    Characterisation of resistance mechanisms developed by basal cell carcinoma cells in response to repeated cycles of photodynamic therapy

    photodynamic therapy (pDt) with methyl-aminolevulinate acid (MAL-pDt) is being used for the treatment of Basal cell carcinoma (BCC), but recurrences have been reported. In this work, we have evaluated resistance mechanisms to MAL-pDt developed by three BCC cell lines (AsZ, BsZ and CsZ), derived from mice on a ptch+/− background and with or without p53 expression, subjected to 10 cycles of PDT (10thG). the resistant populations showed mesenchymal-like structure and diminished proliferative capacity and size compared to the parental (p) cells. the resistance was dependent on the production of the endogenous photosensitiser protoporphyrin IX in the CsZ cell line and on its cellular localisation in AsZ and BsZ cells. Moreover, resistant cells expressing the p53 gene presented lower proliferation rate and increased expression levels of N-cadherin and Gsk3β (a component of the Wnt/β-catenin pathway) than P cells. In contrast, 10thG cells lacking the p53 gene showed lower levels of expression of Gsk3β in the cytoplasm and of e-cadherin and β-catenin in the membrane. In addition, resistant cells presented higher tumorigenic ability in immunosuppressed mice. Altogether, these results shed light on resistance mechanisms of BCC to pDt and may help to improve the use of this therapeutic approac

    En esta obra se recogen las aportaciones de diversos autores, que en algunos casos son profesionales dedicados a la actividad de Protección Civil o, en otros, son profesionales del mundo académico, con conocimientos especializados en este campo, y cuyas aportaciones, en conjunto, dan una visión panorámica respecto a la configuración actual del Sistema Nacional de Protección Civil, y resuelven la cuestión de cómo las competencias autonómicas se insertan en el mismo, con tratamiento particularizado del contenido normativo vigente en la Comunidad Autónoma Valenciana. Se hace referencia también, a cómo el Sistema Nacional de Protección Civil se encuadra en el Mecanismo Europeo de Protección Civil, que supone un nivel competencial más, pero organizado en base a los mismos principios de coordinación, cooperación y solidaridad de los territorios, y que al insertarse aquel como sistema integrado en el plano europeo, respeta completamente la distribución competencial, sin que esta participación en la dimensión competencias autonómicas.Esta edición se ha financiado mediante la concesión de la subvención de la Generalitat Valenciana, a través de la Conselleria de Governació i Justícia, aprobada por Resolución de 12 de marzo de 2015, con la cual se financiaron también las Jornadas sobre el desarrollo de las competencias de Protección Civil en la Generalitat Valenciana

    Prevalence, features and predictive factors of liver nodules in Fontan surgery patients: The VALDIG Fonliver prospective cohort

    Background & Aims: Fontan surgery is used to treat a variety of congenital heart malformations, and may lead to advanced chronic liver disease in the long-term. This study examines the prevalence, characteristics and predictors of liver nodules in patients following Fontan surgery. Methods: This was a prospective, cross-sectional, observational study conducted at 8 European centres. Consecutive patients who had undergone Fontan surgery underwent blood tests, abdominal ultrasonography (US), transient elastography (Fibroscan®), echocardiography, haemodynamic assessments, and abdominal MRI/CT scan. The primary outcome measure was liver nodules detected in the MRI/CT scan. Predictors of liver nodules were identified by multivariate logistic regression. Results: One hundred and fifty-two patients were enrolled (mean age 27.3 years). The mean time elapsed from surgery to inclusion was 18.3 years. Liver nodule prevalences were 29.6% (95% CI 23–37%) on US and 47.7% (95% CI 39–56%) on MRI/CT. Nodules were usually hyperechoic (76.5%), round-shaped (>80%), hyperenhancing in the arterial phase (92%) and located in the liver periphery (75%). The sensitivity and specificity of US were 50% (95% CI 38–62%) and 85.3% (95% CI 75–92%), respectively. Inter-imaging test agreement was low (adjusted kappa: 0.34). In the multivariate analysis, time since surgery >10 years was the single independent predictor of liver nodules (odds ratio 4.18; p = 0.040). Hepatocellular carcinoma was histologically diagnosed in 2 of the 8 patients with hypervascular liver nodules displaying washout. Conclusion: While liver nodules are frequent in Fontan patients, they may go unnoticed in US. Liver nodules are usually hyperechoic, hypervascular and predominantly peripheral. This population is at risk of hepatocellular carcinoma, the diagnosis of which requires confirmatory biopsy

    Microbial responses to perfluoroalkyl substances and perfluorooctanesulfonate (PFOS) desulfurization in the Antarctic marine environment

    This research is part of POLARCSIC activities.-- 12 pages, 8 figures, supplementary data https://doi.org/10.1016/j.watres.2019.115434Perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA) acids are ubiquitous in the oceans, including remote regions, and are toxic to fish and mammals. The impact to the lowest trophic levels of the food web, however, remains unknown. We challenged natural bacterial communities inhabiting Antarctic coastal waters (Deception Island) with PFOS and PFOA concentrations ranging from 2 ng/L to 600 ng/L that selected for tolerant taxa. After 48 h, concentrations of PFOS decreased by more than 50% and sulfur metabolism-related transcripts were significantly enriched in the treatments suggesting desulfurization of PFOS. Conversely, no significant differences were found between initial and final PFOA concentrations. Gammaproteobacteria and Roseobacter, two abundant groups of marine bacteria, increased their relative activity after 24 h of incubation, whereas Flavobacteriia became the main contributor in the treatments after 6 days. Community activities (extracellular enzyme activity and absolute number of transcripts) were higher in the treatments than in the controls, while bacterial abundances were lower in the treatments, suggesting a selection of PFOS and PFOA tolerant community in the exposed treatments. Our results show a direct effect of PFOS and PFOA exposure on the composition and functionality of natural Antarctic marine microbial communities.
