    Brain injury : the impact on family members living with a brain injured adult and implications for rehabilitation services

    Chapter One: This chapter reviews the relevant literature published since 1990 that has addressed the psychological impact both progressive and acute brain injury can have upon spouses of patients. The papers reviewed suggest that psychological strain or burden experienced by spouse carers is associated with a number of factors irrespective of illness type. A number of methodological limitations of the studies reviewed are discussed along with implications for clinical practice. Further research is required that explores the experience of family members to ascertain the best way forward for rehabilitation services in terms of providing family orientated interventions and support. Chapter Two: Chapter two presents a study that explores the views, beliefs and experiences of brain injury rehabilitation professionals on working collaboratively with families. Using a qualitative research approach, two focus groups were conducted with a total of 12 professionals from a range of disciplines. Focus groups were analysed using thematic analysis. A number of salient themes emerged that reflected the narrative collected. Implications for brain injury rehabilitation services and suggestions for further research are made. Chapter Three: Chapter three presents an empirical study that explores children's experiences of parental brain injury. Using a qualitative research approach interviews were conducted with twelve participants aged between 11 and 18 years living at home with a brain-injured parent. Interview transcripts were analysed using Interpretative Phenomenological Analysis (IPA). A number of themes emerged that reflected the narrative collected. A preliminary model of adaptation to parental brain injury is proposed. Implications for brain injury rehabilitation services in terms of addressing the needs of children and suggestions for further research are discussed. Chapter Four: This chapter documents the first author's experience of conducting the two empirical papers presented in chapters two and three. Specifically it highlights a number of ethical and methodological concerns associated to conducting focus groups and interviewing children.     Non-destructive Detection of an Invasive Drywood Termite, Cryptotermes brevis (Blattodea: Kalotermitidae), in Timber

    Reliable drywood termite detection in structures is challenging but is critical for effective management. A microwave-based non-destructive method was evaluated for detecting termite activity. This study evaluated factors affecting the ability of this device to reliably detect Cryptotermes brevis in timber. The device displayed a high probability of successfully detecting C. brevis in naturally infested boards. The system detected termites 97% of the time when used at the highest sensitivity level, while producing few false positives. The number of termites did not affect detection ability, and detectable signals were produced even when a single termite was present. Detection success decreased with both increasing wood density and testing perpendicular to the grain in abrupt transition timber species. The device detected termites to a maximum depth of 45 mm in southern pine (Pinus spp.), but sensitivity declined with increased wood density with the detection limit declining to only 20 mm in denser Tasmanian oak (Eucalyptus spp). The device could only detect termites in samples with densities between 392 to 511 kg/m3 in 38 mm thick radiata pine samples. The results support the ability of microwaves to reliably detect C. brevis in timber.

    Age and Prostate-Specific Antigen Level Prior to Diagnosis Predict Risk of Death from Prostate Cancer.

    A single early prostate-specific antigen (PSA) level has been correlated with a higher likelihood of prostate cancer diagnosis and death in younger men. PSA testing in older men has been considered of limited utility. We evaluated prostate cancer death in relation to age and PSA level immediately prior to prostate cancer diagnosis. Using the Veterans Affairs database, we identified 230,081 men aged 50-89 years diagnosed with prostate cancer and at least one prior PSA test between 1999 and 2009. Prostate cancer-specific death over time was calculated for patients stratified by age group (e.g., 50-59 years, through 80-89 years) and PSA range at diagnosis (10 ranges) using Kaplan-Meier methods. Risk of 10-year prostate cancer mortality across age and PSA was compared using log-rank tests with a Bonferroni adjustment for multiple testing. 10.5% of men diagnosed with prostate cancer died of cancer during the 10-year study period (mean follow-up = 3.7 years). Higher PSA values prior to diagnosis predict a higher risk of death in all age groups (p < 0.0001). Within the same PSA range, older age groups are at increased risk for death from prostate cancer (p < 0.0001). For PSA of 7-10 ng/mL, cancer-specific death, 10 years after diagnosis, increased from 7% for age 50-59 years to 51% for age 80-89 years. Men older than 70 years are more likely to die of prostate cancer at any PSA level than younger men, suggesting prostate cancer remains a significant problem among older men (even those aged 80+) and deserves additional study

    Spectral Variations of Of?p Oblique Magnetic Rotator Candidates in the Magellanic Clouds

    Optical spectroscopic monitoring has been conducted of two O stars in the Small and one in the Large Magellanic Cloud, the spectral characteristics of which place them in the Of?p category, which has been established in the Galaxy to consist of oblique magnetic rotators. All of these Magellanic stars show systematic spectral variations typical of the Of?p class, further strengthening their magnetic candidacy to the point of virtual certainty. The spectral variations are related to photometric variations derived from OGLE data by Naze et al. (2015) in a parallel study, which yields rotational periods for two of them. Now circular spectropolarimetry is required to measure their fields, and ultraviolet spectroscopy to further characterize their low-metallicity, magnetically confined winds, in support of hydrodynamical analyses.Comment: 18 pages, 6 figures, accepted for publication by A

    Background. A birth cohort study, the Birth to Ten (BIT) study, commenced in the greater Johannesburg/Soweto metropole in South Africa in 1990. The overall BIT project collected antenatal, birth and early development information on these children as well as information that could help identify factors related to the emergence of risk of cardiovascular diseases (CVDs) in children.Objective. To determine CVD risk profiles and their determinants in 5-year-old children living in an urban environment in South Africa.Methods. Demographic and birth characteristics were collected on a sample of 964 5-year-olds whose parents agreed for blood samples to be taken from their children. The children's height and weight were measured using standardised procedures; blood pressure (BP) was measured with a Dinamap Vital Signs Monitor, and a nonfasting blood sample was drawn for lipid deterrninations. Wormation on exposure to tobacco smoke and additional health-related data were obtained by interview.Results. No differences were found between the birth weight and gestational age of the 5-year-old CVD participants and the remainder of the children studied at birth.     Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood.

    Historically, the limited availability of primary endothelial cells from patients with vascular disorders has hindered the study of the molecular mechanisms underlying endothelial dysfunction in these individuals. However, the recent identification of blood outgrowth endothelial cells (BOECs), generated from circulating endothelial progenitors in adult peripheral blood, may circumvent this limitation by offering an endothelial-like, primary cell surrogate for patient-derived endothelial cells. Beyond their value to understanding endothelial biology and disease modeling, BOECs have potential uses in endothelial cell transplantation therapies. They are also a suitable cellular substrate for the generation of induced pluripotent stem cells (iPSCs) via nuclear reprogramming, offering a number of advantages over other cell types. We describe a method for the reliable generation, culture and characterization of BOECs from adult peripheral blood for use in these and other applications. This approach (i) allows for the generation of patient-specific endothelial cells from a relatively small volume of adult peripheral blood and (ii) produces cells that are highly similar to primary endothelial cells in morphology, cell signaling and gene expression

    Non–Destructive Imaging of Phytosulfokine Trafficking Using a Fiber–Optic Fluorescence Microscope

    Plants secrete peptide ligands and use receptor signaling to respond to stress and control development. Understanding the signaling mechanisms and associated molecular trafficking is key to improving plant health and productivity for food, fiber and energy applications. However, one of the challenges to elucidating communication pathways in plants is to study the trafficking of molecules and signals iteratively and non-destructively. This study focuses on using fiber-optic fluorescence microscopy to image live plants iteratively and non-destructively after delivering both labeled and unlabeled phytosulfokine (PSK) into the plant. PSK is a sulfated peptide hormone involved in the regulation of plant cell division and growth via specific receptors, PSKRs. It also plays a role in regulating how plants are able to tolerate stress conditions. The microscope provides two-color (FITC/TRITC) optics and provides high-resolution (3–5 µm) epifluorescence micrographs via a 1-m coherent imaging fiber and a GRIN objective lens. To obtain high-quality images, the fiber was mounted either to a conventional upright microscope body equipped with a leaf compressor, or to a leaf clip with 5-axis positioning (X–Y–Z plus pitch and yaw) mounted on an extensible arm. PSK and TAMRA-labelled PSK were delivered into the roots of various Arabidopsis thaliana genotypes (wt; receptor-deficient: pskr1/pskr2; and tagged receptor overproducing: PSKR1‑GFP), and their movement in roots and leaves was tracked with the fiber-optic fluorescence microscope. Peptide trafficking was successfully observed in live plants non- destructively, confirming that PSK is mobile in both wt and receptor-deficient plants. Preliminary results suggest that the level of receptor PSKR1 may change in response to PSK, and that levels of PSKR1, PSKR2 or both may impact the trafficking of PSK. Understanding how PSK is trafficked in plants will offer insights into how we can improve plants health and productivity

    The Carnegie Supernova Project: Analysis of the First Sample of Low-Redshift Type-Ia Supernovae

    We present the analysis of the first set of low-redshift Type Ia supernovae (SNe Ia) by the Carnegie Supernova Project. Well-sampled, high-precision optical (ugriBV) and near-infrared (NIR; YJHKs) light curves obtained in a well-understood photometric system are used to provide light-curve parameters, and ugriBVYJH template light curves. The intrinsic colors at maximum light are calibrated to compute optical--NIR color excesses for the full sample, thus allowing the properties of the reddening law in the host galaxies to be studied. A low value of Rv~1.7, is derived when using the entire sample of SNe. However, when the two highly reddened SNe in the sample are excluded, a value Galactic standard of Rv~3.2 is obtained. The colors of these two events are well matched by a reddening model due to circumstellar dust. The peak luminosities are calibrated using a two-parameter linear fit to the decline rates and the colors, or alternatively, the color excesses. In both cases, dispersions in absolute magnitude of 0.12--0.16 mag are obtained, depending on the filter-color combination. In contrast to the results obtained from color excesses, these fits give Rv~1--2, even when the two highly reddened SNe are excluded. This discrepancy suggests that, beyond the "normal" interstellar reddening produced in the host galaxies, there is an intrinsic dispersion in the colors of SNe Ia which is correlated with luminosity but independent of the decline rate. Finally, a Hubble diagram is produced by combining the results of the fits for each filter. The resulting scatter of 0.12 mag appears to be limited by peculiar velocities as evidenced by the strong correlation between the distance-modulus residuals among the different filters.
