    A conserved domain targets exported PHISTb family proteins to the periphery of Plasmodium infected erythrocytes

    During blood-stage infection, malaria parasites export numerous proteins to the host erythrocyte. The Poly-Helical Interspersed Sub-Telomeric (PHIST) proteins are an exported family that share a common ‘PRESAN’ domain, and include numerous members in Plasmodium falciparum, Plasmodium vivax and Plasmodium knowlesi. In P. falciparum, PHIST proteins have been implicated in protein trafficking and intercellular communication. A number of PHIST proteins are essential for parasite survival. Here, we identify nine members of the PHISTb sub-class of PHIST proteins, including one protein known to be essential for parasite survival, that localise to the erythrocyte periphery. These proteins have solubility characteristics consistent with their association with the erythrocyte cytoskeleton. Together, an extended PRESAN domain, comprising the PRESAN domain and preceding sequence, form a novel targeting-domain that is sufficient to localise a protein to the erythrocyte periphery. We validate the role of this domain in RESA, thus identifying a cytoskeleton-binding domain in RESA that functions independently of its known spectrin-binding domain. Our data suggest that some PHISTb proteins may act as cross-linkers of the erythrocyte cytoskeleton. We also show for the first time that peripherally-localised PHISTb proteins are encoded in genomes of P. knowlesi and vivax indicating a conserved role for the extended PRESAN domain of these proteins in targeting to the erythrocyte peripher

    Pulsar Wind Nebulae with Bow Shocks: Non-thermal Radiation and Cosmic Ray Leptons

    Pulsars with high spin-down power produce relativistic winds radiating a non-negligible fraction of this power over the whole electromagnetic range from radio to gamma-rays in the pulsar wind nebulae (PWNe). The rest of the power is dissipated in the interactions of the PWNe with the ambient interstellar medium (ISM). Some of the PWNe are moving relative to the ambient ISM with supersonic speeds producing bow shocks. In this case, the ultrarelativistic particles accelerated at the termination surface of the pulsar wind may undergo reacceleration in the converging flow system formed by the plasma outflowing from the wind termination shock and the plasma inflowing from the bow shock. The presence of magnetic perturbations in the flow, produced by instabilities induced by the accelerated particles themselves, is essential for the process to work. A generic outcome of this type of reacceleration is the creation of particle distributions with very hard spectra, such as are indeed required to explain the observed spectra of synchrotron radiation with photon indices Γâ‰Č 1.5. The presence of this hard spectral component is specific to PWNe with bow shocks (BSPWNe). The accelerated particles, mainly electrons and positrons, may end up containing a substantial fraction of the shock ram pressure. In addition, for typical ISM and pulsar parameters, the e+ released by these systems in the Galaxy are numerous enough to contribute a substantial fraction of the positrons detected as cosmic ray (CR) particles above few tens of GeV and up to several hundred GeV. The escape of ultrarelativistic particles from a BSPWN—and hence, its appearance in the far-UV and X-ray bands—is determined by the relative directions of the interstellar magnetic field, the velocity of the astrosphere and the pulsar rotation axis. In this respect we review the observed appearance and multiwavelength spectra of three different types of BSPWNe: PSR J0437-4715, the Guitar and Lighthouse nebulae, and Vela-like objects. We argue that high resolution imaging of such objects provides unique information both on pulsar winds and on the ISM. We discuss the interpretation of imaging observations in the context of the model outlined above and estimate the BSPWN contribution to the positron flux observed at the Earth

    Factors Associated with Revision Surgery after Internal Fixation of Hip Fractures

    Background: Femoral neck fractures are associated with high rates of revision surgery after management with internal fixation. Using data from the Fixation using Alternative Implants for the Treatment of Hip fractures (FAITH) trial evaluating methods of internal fixation in patients with femoral neck fractures, we investigated associations between baseline and surgical factors and the need for revision surgery to promote healing, relieve pain, treat infection or improve function over 24 months postsurgery. Additionally, we investigated factors associated with (1) hardware removal and (2) implant exchange from cancellous screws (CS) or sliding hip screw (SHS) to total hip arthroplasty, hemiarthroplasty, or another internal fixation device. Methods: We identified 15 potential factors a priori that may be associated with revision surgery, 7 with hardware removal, and 14 with implant exchange. We used multivariable Cox proportional hazards analyses in our investigation.     Blood chemistry and haematological parameters have been determined in the Antarctic teleosts, Notothenia neglecta Nybelin and Notothenia rossii Richardson at 2° C. Samples were taken using chronically implanted dorsal aortic cannulae following a minimum of 24–36 h recovery. Broadly similar results were obtained for the two species. In N. neglecta, routinely active specimens had high values of arterial pH (7.81) and PO2 (9.26 kPa), and modest haemoglobin levels (5.6 g dl−1) relative to temperate species. Following 3 min strenuous activity there was a decrease in arterial pH (7.63) and a small rise in lactate from 0.41 to 0.68 mm, but no significant change in the calculated net metabolic acid load (ήHm+). PaO2 and PaCO2 varied inversely during exercise, and oxygen content declined by 22%. pHa and most other haematological parameters returned to routine values between 1–3 h post‐exercise. The results suggest that the major effect of strenuous activity in Notothenia spp. is a respiratory, rather than a metabolic acidosis
