Research Problems in the Management of the Northern White Pine in the Northeastern United States
Although the northern white pine type is one of the most extensive in New England, specific funds for investigative work in white pine were not made available to the Forest Service until the fiscal year 1939. However, the white pine studies conducted by the Forest Service in the early part of the century and more recently by various agencies within the region have laid a foundation for the proposed work by the Northeastern Forest Experiment Station
Polarised epithelial monolayers of the gastric mucosa reveal insights into mucosal homeostasis and defence against infection
Objective Helicobacter pylori causes life-long colonisation of the gastric mucosa, leading to chronic inflammation with increased risk of gastric cancer. Research on the pathogenesis of this infection would strongly benefit from an authentic human in vitro model. Design Antrum-derived gastric glands from surgery specimens served to establish polarised epithelial monolayers via a transient airâliquid interface culture stage to study cross-talk with H. pylori and the adjacent stroma. Results The resulting âmucosoid culturesâ, so named because they recapitulate key characteristics of the gastric mucosa, represent normal stem cell-driven cultures that can be passaged for months. These highly polarised columnar epithelial layers encompass the various gastric antral cell types and secrete mucus at the apical surface. By default, they differentiate towards a foveolar, MUC5AC-producing phenotype, whereas Wnt signalling stimulates proliferation of MUC6-producing cells and preserves stemnessâreminiscent of the gland base. Stromal cells from the lamina propria secrete Wnt inhibitors, antagonising stem-cell niche signalling and inducing differentiation. On infection with H. pylori, a strong inflammatory response is induced preferentially in the undifferentiated basal cell phenotype. Infection of cultures for several weeks produces foci of viable bacteria and a persistent inflammatory condition, while the secreted mucus establishes a barrier that only few bacteria manage to overcome. Conclusion Gastric mucosoid cultures faithfully reproduce the features of normal human gastric epithelium, enabling new approaches for investigating the interaction of H. pylori with the epithelial surface and the cross-talk with the basolateral stromal compartment. Our observations provide striking insights in the regulatory circuits of inflammation and defence.</p
Detection of viral antigen, IgM and IgG antibodies in cerebrospinal fluid of Chikungunya patients with neurological complications
BACKGROUND: During Chikungunya virus (CHIKV) epidemic in Nagpur, India, we identified some suspected Chikungunya patients with neurological complications. Early and cost-effective diagnosis of these patients remains problematic despite many new advanced diagnostic methods. A reliable diagnostic test, which could be performed in any standard pathology laboratory, would help to obtain definitive early diagnosis of CHIKV patients with neurological complications. In our laboratory, in-house ELISA protocol for viral antigen, immunoglobulin M (IgM) and IgG detection has been developed and assessed for the diagnosis of CHIKV patients with neurological complications. METHOD: Cerebrospinal fluid samples of forty-six patients who developed neurological symptoms within two months of CHIKV infections along with control subjects were included in the study and were analyzed for the presence of antigens and of IgM and IgG using an ELISA protocol. RESULTS: The ELISA method for antigen detection yielded 80% sensitivity and 87% specificity for the diagnosis of CHIKV patients with neurological complications. The sensitivity for detection of IgM 48% or IgG 63% was significantly lower than the antigen assay (80%). CONCLUSION: The detection of viral antigen in CSF of CHIKV patients with neurological complications by ELISA method gave a more reliable diagnosis than antibodies detection that can be used to develop an immunodiagnostic assay with increased sensitivity and specificity
The Role of Mesotocin on Social Bonding in Pinyon Jays
The neuropeptide oxytocin influences mammalian social bonding by facilitating the building and maintenance of parental, sexual, and sameâsex social relationships. However, we do not know whether the function of the avian homologue mesotocin is evolutionarily conserved across birds. While it does influence avian prosocial behavior, mesotocin\u27s role in avian social bonding remains unclear. Here, we investigated whether mesotocin regulates the formation and maintenance of sameâsex social bonding in pinyon jays (Gymnorhinus cyanocephalus), a member of the crow family. We formed squads of four individually housed birds. In the first, âpairâformationâ phase of the experiment, we repeatedly placed pairs of birds from within the squad together in a cage for short periods of time. Prior to entering the cage, we intranasally administered one of three hormone solutions to both members of the pair: mesotocin, oxytocin antagonist, or saline. Pairs received repeated sessions with administration of the same hormone. In the second, âpairâmaintenanceâ phase of the experiment, all four members of the squad were placed together in a large cage, and no hormones were administered. For both phases, we measured the physical proximity between pairs as our proxy for social bonding. We found that, compared with saline, administering mesotocin or oxytocin antagonist did not result in different proximities in either the pairâformation or pairâmaintenance phase of the experiment. Therefore, at the dosages and time frames used here, exogenously introduced mesotocin did not influence sameâsex social bond formation or maintenance. Like oxytocin in mammals, mesotocin regulates avian prosocial behavior; however, unlike oxytocin, we do not have evidence that mesotocin regulates social bonds in birds

Integrated cross-domain object storage in working memory: Evidence from a verbal-spatial memory task
Working-memory theories often include domain-specific verbal and visual stores (e.g., the phonological and visuospatial buffers of Baddeley, 1986), and some also posit more general stores thought to be capable of holding verbal or visuospatial materials (Baddeley, 2000; Cowan, 2005). However, it is currently unclear which type of store is primarily responsible for maintaining objects that include components from multiple domains. In these studies, a spatial array of letters was followed by a single probe identical to an item in the array or differing systematically in spatial location, letter identity, or their combination. Concurrent verbal rehearsal suppression impaired memory in each of these trial types in a task that required participants to remember verbal-spatial binding, but did not impair memory for spatial locations if the task did not require verbal-spatial binding for a correct response. Thus, spatial information might be stored differently when it must be bound to verbal information. This suggests that a cross-domain store such as the episodic buffer of Baddeley (2000) or the focus of attention of Cowan (2001) might be used for integrated object storage, rather than the maintenance of associations between features stored in separate domain-specific buffers
Residual cognitive deficits 50 years after lead poisoning during childhood
The long term neurobehavioural consequences of childhood lead poisoning are not known. In this study adult subjects with a documented history of lead poisoning before age 4 and matched controls were examined with an abbreviated battery of neuropsychological tests including measures of attention, reasoning, memory, motor speed, and current mood. The subjects exposed to lead were inferior to controls on almost all of the cognitive tasks. This pattern of widespread deficits resembles that found in children evaluated at the time of acute exposure to lead rather than the more circumscribed pattern typically seen in adults exposed to lead. Despite having completed as many years of schooling as controls, the subjects exposed to lead were lower in lifetime occupational status. Within the exposed group, performance on the neuropsychological battery and occupational status were related, consistent with the presumed impact of limitations in neuropsychological functioning on everyday life. The results suggest that many subjects exposed to lead suffered acute encephalopathy in childhood which resolved into a chronic subclinical encephalopathy with associated cognitive dysfunction still evident in adulthood. These findings lend support to efforts to limit exposure to lead in childhood
Persistent Transport Barrier on the West Florida Shelf
Analysis of drifter trajectories in the Gulf of Mexico has revealed the
existence of a region on the southern portion of the West Florida Shelf (WFS)
that is not visited by drifters that are released outside of the region. This
so-called ``forbidden zone'' (FZ) suggests the existence of a persistent
cross-shelf transport barrier on the southern portion of the WFS. In this
letter a year-long record of surface currents produced by a Hybrid-Coordinate
Ocean Model simulation of the WFS is used to identify Lagrangian coherent
structures (LCSs), which reveal the presence of a robust and persistent
cross-shelf transport barrier in approximately the same location as the
boundary of the FZ. The location of the cross-shelf transport barrier undergoes
a seasonal oscillation, being closer to the coast in the summer than in the
winter. A month-long record of surface currents inferred from high-frequency
(HF) radar measurements in a roughly 60 km 80 km region on the WFS off
Tampa Bay is also used to identify LCSs, which reveal the presence of robust
transient transport barriers. While the HF-radar-derived transport barriers
cannot be unambiguously linked to the boundary of the FZ, this analysis does
demonstrate the feasibility of monitoring transport barriers on the WFS using a
HF-radar-based measurement system. The implications of a persistent cross-shelf
transport barrier on the WFS for the development of harmful algal blooms on the
shoreward side of the barrier are considered.Comment: Submitted to Geophysical Research Letter
Symbolic powers of monomial ideals and Cohen-Macaulay vertex-weighted digraphs
In this paper we study irreducible representations and symbolic Rees algebras
of monomial ideals. Then we examine edge ideals associated to vertex-weighted
oriented graphs. These are digraphs having no oriented cycles of length two
with weights on the vertices. For a monomial ideal with no embedded primes we
classify the normality of its symbolic Rees algebra in terms of its primary
components. If the primary components of a monomial ideal are normal, we
present a simple procedure to compute its symbolic Rees algebra using Hilbert
bases, and give necessary and sufficient conditions for the equality between
its ordinary and symbolic powers. We give an effective characterization of the
Cohen--Macaulay vertex-weighted oriented forests. For edge ideals of transitive
weighted oriented graphs we show that Alexander duality holds. It is shown that
edge ideals of weighted acyclic tournaments are Cohen--Macaulay and satisfy
