    Role of the JP45-Calsequestrin Complex on Calcium Entry in Slow Twitch Skeletal Muscles

    We exploited a variety of mouse models to assess the roles of JP45-CASQ1 (CASQ, calsequestrin) and JP45-CASQ2 on calcium entry in slow twitch muscles. In flexor digitorum brevis (FDB) fibers isolated from JP45-CASQ1-CASQ2 triple KO mice, calcium transients induced by tetanic stimulation rely on calcium entry via La3+- and nifedipine-sensitive calcium channels. The comparison of excitation-coupled calcium entry (ECCE) between FDB fibers from WT, JP45KO, CASQ1KO, CASQ2KO, JP45-CASQ1 double KO, JP45-CASQ2 double KO, and JP45-CASQ1-CASQ2 triple KO shows that ECCE enhancement requires ablation of both CASQs and JP45. Calcium entry activated by ablation of both JP45-CASQ1 and JP45-CASQ2 complexes supports tetanic force development in slow twitch soleus muscles. In addition, we show that CASQs interact with JP45 at Ca2+ concentrations similar to those present in the lumen of the sarcoplasmic reticulum at rest, whereas Ca2+ concentrations similar to those present in the SR lumen after depolarization-induced calcium release cause the dissociation of JP45 from CASQs. Our results show that the complex JP45-CASQs is a negative regulator of ECCE and that tetanic force development in slow twitch muscles is supported by the dynamic interaction between JP45 and CASQs

    Simultaneous sampling of vapor and particle-phase carcinogenic polycyclic aromatic hydrocarbons on functionalized glass fiber filters

    The sampling of polycyclic aromatic hydrocarbons (PAHs) in the atmosphere is often performed on filters, which retain only aerosol particles, disregarding the vapor fraction; after the filter, an adsorbent (e.g., polyurethane foam, PUF, or styrene/divinylbenzene, XAD) is sometimes used for sampling vapors not retained from the filter. However, the use of an adsorbent may lead to many disadvantages: contaminations, analysis time and costs, and size problems when developing automated or personal samplers. In this work, a functionalized glass fiber filter for the simultaneous sampling of aerosol particles and vapor fraction is presented for the sampling of PAHs in air. A low sampling efficiency was observed for 3 ring PAHs, but all carcinogenic PAHs (according to IARC) were totally retained on functionalized filters. On the other hand, a comparison with normal filter sampling was performed, and results obtained confirm that > 10% of benzo(a)pyrene can be lost from normal filters. Together with size reduction, another advantage of the functionalized filter is an enhancement in the extraction and purification recovery. © Taiwan Association for Aerosol Research

    ABSTRACT The aim was to compare the effect of dimethylacetamide (DMA) and N-methylacetamide (NMA) concentrations on the quality and fertility of post-thaw chicken semen. Ejaculates were obtained from 30 Hi-Line White roosters and processed according to the following treatments: lake pre-freezing extender + 0.1M trehalose (LPF-T) + 6% DMA (control treatment), LPF-T + 9% DMA, LPF-T + 6% NMA, and LPF-T + 9% NMA. Sperm quality (viability, motility, and kinetic traits) was assessed before and after cryopreservation. A total of 15 laying hens per treatment were inseminated to assess fertility and embryo viability. Sperm cryopreserved in presence of DMA had significantly better in vitro quality compared to NMA, showing the highest proportion of viable and progressive motile sperm recovered after thawing. Furthermore, proportion of progressive motile sperm and the VCL, LIN, ALH, and WOB mean values were significantly improved in semen samples frozen/thawed with 6% compared to 9% cryoprotectant concentration. However, the best cryoprotective action on sperm quality played by DMA and the lowest cryoprotectant concentration did not translate into a concomitant advantage in in vivo semen fertility that showed no differences between cryoprotectant and cryoprotectant concentration treatments. Finally, the cryoprotectant DMA and NMA showed an opposite effect on embryo viability in comparison with the effect played on in vitro semen quality, being NMA more efficient than DMA on preserving viable embryos. The present results suggest the urgency to further decrease the cryoprotectant concentration in poultry semen freezing procedures and to assess the specific toxic effect of cryoprotectant on sperm integrity, fertility, and embryo development

    The immediate impacts of COVID-19 on European electricity systems: a first assessment and lessons learned

    The worldwide spread of the COVID-19 pandemic in 2020 forced most countries to intervene with policies and actions—including lockdowns, social-distancing and smart working measures—aimed at mitigating the health system and socio-economic disruption risks. The electricity sector was impacted as well, with performance largely reflecting the changes in the industrial and commercial sectors operations and in the social behavior patterns. The most immediate consequences concerned the power demand profiles, the generation mix composition and the electricity price trends. As a matter of fact, the electricity sectors experienced a foretaste of the future, with higher renewable energy penetration and concerns for security of supply. This paper presents a systemic approach toward assessing the impacts of the COVID-19 pandemic on the power sector. This is aimed at supporting decision making—particularly for policy makers, regulators, and system operators—by quantifying shorter term effects and identifying longer term impacts of the pandemic waves on the power system. Various metrics are defined in different areas—system operation, security, and electricity markets—to quantify those impacts. The methodology is finally applied to the European power system to produce a comparative assessment of the effects of the lockdown in the European context

    Inhibition of Poly(ADP-ribose)polymerase impairs Epstein Barr Virus lytic cycle progression

    <p>Abstract</p> <p>Background</p> <p>Poly(ADP-ribosylation) is a post-translational modification of nuclear proteins involved in several cellular events as well as in processes that characterize the infective cycle of some viruses. In the present study, we investigated the role of poly(ADP-ribosylation) on Epstein-Barr Virus (EBV) lytic cycle activation.</p> <p>Results</p> <p>Inhibition of PARP-1 by 3-aminobenzamide (3-ABA) during EBV induction, diminished cell damage and apoptosis in the non-productive Raji cell line while markedly reducing the release of viral particles in the productive Jijoye cells. Furthermore, incubation with 3-ABA up-regulated the levels of LMP1 and EBNA2 latent viral proteins. At the same time, it slightly affected the expression of the immediate early BZLF1 gene, but largely down-regulated the levels of the early BFRF1 protein. The modulation of the expression of both latent and lytic EBV genes appeared to be post-transcriptionally regulated.</p> <p>Conclusion</p> <p>Taken together the data indicate that PARP-1 plays a role in the progression of EBV lytic cycle and therefore, PARP inhibitors might represent suitable pharmacological adjuncts to control viral spread in EBV productive infection.</p

    Free-range rearing density for male and female Milanino chickens: carcass yield and qualitative meat traits

    SUMMARY The Milanino is a heavy Italian chicken breed and represents an important genetic resource for alternative production systems. In order to support its promotion in the market according to consumer expectations on healthy nutrition and animal welfare, this trial aims to study the slaughter performance and the meat quality in male and female Milanino chickens kept at different rearing density in a separate-sex free-range system. A total of 140 birds (70M:70F) were reared in outdoor pens from 75 to 235 d of life according to the following experimental groups (35 birds/group): M2) males in 2 m2/bird; F2) females in 2 m2/bird; M10) males in 10 m2/bird; F10) females in 10 m2/bird. At 235 d of age, 6 birds per group were slaughtered. Slaughter performance and meat quality were assessed. The Milanino chicken is characterized by high carcass weight and carcass yield among local chicken breeds, and a rearing density of 10 m2/bird is recommended for male birds to obtain heavier carcasses. Milanino meat appears bright and intensely colored, and it is characterized by high protein and low fat contents compared with the standard broiler meat. Total lipids of Milanino meat are characterized by a healthy fatty acid composition, corresponding to a high PUFA/SFA ratio. Milanino breast meat is a good supply of nutraceutical PUFA with a positive low n-6/n-3 PUFA ratio. The ability of the Milanino breed either to synthesize or to transfer to tissue a high quantity of PUFA relevant for human health could be a key factor for its economic valorization

    Achieving energy sustainability in future neighborhoods through building refurbishment and energy hub concept: a case study in Hemberg-Switzerland

    This study aims to investigate the role of distributed generation through the energy hub concept and refurbishment of existing buildings. More specifically, a computational platform combining (i) the urban energy modelling tool CitySimPro and (ii) the microgrid simulation tool Homer is developed. The energy flow on hourly basis is assessed for buildings in Hemberg, a small village in Switzerland, considering occupancy, lighting and appliances profiles, as defined by the Swiss normative. System design of the energy hub is optimized considering three energy system configurations: (i) present scenario, (ii) energy hub catering electrical demand, (iii) energy hub catering both electrical and thermal demand including heat pumps. Results for the integration of an energy hub on site show that the current use of renewables can be increased from 0.15% to over 60% by integrating heat pumps in the city energy network
