    Immune-Endocrine Links to Gregariousness in Wild House Mice

    Social interactions are critically important for survival and impact overall-health, but also impose costs on animals, such as exposure to contagious agents. The immune system can play a critical role in modulating social behavior when animals are sick, as has been demonstrated within the context of “sickness behaviors.” Can immune molecules affect or be affected by social interactions even when animals are not sick, therefore serving a role in mediating pathogen exposure? We tested whether markers of immune function in both the blood and the brain are associated with gregariousness, quantified as number of animals interacted with per day. To do this, we used remote tracking of social interactions of a wild population of house mice (Mus musculus domesticus) to categorize animals in terms of gregariousness. Blood, hair, brain and other tissue samples from animals with extreme gregariousness phenotypes were collected. We then tested whether the levels of three important cytokines (TNF-a, IFN-g and IL- 1b) in the serum, cortex and hypothalamus of these animals could be explained by the gregariousness phenotype and/or sex of the mice. Using the hair as a long-term quantification of steroid hormones, we also tested whether corticosterone, progesterone and testosterone differed by social phenotype. We found main effects of gregariousness and sex on the serum levels of TNF-a, but not on IFN-g or IL-1b. Brain gene expression levels were not different between phenotypes. All hair steroids tended to be elevated in animals of high gregariousness phenotype, independent of sex. In sum, elements of the immune system may be associated with gregariousness, even outside of major disease events. These results extend our knowledge of the role that immune signals have in contributing to the regulation of social behaviors outside periods of illness

    Curriculum Transformation: Redesigning a Construction Management program to a Vertically and Horizontally Integrated Curriculum with Authentic Project-Based Learning

    Short Abstract: The Construction Management program at Purdue University is redesigning their curriculum to a vertically and horizontally integrated program. The curriculum will utilize authentic projects from industry partners, providing a basis for project-based learning and instruction. All faculty will team-teach the large project based courses developed for the new curriculum. Full Abstract: Traditional higher education provides students with a variety of specialized courses, combined to create a curriculum, which then translates into a major area of study. While prerequisites and course numbers provide a road map to the students for the order of classes, there is no guarantee the concepts from one course are built upon by another course. Typically, a senior capstone is the first time students are required to synthesize accumulated skills and knowledge. For the past two years, the Construction Management program at Purdue University has been redesigning their curriculum to integrate construction concepts both vertically and horizontally throughout the entire 4-year program. The new curriculum will also utilize authentic projects obtained from industry partners, which will provide the basis of the project-based learning and instruction. Team teaching by all faculty within the department will be applied in the large project based courses. While the technical information presented will remain the same, it is the hope of the faculty this new curriculum will develop students who are better prepared for the construction industry by having a higher level of written and verbal communication, increased critical thinking and problem solving skills, and display greater professionalism and initiative

    CCL2 Overexpression in the Brain Promotes Glial Activation and Accelerates Tau Pathology in a Mouse Model of Tauopathy

    Innate immune activation is a major contributor to Alzheimer’s Disease (AD) pathophysiology, although the mechanisms involved are poorly understood. Chemokine C-C motif ligand (CCL) 2 is produced by neurons and glial cells and is upregulated in the AD brain. Transgene expression of CCL2 in mouse models of amyloidosis produces microglia-induced amyloid β oligomerization, a strong indication of the role of these activation pathways in the amyloidogenic processes of AD. We have previously shown that CCL2 polarizes microglia in wild type mice. However, how CCL2 signaling contributes to tau pathogenesis remains unknown. To address this question, CCL2 was delivered via recombinant adeno-associated virus serotype 9 into both cortex and hippocampus of a mouse model with tau pathology (rTg4510). We report that CCL2 overexpression aggravated tau pathology in rTg4510 as shown by the increase in Gallyas stained neurofibrillary tangles as well as phosphorylated tau-positive inclusions. In addition, biochemical analysis showed a reduction in the levels of detergent-soluble tau species followed by increase in the insoluble fraction, indicating a shift toward larger tau aggregates. Indeed, increased levels of high molecular weight species of phosphorylated tau were found in the mice injected with CCL2. We also report that worsening of tau pathology following CCL2 overexpression was accompanied by a distinct inflammatory response. We report an increase in leukocyte common antigen (CD45) and Cluster of differentiation 68 (CD68) expression in the brain of rTg4510 mice without altering the expression levels of a cell-surface protein Transmembrane Protein 119 (Tmem119) and ionized calcium-binding adaptor molecule 1 (Iba-1) in resident microglia. Furthermore, the analysis of cytokines in brain extract showed a significant increase in interleukin (IL)-6 and CCL3, while CCL5 levels were decreased in CCL2 mice. No changes were observed in IL-1α, IL-1β, TNF-α. IL-4, Vascular endothelial growth factor-VEGF, IL-13 and CCL11. Taken together our data report for the first time that overexpression of CCL2 promotes the increase of pathogenic tau species and is associated with glial neuroinflammatory changes that are deleterious. We propose that these events may contribute to the pathogenesis of Alzheimer’s disease and other tauopathies

    The properties of the 2175AA extinction feature discovered in GRB afterglows

    The unequivocal, spectroscopic detection of the 2175 bump in extinction curves outside the Local Group is rare. To date, the properties of the bump have been examined in only two GRB afterglows (GRB 070802 and GRB 080607). In this work we analyse in detail the detections of the 2175 extinction bump in the optical spectra of the two further GRB afterglows: GRB 080605 and 080805. We gather all available optical/NIR photometric, spectroscopic and X-ray data to construct multi-epoch SEDs for both GRB afterglows. We fit the SEDs with the Fitzpatrick & Massa (1990) model with a single or broken PL. We also fit a sample of 38 GRB afterglows, known to prefer a SMC-type extinction curve, with the same model. We find that the SEDs of GRB 080605 and GRB 080805 at two epochs are fit well with a single PL with a derived extinction of A_V = 0.52(+0.13 -0.16) and 0.50 (+0.13 -0.10), and 2.1(+0.7-0.6) and 1.5+/-0.2 respectively. While the slope of the extinction curve of GRB 080805 is not well-constrained, the extinction curve of GRB 080605 has an unusual very steep far-UV rise together with the 2175 bump. Such an extinction curve has previously been found in only a small handful of sightlines in the MW. One possible explanation of such an extinction curve may be dust arising from two different regions with two separate grain populations, however we cannot distinguish the origin of the curve. We finally compare the four 2175 bump sightlines to the larger GRB afterglow sample and to Local Group sightlines. We find that while the width and central positions of the bumps are consistent with what is observed in the Local Group, the relative strength of the detected bump (A_bump) for GRB afterglows is weaker for a given A_V than for almost any Local Group sightline.     Technological Advances to Address Current Issues in Entomology: 2020 Student Debates

    The 2020 Student Debates of the Entomological Society of America (ESA) were live-streamed during the Virtual Annual Meeting to debate current, prominent entomological issues of interest to members. The Student Debates Subcommittee of the National ESA Student Affairs Committee coordinated the student efforts throughout the year and hosted the live event. This year, four unbiased introductory speakers provided background for each debate topic while four multi-university teams were each assigned a debate topic under the theme ‘Technological Advances to Address Current Issues in Entomology’. The two debate topics selected were as follows: 1) What is the best taxonomic approach to identify and classify insects? and 2) What is the best current technology to address the locust swarms worldwide? Unbiased introduction speakers and debate teams began preparing approximately six months before the live event. During the live event, teams shared their critical thinking and practiced communication skills by defending their positions on either taxonomical identification and classification of insects or managing the damaging outbreaks of locusts in crops

    Overview of the marine litter status in the Atlantic Area: beach, floating and seabed litter

    CleanAtlantic is an INTERREG Atlantic Area Programme project that aimed at protecting biodiversity and ecosystem services in the Atlantic Area by improving capabilities to monitor, prevent and remove (macro) marine litter. Besides, the project also contributed to raise awareness and change attitudes among stakeholders and to improve marine litter managing systems. To achieve these aims, the work was organised in 8 work packages. The present deliverable aims at synthesizing the main results obtained on the frame of the action 1 of work package 4, which focused on the Regional characterisation of marine litter in the Atlantic Area. With this purpose, an overview of marine litter status in beach, floating and seabed compartments in the Atlantic Area is presented. Additionally, the major key findings, gaps on monitoring and research as well as potential improvements and recommendations are identified. Links to the complete dedicated reports for each compartment are included in the references section. Also, an interactive map for spatial visualization of data on beach, floating and seabed litter composition and abundance in the Atlantic Area was created and is presented at the end of this report
