    Antibodies To Neospora Caninum In Sheep From Slaughterhouses In The State Of Sao Paulo, Brazil

    Neosporosis is an emergent disease responsible for considerable economic impact due to reproductive losses. Its zoonotic potential remains unknown. This study involved a survey of antibodies to Neospora caninum in slaughtered sheep and their association with epidemiological variables. Serum samples from 596 sheep from the states of Sao Paulo and Rio Grande do Sul, Brazil, were collected in two slaughterhouses located in Sao Paulo and evaluated by indirect fluorescence antibody test (IFAT), using cut-off titers of 25. Among these samples, 353/596 (59.23%; 95%CI 55.23-63.10) were positive and 263/353 (74.50%; 95%CI 69.71-78.77%) were from Rio Grande do Sul. Statistical associations were determined in the univariate analysis between the serological results and sex, breed and municipality of origin. Sheep that came from extensive breeding system showed higher chance (OR=2.09) of presenting antibodies to N. caninum in relation to those from semi-intensive system. Higher chance was also observed for the different studied breeds, except Bergamacia, in relation to Hampshire Down. The results revealed the presence of infection by N. caninum in sheep from slaughterhouses.2419510

    ABI3, a component of the WAVE2 complex, is potentially regulated by PI3K/AKT pathway

    We previously reported that ABI3 expression is lost in follicular thyroid carcinomas and its restoration significantly inhibited cell growth, invasiveness, migration, and reduced tumor growth in vivo. The mechanistic basis by which ABI3 exerts its tumor suppressive effects is not fully understood. In this study, we show that ABI3 is a phosphoprotein. Using proteomic array analysis, we showed that ABI3 modulated distinct cancer-related pathways in thyroid cancer cells. The KEA analysis found that PI3K substrates were enriched and forced expression of ABI3 markedly decreased the phosphorylation of AKT and the downstream-targeted protein pGSK3 beta. We next used immunoprecipitation combined with mass spectrometry to identify ABI3-interacting proteins that may be involved in modulating/integrating signaling pathways. We identified 37 ABI3 partners, including several components of the canonical WAVE regulatory complex (WRC) such as WAVE2/CYF1P1/NAP1, suggesting that ABI3 function might be regulated through WRC. Both, pharmacological inhibition of the PI3K/AKT pathway and mutation at residue S342 of ABI3, which is predicted to be phosphorylated by AKT, provided evidences that the non-phosphorylated form of ABI3 is preferentially present in the WRC protein complex.     A violação dos direitos fundamentais mostra-se cada vez mais recorrente no âmbito do Sistema Único de Saúde (SUS) ante às precárias condições de atendimento à população. A grande consequência disso é a mistanásia, a morte miserável, sem assistência à saúde e resultado da ausência de prestação do serviço de saúde ou da ineficiência deste. Neste sentido, o presente artigo tem por objetivo analisar a utilização da auditoria como forma de prevenção da mistanásia no âmbito do Sistema Único de Saúde (SUS), mediante a possibilidade de melhora na assistência e no acesso à saúde pública, uma vez que esta, enquanto política pública de Estado, pode diminuir o número de mortes miseráveis.  Para tanto, a presente pesquisa utilizou o método teórico, fundamentado em pesquisa e revisão bibliográfica de obras, artigos científicos, periódicos, legislação, doutrina e jurisprudência aplicáveis ao caso e pertinentes à propositura de soluções à problemática

    Evaluation of the monitoring network of a river basin of Rio Grande do Sul, Brazil

    The Sinos River is one of the most polluted rivers in Brazil. This article presents a survey of information about the hydrological monitoring network of Sinos River Basin, state of Rio Grande do Sul. It aimed to identify the institutional actions in progress to evaluate the water quality of the river. A documentary research was conducted through which the entities responsible for the hydrological monitoring of the Sinos River and its tributaries were identified. It was found that there are several responsible organisms and that the sampling frequencies of water quality are varied. The main entity responsible for this monitoring is the State Foundation for Environmental Protection, which performs the analysis of physical, chemical and biological parameters in quarterly frequency. The owners of the sanitation services verify the hourly, daily and monthly frequency of the adduced water and the effluent standards dumped in the river. Other entities, state and federal, perform the monitoring in fluviometric and rainfall stations. It was verified, then, that the river is highly monitored, but there isn’t integration of the data. The quality results are available with a three-month time lapse and the daily results are not disclosed by the sanitation companies to the public

    RESUMO: No referido artigo dedicou-se a análise da violência de gênero ao longo dos tempos na legislação nacional e internacional, onde se obteve o cuidado para circundar o tema com o marco teórico propondo um diálogo com os pensadores como Deleuze, Michel Foucault, Pierre Bourdieu e Judith Butler sob o enfoque da teoria do poder que impera nas relações humanas, sociais e jurídicas. Houve a preocupação em repensar o conceito de gênero, a partir da desconstrução dos seus elementos estruturais ligados a condição biológica, social e histórica. Além dessa desconstrução, outros conceitos foram trabalhados, como: sexo, sexualidade e identidade.  Nessa trajetória inicial enfatizou-se que os homens e as mulheres são cidadãos plurais e complexos, e em meio a isto está a violência que também é fruto dos conflitos que demandam desse espaço, carente de diálogo e compreensão. E por fim, como mecanismo de construção do texto utilizou-se o método hipotético-dedutivo, baseado em pesquisa bibliográfica

    Immunosensor for the Diagnostics of Autoimmune Hemolytic Anemia (AIHA) Based on Immobilization of a Monoclonal Antibody on a Layer of Silk Fibroin

    European Regional Development Fund (ERDF)The diagnostics of the autoimmune hemolytic anemia (AIHA), a rare disease caused by autoantibody-induced hemolysis, is still prone to false positives for it is based on visual observation in the so-called Direct Coombs test. In this study, we developed a specific IgG hemolysis immunosensor produced with layer-by-layer (LbL) films containing a monoclonal antibody against human immunoglobulin (mAbIMUG) deposited along with a layer of silk fibroin (SF) derived from Bombyx mori cocoons. Adsorption of mAbIMUG on a SF layer was confirmed by the fluorescence emission band at 326 nm. Immunosensors were prepared with LbL films deposited on interdigitated gold electrodes for impedance spectroscopy and on screen printed carbon electrodes for electrochemical measurements. When the SF/mAbIMUGLbL film was exposed to healthy red blood cells (RBCs), no cell binding was observed by the optical microscopy images. In addition, no major changes were observed in the signals of the square wave voltammogram and in the impedance spectra. In contrast, the electrochemical signal was significantly increased and the dielectric loss curve shifted for the sensing units containing RBCs with the antibody attached on the surface (“sick cells”). Furthermore, cell attachment was so strong that optical images still showed covered electrodes even after washing in PBS buffer. The detection with two distinct methods seems promising for an effective diagnosis of AIHA

    The quality of fish is affected by various factors such as, source species, freshness, and storage conditions. In free markets, exposure and handling of fish are intense, and often it is not known the origin and date of capture. The aim this work was study physicochemical, microbiological and sensory parameters to determine the shelf life of Yellow Hake sold in street markets in the city of Santos-SP. Samples were acquired in street markets from different districts of the city and stored in a domestic refrigerator for 72 hours. And it was found that the Yellow Hake acquired on open street markets in the city of Santos-SP should be for immediate consumption. The changes are noticeable deterioration after 24 hours of storage. The results reinforce the importance of the implementation of quality programs in all sectors of sale to ensure food safety for consumers.10217418

    AGK-BRAF gene fusion is a recurrent event in sporadic pediatric thyroid carcinoma

    Thyroid cancer is the fastest increasing cancer worldwide in all age groups. Papillary thyroid carcinoma (PTC) is the most common type of thyroid cancer in both adults and children. PTC genomic landscape has been extensively studied in adults, but information regarding sporadic pediatric patients is lacking. Although BRAF V600E mutation is highly prevalent in adults, this mutation is uncommon in pediatric cases. As adult and pediatric PTC is a mitogen-activated protein kinase-driven cancer, this altered pathway might be activated by different genetic events. The aim of this study was to investigate the occurrence of AGK-BRAF fusion gene, recently described in radiation-exposed pediatric PTC, in a cohort of exclusively sporadic pediatric PTC. The series consisted of 30 pediatric PTC younger than 18 years of age at the time of diagnosis and 15 matched lymph node metastases (LNM). Primary tumors and matched LNM were screened for the presence of the AGK-BRAF fusion transcript by RT-PCR. To confirm the identity of the amplified products, randomly selected samples positive for the presence of the fusion transcripts were sequenced. Moreover, BRAF dual-color, break-apart probes confirmed BRAF rearrangement. Overall, the AGK-BRAF fusion gene was detected in 10% (3/30) of primary tumors. For one of these cases, paired LNM was also available, which also shows the presence of AGK-BRAF fusion gene. This study described, for the first time, the presence of AGK-BRAF in sporadic pediatric PTC.     Evaluation of antimicrobial and antitumoral activity of Garcinia mangostana L. (mangosteen) grown in Southeast Brazil

    PURPOSE:To characterize the anatomy of the fruit and leaf and the presence of phytocompounds. To evaluate the antitumor and antimicrobial activity of ethanolic extract of Garcinia mangostana L. (mangosteen) cultivated in southeastern Brazil.METHODS:Anatomical characterization and histochemical reactions were performed for structural identification and the presence of phytocompounds. Preparation of ethanolic extract of the fruit, leaf and resin of mangosteen. Culture B16-F10 melanoma cells for treatment with mangosteen ethanolic extract to determine cell viability by MTT and genotoxic effect by comet assay. Evaluation by antimicrobial activity against Staphylococcus aureus and Escherichia coli by agar diffusion test and by determination of Minimum Inhibitory Concentration (MIC).RESULTS:Our results showed many secretory canals in resin fruit and leaf; identifying lipids, starch, lignin and phenolic compounds. The leaf extract induced genotoxicity and apoptosis in B16-F10 cells, since the fragmentation of DNA in the comet assay.
