    In-situ methane enrichment in continuous anaerobic digestion of pig slurry by zero-valent iron nanoparticles addition under mesophilic and thermophilic conditions

    The effect of zero-valent iron nanoparticles (nZVI) addition on methane production during anaerobic digestion of pig slurry was assessed. Experiments were conducted using two experimental set-ups: batch and long-term continuous operation at a fixed nZVI dosage.     ARID2 deficiency promotes tumor progression and is associated with higher sensitivity to chemotherapy in lung cancer

    The survival rate in lung cancer remains stubbornly low and there is an urgent need for the identification of new therapeutic targets. In the last decade, several members of the SWI/SNF chromatin remodeling complexes have been described altered in different tumor types. Nevertheless, the precise mechanisms of their impact on cancer progression, as well as the application of this knowledge to cancer patient management are largely unknown. In this study, we performed targeted sequencing of a cohort of lung cancer patients on genes involved in chromatin structure. In addition, we studied at the protein level the expression of these genes in cancer samples and performed functional experiments to identify the molecular mechanisms linking alterations of chromatin remodeling genes and tumor development. Remarkably, we found that 20% of lung cancer patients show ARID2 protein loss, partially explained by the presence of ARID2 mutations. In addition, we showed that ARID2 deficiency provokes profound chromatin structural changes altering cell transcriptional programs, which bolsters the proliferative and metastatic potential of the cells both in vitro and in vivo. Moreover, we demonstrated that ARID2 deficiency impairs DNA repair, enhancing the sensitivity of the cells to DNA-damaging agents. Our findings support that ARID2 is a bona fide tumor suppressor gene in lung cancer that may be exploited therapeutically.Financial Support: I.     The synthetic molecule stauprimide impairs cell growth and migration in triple-negative breast cancer

    Stauprimide, a semi-synthetic derivative of staurosporine, is known mainly for its potent differentiation-enhancing properties in embryonic stem cells. Here, we studied the effects of stauprimide in cell growth and migration of triple-negative breast cancer cells in vitro, evaluating its potential antitumoral activity in an orthotopic mouse model of breast cancer in vivo. Our results from survival curves, EdU incorporation, cell cycle analysis and annexin-V detection in MDA-MB-231 cells indicated that stauprimide inhibited cell proliferation, arresting cell cycle in G2/M without induction of apoptosis. A decrease in the migratory capability of MDA-MB-231 was also assessed in response to stauprimide. In this work we pointed to a mechanism of action of stauprimide involving the modulation of ERK1/2, Akt and p38 MAPK signalling pathways, and the downregulation of MYC in MDA-MB-231 cells. In addition, orthotopic MDA-MB-231 xenograft and 4T1 syngeneic models suggested an effect of stauprimide in vivo, increasing the necrotic core of tumors and reducing metastasis in lung and liver of mice. Together, our results point to the promising role of stauprimide as a putative therapeutic agent in triple-negative breast cancer.MRI experiments were performed in the ICTS “NANBIOSIS”, more specifically in the U28 Unit at the Andalusian Centre for Nanomedicine & Biotechnology (BIONAND).     Palimpsestos y cambios culturales en el límite Pleistoceno-Holoceno: el conjunto lítico de Picamoixons (Alt Camp, Tarragona)

    This paper presents the technological and typological study of the lithic assemblage recovered at Picamoixons during the field season carried out in 1993. Its technotypological features, the differential conservation of the assemblage, the spatial distribution of lithic remains and the radiometric dates allow us to divide the assemblage into two different sets and suggest that they belong to different chronological and cultural periods. The items included in the first one (CP) show a strong patina and should be included in the macrolithic Mesolithic, while the second one’s items (CNP) are not patinated and belong to the final part of upper Palaeolithic.En este artículo se presenta el análisis tecnológico y tipológico de la industria lítica recuperada en el año 1993 en el yacimiento de Picamoixons. Dadas sus características, su estado de conservación diferencial, su distribución espacial y las dataciones radiométricas disponibles, podemos concluir la existencia de dos conjuntos líticos pertenecientes a dos momentos cronológicos y culturales diferentes.     News and updates in the treatment of localized stage triple-negative breast cancer

    Compared to other breast cancer subtypes, triple-negative breast cancer presents a worse prognosis and higher mortality. Even in localized stages, the risk of relapse is high, especially in patients with ≥ cT2 and/or ≥ cN1. We know that those patients who achieve a complete pathologic response after neoadjuvant treatment have better disease-free survival. Therefore, many research efforts have been made to try to optimize neoadjuvant chemo/immunotherapy to increase pathologic complete response rates. The available evidence related to that subject matter is summarized in this article. In the field of adjuvant therapy, the challenge of improving disease-free survival in those patients who do not achieve pathologic complete response after neoadjuvant therapy stands out. The second part of this article will deal with the challenges inherent to this issue

    Psychoeducational intervention in psychology students on suicide through a lecture and the implementation of an emotional kit

    [EN] Suicide is a global health problem that has been aggravated by the COVID-19 pandemic. Despite its great psychosocial relevance, there are a large number of myths about suicide. The aim of the study was to raise awareness of basic aspects of suicide through attractive teaching methodologies that combine the virtual and face-to-face environments and that allow the theoretical and practical knowledge necessary to assess, intervene and prevent the suicide to be integrated in a cooperative manner. Ninety-two students from the Psychology Degree at the University of Valencia participated (M=21.04; SD=1.81). First, they completed a battery of questions about myths and realities of suicide; then, they received the training; and, finally, they completed the battery again. The results showed that students obtained a higher mean number of most psychologically accepted responses answers in the identification of myths and realities after the training (t=-5.87, p<.001). The presence of myths related to suicide among university students is evident. It is also concluded that the theoretical-practical training presented in this study could be a useful tool to enhance the acquisition of knowledge and skills in students about suicide assessment, prevention and intervention.[ES] El suicidio consituye un problema de salud mundial que ha sido agravado por la pandemia de COVID-19. A pesar de su gran relevancia psicosocial, existe un elevado número de mitos acerca del suicidio. El objetivo del estudio fue dar a conocer aspectos básicos sobre el suicidio a través de metodologías docentes atractivas que combinan el entorno virtual con el presencial y que permiten integrar de forma cooperativa los conocimientos teórico-prácticos necesarios para evaluar, intervenir y prevenir el suicidio. Participaron 92 estudiantes del Grado de Psicología de la Universitat de València (M=21,04; DT=1,81). Primero, completaron una batería de preguntas acerca de mitos y realidades del suicidio; a continuación, recibieron la formación; y, finalmente, completaron la batería de nuevo. Los resultados mostraron que el alumnado obtuvo una media de respuestas psicológicamente más aceptadas más elevada en la identificación de mitos y realidades tras la formación (t=-5,87; p&lt;,001). Se evidencia la presencia de mitos relacionados con el suicidio entre el estudiantado universitario.     Application of new indicators to assess the quality of antimicrobial use in intensive care units

    This study explored the feasibility of a bundle of indicators aimed at assessing the quality of antimicrobial use in intensive care units (ICUs) through an observational prospective study spanning 12 quarters (January 2019-December 2021) in a 1290-bed teaching hospital in Spain. Members of the antimicrobial stewardship programme team selected the indicators to analyse the quality of antimicrobial use based on consumption data from a list proposed in a previous study. Antimicrobial use in the ICU was measured as defined daily dose (DDD) per 100 occupied bed-days. Trends and points of change were analysed with segmented regression. The intravenous macrolides/intravenous respiratory fluoroquinolones ratio in the ICU increased progressively, although not significantly, by 11.14% per quarter, likely related to prioritization of the use of macrolides in serious community-acquired pneumonia and the coronavirus disease 2019 pandemic. A remarkable upward trend of 2.5% per quarter was detected in the anti-methicillin-susceptible Staphylococcus aureus/anti-methicillin-resistant S. aureus agents ratio in the ICU, which could be explained by the low prevalence of methicillin-resistant S. aureus at the study centre. Patterns of amoxicillin-clavulanic acid/piperacillin-tazobactam ratio and diversification of anti-pseudomonal beta-lactams showed an increment in use over the study. The use of these novel indicators provides additional information for the current analysis of DDD.
