Computed Tomography Evaluation of Normal Canine Abdominal Lymph Nodes: Retrospective Study of Size and Morphology According to Body Weight and Age in 45 Dogs
The morphological characteristics of the largest lymphatic vessels and lymph nodes of the body have been described through ultrasonography, although food and gas in the gastrointestinal tract can often have negative effects on the response of small abdominal structures. The aim of the study was to describe the size of normal abdominal lymph nodes (ALs) in dogs affected by disease, not including lymphadenomegaly or lymphadenopathy, and divided according to body weight and age. The ALs studied included the jejunal, medial iliac, portal, gastric, splenic, and pancreaticoduodenal lymph nodes. Statistical correlation considering body weight and age as continuous variables showed that all measurements of the ALs increased according to body weight changes (p p p < 0.05). Other characteristics (shape, attenuation, and enhancement) are subsequently reported. The resulting data can be used to categorize CT measurements of normal ALs displayed based on the body weight and age of the subjects. Non-exhaustive DNA methylation-mediated transposon silencing in the black truffle genome, a complex fungal genome with massive repeat element content
Background: We investigated how an extremely transposon element (TE)-rich organism such as the plant-symbiotic ascomycete truffle Tuber melanosporum exploits DNA methylation to cope with the more than 45,000 repeated elements that populate its genome. Results: Whole-genome bisulfite sequencing performed on different developmental stages reveals a high fraction of methylated cytosines with a strong preference for CpG sites. The methylation pattern is highly similar among samples and selectively targets TEs rather than genes. A marked trend toward hypomethylation is observed for TEs located within a 1 kb distance from expressed genes, rather than segregated in TE-rich regions of the genome. Approximately 300 hypomethylated or unmethylated TEs are transcriptionally active, with higher expression levels in free-living mycelium compared to fruitbody. Indeed, multiple TE-enriched, copy number variant regions bearing a significant fraction of hypomethylated and expressed TEs are found almost exclusively in free-living mycelium. A reduction of DNA methylation, restricted to non-CpG sites and accompanied by an increase in TE expression, is observed upon treatment of free-living mycelia with 5-azacytidine. Conclusions: Evidence derived from analysis of the T. melanosporum methylome indicates that a non-exhaustive, partly reversible, methylation process operates in truffles. This allows for the existence of hypomethylated, transcriptionally active TEs that are associated with copy number variant regions of the genome. Non-exhaustive TE methylation may reflect a role of active TEs in promoting genome plasticity and the ability to adapt to sudden environmental changes
Adsorção de corante azul de metileno por diferentes métodos de obtenção de quitina de resÃduo de camarão
A indústria têxtil, muito importante para a economia mundial, gera um efluente que contém corantes e, quando descartado em corpos hÃdricos sem o tratamento adequado, pode causar impactos na saúde humana e no meio ambiente. Um desses corantes amplamente utilizado é o azul de metileno, cujas caracterÃsticas são a alta solubilidade em água e seu potencial tóxico, causando desde irritações nos olhos, náuseas e vômitos até confusão mental. Entre os potenciais adsorventes desse corante está a quitina, que é um biopolÃmero extraÃdo do exoesqueleto do camarão. Objetivando o desenvolvimento de um material adsorvente de baixo custo com potencial uso na indústria de tratamento de efluentes têxteis, verificou-se a capacidade de remoção de corante azul de metileno por quitina de resÃduo de camarão, obtida por onze diferentes metodologias. Os três tratamentos mais eficientes alcançaram aproximadamente 75% de remoção do corante, comprovando o alto poder de adsorção do resÃduo de camarão. Além de proporcionar desenvolvimento tecnológico de materiais, a pesquisa traz benefÃcios socioeconômicos para a colônia de pescadores com a utilização de resÃduo de camarão para a adsorção de outro resÃduo proveniente da indústria têxtil, contribuindo para a sustentabilidade de ambas as atividades e reduzindo o impacto ambiental.The textile industry, very important for the world economy, generates an effluent containing dyes, and which, when discarded in water bodies without proper treatment, can cause impacts to human health and the environment. One of these widely used dyes is methylene blue, whose characteristics are high solubility in water and its toxic potential, and which effects range from eye irritations, nausea, vomiting and even mental confusion. Among the potential adsorbents of this dye is chitin, which is a biopolymer extracted from the shrimp exoskeleton. Aiming at the development of a low-cost adsorbent material with potential use in the textile effluent treatment industry, the ability to remove methylene blue dye by shrimp residue chitin, obtained by eleven different methodologies, was verified. The three most efficient treatments reached approximately 75% of dye removal, proving the high adsorption power of shrimp residue. In addition to providing technological development of materials, the research brings socioeconomic benefits to the fishermen’s colony with the use of shrimp residue for the adsorption of other waste from the textile industry, contributing to the sustainability of both activities and reducing the environmental impact
Cerebrospinal fluid CXCL13 in clinically isolated syndrome patients: Association with oligoclonal IgM bands and prediction of Multiple Sclerosis diagnosis
Cerebrospinal fluid (CSF) CXCL13 was shown to correlate with markers of intrathecal inflammation and CSF oligoclonal IgM bands (IgMOB) have been associated with a more severe Multiple Sclerosis (MS) course.We correlated CSF CXCL13 levels with clinical, MRI and CSF parameters, including CSF IgMOB, in 110 Clinically Isolated Syndrome (CIS) patients.CSF CXCL13 levels correlated with CSF cell count, total protein, IgG Index and with the presence of CSF IgGOB and IgMOB.CSF CXCL13 levels ≥. 15.4. pg/ml showed a good positive predictive value and specificity for a MS diagnosis and for a clinical relapse within one year from onset
Advanced multimodal laser imaging tool for urothelial carcinoma diagnosis (AMPLITUDE)
Bladder cancer (BC) is the eleventh most diagnosed cancer worldwide. The age-standardized incidence rate (per 100 000 person/years) is 9.0 for men and 2.2 for women [1]. Urothelial carcinoma (UC) represents about 90% of all bladder tumors, thus carrying an enormous social and economic burden [2]. UCs are classified in different stages and grades, depending on their invasiveness and on their degree of cytological abnormalities. The key aspect for a positive prognosis is the early and accurate diagnosis of the lesion stage, in order to identify the most aggressive disease forms and treat them promptly. It is well known that tissue metabolism constitutes a basic mechanism, which is at the base of many pathologies, especially BC. Being able to detect and characterize tissue metabolism and molecular fingerprints at the cellular level could be a key aspect in characterizing the pathology and enabling both early detection and therapy monitoring. The new European Union Horizon 2020 project called AMPLITUDE, the ‘Advanced Multimodal Photonics Laser Imaging Tool for Urothelial Diagnosis in Endoscopy’, starting in January 2020, proposes the development of an advanced multi-modal imaging tool exploiting new laser technologies in an approach combining confocal and non-linear imaging to fulfil unmet clinical needs in terms of the specificity and accuracy of urothelial cancer diagnosis and therapy monitoring. The project is coordinated by Tampere University (Finland) and carried out in cooperation with leading European research organizations including Aston Insitute of Photonic Technologies—AIPT (UK), Consiglio Nazionale delle Ricerche—CNR (Italy), Institute of Photonic Sciences—ICFO (Spain), University of Milan-Bicocca, Modus Research and Innovation Ltd. (UK) and University of Florence (Italy), as well as industrial partners: Ampliconyx Oy (Finland), Femtonics Ltd. (Hungary), HC Photonics (Taiwan), and LEONI Fiber Optics GmbH (Germany)
A comprehensive resource of genomic, epigenomic and transcriptomic sequencing data for the black truffle Tuber melanosporum
Abstract Background: Tuber melanosporum, also known in the gastronomic community as "truffle", features one of the largest fungal genomes (125 Mb) with an exceptionally high transposable element (TE) and repetitive DNA content (>58%). The main purpose of DNA methylation in fungi is TE silencing. As obligate outcrossing organisms, truffles are bound to a sexual mode of propagation, which together with TEs is thought to represent a major force driving the evolution of DNA methylation. Thus, it was of interest to examine if and how T. melanosporum exploits DNA methylation to maintain genome integrity. Findings: We performed whole-genome DNA bisulfite sequencing and mRNA sequencing on different developmental stages of T. melanosporum; namely, fruitbody ("truffle"), free-living mycelium and ectomycorrhiza. The data revealed a high rate of cytosine methylation (>44%), selectively targeting TEs rather than genes with a strong preference for CpG sites. Whole genome DNA sequencing uncovered multiple TE-enriched, copy number variant regions bearing a significant fraction of hypomethylated and expressed TEs, almost exclusively in free-living mycelium propagated in vitro. Treatment of mycelia with 5-azacytidine partially reduced DNA methylation and increased TE transcription. Our transcriptome assembly also resulted in the identification of a set of novel transcripts from 614 genes
Non-pathogenic Neisseria species of the oropharynx as a reservoir of antimicrobial resistance: a cross-sectional study
Commensal Neisseria species of the oropharynx represent a significant reservoir of antimicrobial resistance determinants that can be transferred to Neisseria gonorrhoeae. This aspect is particularly crucial in ‘men having sex with men’ (MSM), a key population in which pharyngeal co-colonization by N. gonorrhoeae and non-pathogenic Neisseria species is frequent and associated with the emergence of antimicrobial resistance. Here, we explored the antimicrobial susceptibility of a large panel of non-pathogenic Neisseria species isolated from the oropharynx of two populations: a group of MSM attending a ‘sexually transmitted infection’ clinic in Bologna (Italy) (n=108) and a group of males representing a ‘general population’ (n=119). We collected 246 strains, mainly belonging to N. subflava (60%) and N. flavescens (28%) species. Their antimicrobial susceptibility was evaluated assessing the minimum inhibitory concentrations (MICs) for azithromycin, ciprofloxacin, cefotaxime, and ceftriaxone using E-test strips. Overall, commensal Neisseria spp. showed high rates of resistance to azithromycin (90%; median MICs: 4.0 mg/L), and ciprofloxacin (58%; median MICs: 0.12 mg/L), whereas resistance to cephalosporins was far less common (<15%). Neisseria strains from MSM were found to have significantly higher MICs for azithromycin (p=0.0001) and ciprofloxacin (p<0.0001) compared to those from the general population. However, there was no significant difference in cephalosporin MICs between the two groups. The surveillance of the antimicrobial resistance of non-pathogenic Neisseria spp. could be instrumental in predicting the risk of the spread of multi-drug resistant gonorrhea. This information could be an early predictor of an excessive use of antimicrobials, paving the way to innovative screening and prevention policies
Quantitation of regional ejection fractions using gated tomographic imaging with Tc-99m-sestamibi
BACKGROUND:
A new oro-dispersible film (ODF) formulation of sildenafil has been developed for the treatment of erectile dysfunction (ED) to overcome the drawbacks that some patients experience when taking the conventional film-coated tablet (FCT).
AIM:
To assess the effectiveness and safety of sildenafil ODF formulation in patients with ED who were using the conventional FCT.
METHODS:
From May 2017 through July 2017, 139 patients with ED were enrolled. Data from penile color-duplex ultrasound, medical history, hormonal evaluation, and patient self-administered questionnaires were collected. All patients were administered sildenafil 100-mg FCT for 4 weeks. Thereafter, they underwent a 2-week washout period and subsequently took sildenafil 75-mg ODF for 4 weeks.
OUTCOMES:
The International Index of Erectile Function (IIEF-15), Hospital Anxiety and Depression Scale (HADS), Patient Global Impressions of Improvement (PGI-I), and Clinician Global Impressions of Improvement (CGI-I) questionnaires were administered and severity of ED was classified as severe (IIEF-15 score ≤ 10), moderate (IIEF-15 score 11-16), or mild (IIEF-15 score = 17-25).
RESULTS:
All patients completed the final protocol. Differences in mean IIEF scores for erectile function, orgasmic function, sexual desire, and intercourse satisfaction were significantly in favor of sildenafil 100-mg FCT, whereas the mean score for overall satisfaction was in favor of sildenafil 75-mg ODF. A significant difference in changes in HADS score was found from washout to final follow-up (mean difference = -0.19; P < .01). For the ODF formulation, the median CGI-I score was 3.5 (interquartile range [IQR] = 2.5-4.5) and the median PGI-I score was 3.0 (IQR = 2.0-4.0). The median action time was 20.0 minutes (IQR = 15.0-30.0) and the median mouth time was 60.0 seconds (IQR = 30.0-120.0).
CLINICAL IMPLICATIONS:
The ODF formulation of a widely known drug, with the same safety and effectiveness of the FCT, was better appreciated by patients in overall satisfaction.
STRENGTHS AND LIMITATIONS:
This is the first clinical trial to assess the efficacy of a new formulation of sildenafil in patients with ED. The limitations of the study are related to the methodology used: it was not a case-control study and the patients were not drug-naïve for ED treatment. Therefore, only the "additional" side effects of the ODF formulation compared with FCT are reported.
CONCLUSION:
The new ODF formulation is as efficient and safe as the FCT formulation and offers a new choice of treatment to specialists for more precisely tailored therapy. Cocci A, Capece M, Cito G, et al. INFN Camera demonstrator for the Cherenkov Telescope Array
The Cherenkov Telescope Array is a world-wide project for a new generation of
ground-based Cherenkov telescopes of the Imaging class with the aim of
exploring the highest energy region of the electromagnetic spectrum. With two
planned arrays, one for each hemisphere, it will guarantee a good sky coverage
in the energy range from a few tens of GeV to hundreds of TeV, with improved
angular resolution and a sensitivity in the TeV energy region better by one
order of magnitude than the currently operating arrays. In order to cover this
wide energy range, three different telescope types are envisaged, with
different mirror sizes and focal plane features. In particular, for the highest
energies a possible design is a dual-mirror Schwarzschild-Couder optical
scheme, with a compact focal plane. A silicon photomultiplier (SiPM) based
camera is being proposed as a solution to match the dimensions of the pixel
(angular size of ~ 0.17 degrees). INFN is developing a camera demonstrator made
by 9 Photo Sensor Modules (PSMs, 64 pixels each, with total coverage 1/4 of the
focal plane) equipped with FBK (Fondazione Bruno Kessler, Italy) Near
UltraViolet High Fill factor SiPMs and Front-End Electronics (FEE) based on a
Target 7 ASIC, a 16 channels fast sampler (up to 2GS/s) with deep buffer,
self-trigger and on-demand digitization capabilities specifically developed for
this purpose. The pixel dimensions of mm lead to a very compact
design with challenging problems of thermal dissipation. A modular structure,
made by copper frames hosting one PSM and the corresponding FEE, has been
conceived, with a water cooling system to keep the required working
temperature. The actual design, the adopted technical solutions and the
