    Effects of a mesoporous bioactive glass on osteoblasts, osteoclasts and macrophages

    A mesoporous bioactive glass (MBG) of molar composition 75SiO2-20CaO-5P2O5 (MBG-75S) has been synthetized as a potential bioceramic for bone regeneration purposes. X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), nitrogen adsorption studies and transmission electron microscopy (TEM) demonstrated that MBG-75S possess a highly ordered mesoporous structure with high surface area and porosity, which would explain the high ionic exchange rate (mainly calcium and silicon soluble species) with the surrounded media. MBG-75S showed high biocompatibility in contact with Saos-2 osteoblast-like cells. Concentrations up to 1 mg/ml did not lead to significant alterations on either morphology or cell cycle. Regarding the effects on osteoclasts, MBG-75S allowed the differentiation of RAW264.7 macrophages into osteoclast-like cells but exhibiting a decreased resorptive activity. These results point out that MBG-75S does not inhibit osteoclastogenesis but reduces the osteoclast bone-resorbing capability. Finally, in vitro studies focused on the innate immune response, evidenced that MBG-75S allows the proliferation of macrophages without inducing their polarization towards the M1 pro-inflammatory phenotype. This in vitro behavior is indicative that MBG-75S would just induce the required innate immune response without further inflammatory complications under in vivo conditions. The overall behavior respect to osteoblasts, osteoclasts and macrophages, makes this MBG a very interesting candidate for bone grafting applications in osteoporotic patients

    SPITZER: Accretion in Low Mass Stars and Brown Dwarfs in the Lambda Orionis Cluster

    We present multi-wavelength optical and infrared photometry of 170 previously known low mass stars and brown dwarfs of the 5 Myr Collinder 69 cluster (Lambda Orionis). The new photometry supports cluster membership for most of them, with less than 15% of the previous candidates identified as probable non-members. The near infrared photometry allows us to identify stars with IR excesses, and we find that the Class II population is very large, around 25% for stars (in the spectral range M0 - M6.5) and 40% for brown dwarfs, down to 0.04 Msun, despite the fact that the H(alpha) equivalent width is low for a significant fraction of them. In addition, there are a number of substellar objects, classified as Class III, that have optically thin disks. The Class II members are distributed in an inhomogeneous way, lying preferentially in a filament running toward the south-east. The IR excesses for the Collinder 69 members range from pure Class II (flat or nearly flat spectra longward of 1 micron), to transition disks with no near-IR excess but excesses beginning within the IRAC wavelength range, to two stars with excess only detected at 24 micron.     Differential Immune-Reactivity and Subcellular Distribution Reveal the Multifunctional Character of Profilin in Pollen as Major Effect of Sequences Polymorphism

    Trabajo presentado al Annual Meeting of the American Society of Agronomy and Crop Science Society and Soil Science Society of America, celebrado en Cincinnati (USA) del 21 al 24 de octubre de 2012.Profilin, one of the major allergen (Ole e 2) of olive (Olea europaea L.) pollen, are broadly distributed actin-monomer-binding proteins (ABP). They display a major regulatory role in actin cytoskeleton dynamics, driving cell morphogenesis, sexual reproduction, and translating signals into cellular responses to different environmental stresses. Plants exhibit multiple profilin isoforms w ith distinctive biochemical properties, and differentially regulated. How ever, it is still an open question w hether these profilin isoforms, generated by multiple gene sequence polymorphism, are functionally different, as well as the role of that polymorphism in pollen allergy. Particularly, in differential epitopes generation, profilin isoforms sensitization and cross-reactivity among cultivars, and even among species. In the present study, w e have used mature pollen from olive, birch, hazel, timothy-grass, and maize, in addition to olive germinating pollen and seeds, w ith the aim to analyze the immune-reactivity and subcellular localization of profilin by using polyclonal and specific isoforms antibodies against olive and maize profilins. The results show ed immune-reactivity differences betw een the five species analyzed, betw een olive cultivars, as w ell as between reproductive and vegetative profilins. Furthermore, the existence of different profilin isoforms w as revealed along pollen germination stages. A differential subcellular distribution of profilin isoforms w as found in olive pollen. They w ere localized in the nucleus, pollen aperture regions, pollen and tube w alls and pollen tip, in addition to a general cytoplasmic distribution, in comparison to controls. Data suggest that profilin family might contain numerous functionally distinctive isoforms, spatial-temporal differentially expressed and regulated during vegetative development, pollen maturation and pollen tube grow th.     A Regenerative Endodontic Approach in Mature Ferret Teeth Using Rodent Preameloblast-conditioned Medium

    Background: This study evaluated the effectiveness of a regenerative endodontic approach to regenerate the pulp tissue in mature teeth of ferret. The presence of odontoblast-like cells in the newly-formed tissue of teeth treated with or without preameloblast-conditioned medium was evaluated based on morphological criteria. Materials and methods: Twenty-four canines from six ferrets were treated. The pulp was removed, and the apical foramen was enlarged. After inducing the formation of a blood clot, a collagen sponge with or without preameloblast-conditioned medium was placed underneath the cementoenamel junction. The samples were analyzed at the eighth week of follow-up. Results: Vascularized connective tissue was observed in 50% of teeth, without differences between groups. The tissue occupied the apical third of the root canals. Odontoblast-like cells were not observed in any group. Conclusion: Revitalization of mature teeth is possible, at least in the apical third of the root canal. Further experimental research is needed to produce more reliable outcomes

    Tuning surface interactions on MgFe2O4 nanoparticles to induce interfacial hyperactivation in Candida rugosa lipase immobilization

    Lipase adsorption on solid supports can be mediated by a precise balance of electrostatic and hydrophobic interactions. A suitable fine-tuning could allow the immobilized enzyme to display high catalytic activity. The objective of this work was to investigate how pH and ionic strength fluctuations affected protein-support interactions during immobilization via physical adsorption of a Candida rugosa lipase (CRL) on MgFe2O5. The highest amount of immobilized protein (IP) was measured at pH 4, and an ionic strength of 90 mM. However, these immobilization conditions did not register the highest hydrolytic activity (HA) in the biocatalyst (CRLa@MgFe2O4), finding the best values also at acidic pH but with a slight shift towards higher values of ionic strength around 110 mM. These findings were confirmed when the adsorption isotherms were examined under different immobilization conditions so that the maximum measurements of IP did not coincide with that of HA. Furthermore, when the recovered activity was examined, a strong interfacial hyperactivation of the lipase was detected towards acidic pH and highly charged surrounding environments. Spectroscopic studies, as well as in silico molecular docking analyses, revealed a considerable involvement of surface hydrophobic protein-carrier interactions, with aromatic aminoacids, especially phenylalanine residues, playing an important role. In light of these findings, this study significantly contributes to the body of knowledge and a better understanding of the factors that influence the lipase immobilization process on magnetic inorganic oxide nanoparticle surfaces.Fil: Morales, Andrés Hernán.     Temas clave en la formación de profesores en Chile desde la perspectiva de docentes y directivos

    Teacher education in Chile and the issues that intersect it are nowadays in the public debate of the Chilean society. This study aims at analyze key issues on teacher education- research, innovation, continuous education, university-school relationship, and management- from the perspective of academics, school-teachers and administrators. The study was conducted on 222 participants from Chilean universities and schools in the context of a non-experimental research design of cross-sectional and descriptive nature. The surveys and semi-structured interviews results show that it is necessary to strengthen the relationship between schools and universities, train teachers that achieve effective performances in the current socio-educational contexts and create the conditions for teachers to do research of their own pedagogical practices.La formación de profesores en Chile y los puntos que la intersectan, están hoy en día en el debate público de la sociedad chilena. El objetivo de este estudio es analizar temas clave de la formación de profesores- investigación, innovación, formación continua, relación universidad-establecimiento educacional, y gestión- desde la perspectiva de profesores universitarios, profesores del sistema escolar y directivos. Para esto, el estudio se realizó con una muestra de 222 participantes de establecimientos educacionales y universidades chilenas en el contexto de un diseño investigativo de carácter no experimental y de tipo transeccional descriptivo. Los resultados de las encuestas y entrevistas aplicadas revelan que es necesario fortalecer la relación establecimiento educacional y universidad, formar profesores que tengan un desempeño efectivo en los contextos socio-educativos actuales y crear las condiciones para que los profesores puedan realizar investigaciones de sus propias prácticas pedagógicas

    Versatile Graphene-Based Platform for Robust Nanobiohybrid Interfaces

    Technologically useful and robust graphene-based interfaces for devices require the introduction of highly selective, stable, and covalently bonded functionalities on the graphene surface, whilst essentially retaining the electronic properties of the pristine layer. This work demonstrates that highly controlled, ultrahigh vacuum covalent chemical functionalization of graphene sheets with a thiol-terminated molecule provides a robust and tunable platform for the development of hybrid nanostructures in different environments. We employ this facile strategy to covalently couple two representative systems of broad interest: metal nanoparticles, via S-metal bonds, and thiol-modified DNA aptamers, via disulfide bridges. Both systems, which have been characterized by a multi-technique approach, remain firmly anchored to the graphene surface even after several washing cycles. Atomic force microscopy images demonstrate that the conjugated aptamer retains the functionality required to recognize a target protein. This methodology opens a new route to the integration of high-quality graphene layers into diverse technological platforms, including plasmonics, optoelectronics, or biosensing.
