fans' Twitter comments concerning the policy within English football across an 18-month period. Design/methodology/approach This research utilised a bespoke search programme to identify and analyse Tweets which focused on the Rooney Rule in English football. A total of 205 posts were thematically analysed and a series of codes were created. Findings The findings illustrated that fans were generally divided over the Rooney Rule. Over half of the participants welcomed counter measures against structural racism although many caveated responses by critiquing the Rule's approach and scope. For others, however, the policy is yet another example of ‘reverse racism’ and ‘political correctness gone mad’. The findings illustrate that there is an undercurrent of hostility towards anti-racist action and a belief that sport is inherently meritocratic and fair. Originality/value While much research has focused on examining online reactions to ‘trigger events’, this chapter provides an empirical insight into contemporary football fan responses towards anti-racist action in the ‘beautiful game’. It demonstrates that there are a series of common misconceptions and misunderstandings towards affirmative action policies in sport. Once we become aware of such misunderstandings, we can attempt to remedy them in order to aid the efficacy of anti-racist action

    The minimum norm multi-input multi-output receptance method for partial pole placement

    A closed-form analytical solution is developed for the first time that fully addresses the problem of choosing feedback gains that minimize the control effort required for partial pole placement in multi-input, multi-output systems. The norm of the feedback gain matrix is shown to take the form of an inverse Rayleigh quotient, such that the optimal closed-loop system eigenvectors are given as a function of the dominant (highest)eigenvectors of the matrix in the quotient. The feedback gains that deliver the required pole placement with minimum effort may then be determined using standard procedures. The original formulation by the receptance method proposed an arbitrary choice of the closed loop eigenvectors that assigned the poles exactly but was generally wasteful of control effort that might otherwise be conserved or put to good use in satisfying additional control objectives. The analytical solution is validated against a set of numerical examples
