    Tumor necrosis factor-alpha inhibition reduces CXCL-8 levels but fails to prevent fibrin generation and does not improve outcome in a rabbit model of endotoxic shock

    The effects of a monoclonal antibody (mAb) to tumor necrosis factor-alpha (TNF-alpha) were examined in a rabbit model of endotoxic shock. Intravenous administration of lipopolysaccharide (100 microg/kg/hr) for 6 hours (n = 11) increased TNF-alpha levels. Fibrinogen was partially consumed, and fibrin deposits were seen in kidney and lungs at 24 hours. Mortality at 24 hours was 64%. Levels of interleukin-8 (aka CXCL-8) were notably increased. Mean arterial pressure (MAP) and leukocyte counts decreased, whereas creatinine levels were enhanced. The anti-TNF-alpha mAb (20 mg/kg i.v. bolus + 5 mg/kg/h i.v. for the first 90 minutes) (n = 10) efficiently inhibited the TNF-activity. Rabbits exhibited lower CXCL-8 levels; MAP improved, the decrease in leukocyte counts was partially prevented and creatinine levels were lower, but fibrinogen, fibrin deposits in kidneys and lungs and mortality, 55%, were similar to the LPS group. Rabbits that did not survive exhibited lower fibrinogen levels, more fibrin in kidneys and lungs and higher CXCL-8 and creatinine levels than survivors, while there were no differences in TNF-alpha, MAP and leukocytes. Thus, the inhibition of TNF-alpha, although beneficial through lowering CXCL-8 levels, is not enough to improve the outcome, which could be partly due to the inability to prevent the fibrin deposits formation in kidneys and lungs

    VEGF121b and VEGF165b are weakly angiogenic isoforms of VEGF-A

    <p>Abstract</p> <p>Background</p> <p>Different isoforms of VEGF-A (mainly VEGF<sub>121</sub>, VEGF<sub>165 </sub>and VEGF<sub>189</sub>) have been shown to display particular angiogenic properties in the generation of a functional tumor vasculature. Recently, a novel class of VEGF-A isoforms, designated as VEGF<sub>xxx</sub>b, generated through alternative splicing, have been described. Previous studies have suggested that these isoforms may inhibit angiogenesis. In the present work we have produced recombinant VEGF<sub>121/165</sub>b proteins in the yeast <it>Pichia pastoris </it>and constructed vectors to overexpress these isoforms and assess their angiogenic potential.</p> <p>Results</p> <p>Recombinant VEGF<sub>121/165</sub>b proteins generated either in yeasts or mammalian cells activated VEGFR2 and its downstream effector ERK1/2, although to a lesser extent than VEGF<sub>165</sub>. Furthermore, treatment of endothelial cells with VEGF<sub>121/165</sub>b increased cell proliferation compared to untreated cells, although such stimulation was lower than that induced by VEGF<sub>165</sub>. Moreover, <it>in vivo </it>angiogenesis assays confirmed angiogenesis stimulation by VEGF<sub>121/165</sub>b isoforms. A549 and PC-3 cells overexpressing VEGF<sub>121</sub>b or VEGF<sub>165</sub>b (or carrying the PCDNA3.1 empty vector, as control) and xenotransplanted into nude mice showed increased tumor volume and angiogenesis compared to controls. To assess whether the VEGF<sub>xxx</sub>b isoforms are differentially expressed in tumors compared to healthy tissues, immunohistochemical analysis was conducted on a breast cancer tissue microarray. A significant increase (p < 0.05) in both VEGF<sub>xxx</sub>b and total VEGF-A protein expression in infiltrating ductal carcinomas compared to normal breasts was observed. A positive significant correlation (r = 0.404, p = 0.033) between VEGF<sub>xxx</sub>b and total VEGF-A was found.</p> <p>Conclusions</p> <p>Our results demonstrate that VEGF<sub>121/165</sub>b are not anti-angiogenic, but weakly angiogenic isoforms of VEGF-A. In addition, VEGF<sub>xxx</sub>b isoforms are up-regulated in breast cancer in comparison with non malignant breast tissues. These results are to be taken into account when considering a possible use of VEGF<sub>121/165</sub>b-based therapies in patients.</p

    Vertical zonation of bacterial assemblages attributed to physical stratification during the summer relaxation of the coastal upwelling off Galicia (NW Spain)

    Research articleWe combined flow cytometry, CARD-FISH, and 16S rRNA gene tag pyrosequencing to investigate bacterioplankton dynamics along a transect in shelf waters off A Coruña (Galicia, NW Spain). Over five days (16-20th July 2012) we sampled during the relaxation of a summer upwelling pulse, providing an opportunity to examine the impact of pulses of cold nutrient-rich water into coastal microbial communities. The hydrographic conditions, characterized by intense density stratification of surface waters and the presence of a deep chlorophyll maximum (DCM) at 20–30 m, were relatively maintained over the sampling period. Indeed, bacterial abundance and composition displayed low day to day variation. Alpha diversity analysis suggested that species richness and diversity increased from coastal to shelf stations and from the surface down to the coastal DCM, which could be caused by the mixing of upwelled bacteria with the coastal surface waters. SAR11, SAR86, and Roseobacter were the most abundant bacteria detected in the samples by using CARD-FISH. The assemblages observed by pyrosequencing displayed a strong vertical zonation along the transect. Rhodobacteraceae (under class Alphaproteobacteria) and Bacteriodetes dominated the surface waters and decreased during the upwelling pulse, while SAR 86 (under class Gammaproteobacteria), Actinobacteria and SAR11 clade increased their relative abundance at the coastal DCM with upwelling relaxation, particularly at the shelf stations. Bacterial assemblages from surface waters were associated with higher temperature and light conditions, while coastal DCM assemblages were rather associated to salinity, inorganic nutrients and a diatom-bloom leading to high chlorophyll-a. Our findings suggest that the vertical variability in environmental conditions induced by the intense density stratification, the exportation of warmer and less saline surface water from the rias to the adjacent shelf, and the fertilizing effect of recently upwelled water at the deeper layer, determined the composition of distinct bacterial assemblages at the subsurface and DCM layers.XUNTA DE GALICIA (O9MMA 027604PR, 10MMA604024PR), IEO (PROYECTO RADIALES) y PLAN NACIONAL (MODUPLAN CTM2011-24008)Versión del editor2,24

    Lung Transplant Improves Survival and Quality of Life Regardless of Telomere Dysfunction

    Introduction: Fibrotic interstitial lung diseases (ILDs) are the first indication for lung transplantation (LT). Telomere dysfunction has been associated with poor post-transplant outcomes. The aim of the study was to evaluate the morbi-mortality and quality of life in fibrotic ILDs after lung transplant depending on telomere biology. Methods: Fibrotic ILD patients that underwent lung transplant were allocated to two arms; with or without telomere dysfunction at diagnosis based on the telomere length and telomerase related gene mutations revealed by whole-exome sequencing. Post-transplant evaluation included: (1) short and long-term mortality and complications and (2) quality of life. Results: Fifty-five percent of patients that underwent LT carried rare coding mutations in telomerase-related genes. Patients with telomere shortening more frequently needed extracorporeal circulation and presented a higher rate of early post-transplant hematological complications, longer stay in the intensive care unit (ICU), and a higher number of long-term hospital admissions. However, post-transplant 1-year survival was higher than 80% regardless of telomere dysfunction, with improvement in the quality of life and oxygen therapy withdrawal. Conclusions: Post-transplant morbidity is higher in patients with telomere dysfunction and differs according to elapsed time from transplantation. However, lung transplant improves survival and quality of life and the associated complications are manageable

    Library of Seleno-Compounds as Novel Agents against Leishmania Species

    The in vitro leishmanicidal activities of a series of 48 recently synthesized selenium derivatives against Leishmania infantum and Leishmania braziliensis parasites were tested using promastigotes and intracellular amastigote forms. The cytotoxicity of the tested compounds for J774.2 macrophage cells was also measured in order to establish their selectivity. Six of the tested compounds (compounds 8, 10, 11, 15, 45, and 48) showed selectivity indexes higher than those of the reference drug, meglumine antimonate (Glucantime), for both Leishmania species; in the case of L. braziliensis, compound 20 was also remarkably selective. Moreover, data on infection rates and amastigote numbers per macrophage showed that compounds 8, 10, 11, 15, 45, and 48 were the most active against both Leishmania species studied. The observed changes in the excretion product profile of parasites treated with these six compounds were also consistent with substantial cytoplasmic alterations. On the other hand, the most active compounds were potent inhibitors of Fe superoxide dismutase (Fe-SOD) in the two parasite species considered, whereas their impact on human CuZn-SOD was low. The high activity, low toxicity, stability, low cost of the starting materials, and straightforward synthesis make these compounds appropriate molecules for the development of affordable antileishmanicidal agents

    Structural analysis of MDM2 RING separates degradation from regulation of p53 transcription activity

    MDM2–MDMX complexes bind the p53 tumor-suppressor protein, inhibiting p53's transcriptional activity and targeting p53 for proteasomal degradation. Inhibitors that disrupt binding between p53 and MDM2 efficiently activate a p53 response, but their use in the treatment of cancers that retain wild-type p53 may be limited by on-target toxicities due to p53 activation in normal tissue. Guided by a novel crystal structure of the MDM2–MDMX–E2(UbcH5B)–ubiquitin complex, we designed MDM2 mutants that prevent E2–ubiquitin binding without altering the RING-domain structure. These mutants lack MDM2's E3 activity but retain the ability to limit p53′s transcriptional activity and allow cell proliferation. Cells expressing these mutants respond more quickly to cellular stress than cells expressing wild-type MDM2, but basal p53 control is maintained. Targeting the MDM2 E3-ligase activity could therefore widen the therapeutic window of p53 activation in tumors

    Tracking bluefin tuna cohorts from east Atlantic Spanish fisheries since the 1980s

    Relative abundance and mean size at age analyses were performed for the catches of three Atlantic Spanish fisheries, in search of an exceptionally abundant cohort in the recent history of the eastern Atlantic and Mediterranean bluefin tuna stock. Length distributions of baitboat fisheries, both in the Bay of Biscay and areas close to the Strait of Gibraltar as well as trap catches from the Spanish Atlantic coast were converted to age distributions using age length keys from calcified structures. Results do validate indirectly the ageing method. The 1994 cohort was clearly tracked for juveniles and young adults, 3 to 7 years old, in baitboat catches, and also for older specimens, 9 to 11 years old, in the trap fisheries. Based on these results, it is suggested that the juvenile and young adults fisheries in the western part of the Iberian Peninsula are interconnected between one another and with the Balearic Islands area of the western Mediterranean, as it is the nearest spawning ground described and because it is also supported by tagging and other studies.Afin de trouver une cohorte exceptionnellement abondante dans la récente histoire du stock de ton rouge de l’Atlantique Est et de la Méditerranée, on a procédé à une analyse de l’abondance relative et de la taille moyenne par âge des captures réalisées par trois pêcheries espagnoles de l’Atlantique. Les distributions par tailles de deux pêcheries de canneurs dans le Golfe de Gascogne et dans les eaux proches du Détroit de Gibraltar, ainsi que les prises des madragues de la côte atlantique espagnole, ont été converties en âges à l’aide des clefs taille-âge obtenues à partir de la lecture de structures calcifiées. Les résultats valident indirectement la méthode de détermination de l’âge. La cohorte de 1994 affiche une forte indication présente chez les juvéniles et les jeunes adultes, de 3 à 7 ans, capturés dans les pêcheries de canneurs ainsi que chez les spécimens d’âge plus avancé, de 9 à 11 ans, pêchés à la madrague. Ces résultats suggèrent que les pêcheries de juvéniles et de jeunes adultes de la partie occidentale de la péninsule ibérique sont interconnectées entre elles ainsi qu’avec la zone des îles Baléares, en Méditerranée occidentale, étant donné qu’il s’agit de la zone de frai la plus proche et qu’il existe des indices de cette relation d’après les études de marquage et d’autres études.Para encontrar una cohorte excepcionalmente abundante en la historia reciente del stock de atún rojo del Atlántico este y Mediterráneo, se realizó un análisis de la abundancia relativa y la talla media por edad de las capturas de tres pesquerías atlánticas españolas. Las distribuciones de tallas de dos pesquerías de cebo vivo, en el Golfo de Vizcaya y en aguas próximas al Estrecho de Gibraltar, así como las capturas de las almadrabas de la costa atlántica española, fueron convertidas a edades mediante el uso de claves talla edad, obtenidas a partir de la lectura de piezas esqueléticas. Los resultados validan indirectamente el método de determinación de la edad. La cohorte de 1994 presenta una fuerte señal que pudo ser encontrada en los juveniles y adultos jóvenes, 3 a 7 años, capturados en las pesquerías de cebo vivo y también en especimenes mayores, 9 a 11 años, pescados con las almadrabas. Con esto resultados se sugiere que las pesquerías de juveniles y adultos jóvenes de la parte occidental de la península Ibérica están interconectadas entre ellas y con el área de las Islas Baleares, en el Mediterráneo occidental, puesto que es la zona de puesta más próxima y porque también hay evidencias de esta relación mediante el marcado y otros estudios

    Molecular characterization of multidrug resistant Enterobacterales strains isolated from liver and kidney transplant recipients in Spain

    The objective of this study was to analyse the mechanisms of resistance to carbapenems and other extended-spectrum-?-lactams and to determine the genetic relatedness of multidrug-resistant Enterobacterales (MDR-E) causing colonization or infection in solid-organ transplantation (SOT) recipients. Prospective cohort study in kidney (n= 142), liver (n= 98) or kidney/pancreas (n= 7) transplant recipients between 2014 and 2018 in seven Spanish hospitals. We included 531 MDR-E isolates from rectal swabs obtained before transplantation and weekly for 4?6 weeks after the procedure and 10 MDR-E from clinical samples related to an infection. Overall, 46.2% Escherichia coli, 35.3% Klebsiella pneumoniae, 6.5% Enterobacter cloacae, 6.3% Citrobacter freundii and 5.7% other species were isolated. The number of patients with MDR-E colonization post-transplantation (176; 71.3%) was 2.5-fold the number of patients colonized pre-transplantation (71; 28.7%). Extended spectrum ?-lactamases (ESBLs) and carbapenemases were detected in 78.0% and 21.1% of MDR-E isolates respectively. In nine of the 247 (3.6%) transplant patients, the microorganism causing an infection was the same strain previously cultured from surveillance rectal swabs. In our study we have observed a low rate of MDR-E infection in colonized patients 4?6 weeks post-transplantation. E. coli producing blaCTX-M-G1 and K. pneumoniae harbouring blaOXA-48 alone or with blaCTX-M-G1 were the most prevalent MDR-E colonization strains in SOT recipients.Acknowledgements The authors thank Mª Jesús Lecea and Laura Álvarez for technical assistance.
