Trabalho de conclusão de curso (graduação)—Universidade de Brasília, 2016.Este trabalho visa estudar a regência do verbo fazer, com semântica de causativo, em contextos de complementação com oração desenvolvida, iniciada pela conjunção que e precedida pela preposição com (“fazer com que”), e sua enorme aceitabilidade pela população com maior ou menor acesso aos letramentos provenientes do meio escolar. Essa dúvida quanto à forma preferível por parte dos falantes surgiu com minha experiência de revisora, em que, independentemente da escolaridade do leitor, sempre que eu revisava um texto e retirava a preposição “com” ambiente supracitado, as pessoas achavam que estava faltando alguma coisa e pediam para que eu deixasse do jeito “errado” mesmo

Metabolic drivers of IC-BEVS productivity: Tackling the production of enveloped viral particles

The Insect Cell-Baculovirus Expression System (IC-BEVS) has a major track record for the production of recombinant proteins and vaccines. Although its widespread use, the physiological aspects that contribute to systems productivity are still to be fully disclosed. In the present work, the metabolic features of the two main insect host cell lines, Sf9 and High Five, were analyzed during cellular growth and after baculovirus infection for the production of enveloped Influenza VLPs (Inf-VLPs). The gathered data were contextualized in a metabolic network representative of central carbon and nitrogen metabolism. Metabolic Flux Analysis (MFA) was performed to have a quantitative overview of the cellular fluxome dynamics that followed infection. In addition, the main carbon sources that contributed most to flux activity were identified. The impact of baculovirus infection on the physiology of High Five and Sf9 host cell lines was assessed by metabolomics, aiming at the identification of metabolic markers of productivity. The information herein generated was used to design tailored supplementation schemes that could boost IC-BEVS production yields of two enveloped viral particles: influenza VLPs (Inf-VLP) as a vaccine candidate and the recombinant baculovirus (BV). The strong correlation observed between the metabolic state of the host cell and baculovirus infection highlights the capacity of this virus to act as a metabolic engineer, re-directing the cellular fluxome to support virus replication and production. The results also show that the viral load influence the cellular responsiveness to the supplements, with lower MOIs retrieving higher improvements in specific productivity. The careful selection of the MOI, along with the supplementation of culture medium with compounds altering cellular redox state and cholesterol metabolism, yielded a 6-fold improvement of specific productivity. These results pave the way to deepen our knowledge on the relationship between host cell and virus, contributing to the disclosure of the metabolic determinants that contribute to productivity

Normalização contabilística em Portugal: algumas implicações para os TOC decorrentes da transição do POC para o SNC

O estudo em apreço pretende analisar algumas das implicações para os Técnicos Oficiais de Contas decorrentes das recentes alterações à normalização contabilística em Portugal. Os resultados obtidos através do estudo empírico revelam, essencialmente, que os Técnicos Oficiais de Contas sentiram uma grande mudança com a entrada em vigor do Sistema de Normalização Contabilística, que necessitaram de assistir a muitas mais horas de formação do que em anos anteriores, que os conceitos se alteraram, a forma de pensar se alterou, que as formações ministradas pela Ordem dos Técnicos Oficiais de Contas são de qualidade, que as áreas temáticas onde se sentem mais dificuldades são na aplicação do justo valor e nos testes de imparidade, que o Sistema de Normalização Contabilística permitirá maior liberdade de pensamento e que o Técnico Oficial de Contas deve ser o parceiro privilegiado do empresário. Contudo, os profissionais da contabilidade demonstram uma capacidade de adaptação positiva, pois na sua maioria sentiram poucas dificuldades na transição do Plano Oficial de Contabilidade para o Sistema de Normalização Contabilística

Which cognitive and perceptual skills best discriminate elite female handball players

The purpose of this study was to identify which cognitive and perceptual skills best discriminate elite female handball players according to the age group to which they belong to. The sample consisted of 73 Portuguese national teams’ female handball players, aged between 11 and 29 years, with an average of 7.5 (± 3.7) years of practice. The following tests were applied: Thurstone Identical Figures Test, Toulouse-Piéron Concentrated Attention Test, and the Nideffer Attentional and Interpersonal Style Inventory to evaluate perceptual skills, the polyreactiometer for Windows – PRWin to evaluate information processing, and an evaluation protocol for precision in anticipation using the temporal occlusion paradigm. The interpretation of the obtained discriminant function was based on the structure of the coefficients greater than |0.30|. The Seniors’ age group is discriminated from that of Talents by their better reaction, perception, and attention times, namely, by the ability to integrate several stimuli simultaneously and by a better capacity to anticipate future events more accurately. Psicoterapia em tempos de pandemia: a transferência em um caso de violência doméstica com criança

O presente relato de experiência tem como objetivo trazer uma discussão sobre uma experiência de atendimento psicoterapêutico ocorrida durante o contexto da pandemia de Covid-19 e toma como eixo norteador o conceito de transferência. Apresentam-se os efeitos da formação ocorrida a partir de um projeto de extensão voltado ao trabalho clínico com crianças e adolescentes envolvidos em situações de violência doméstica.

A click chemistry strategy to specifically monitor and improve purification of Influenza virus-like particles

Virus-like particles (VLPs) constitute a promising platform in vaccine development and targeted drug-delivery. However, most applications use simple, non-enveloped VLPs that present less technical challenges, not only to produce and purify, but also in terms of characterization, compared to enveloped VLPs. Recent advances in upstream processing, new product quality requirements and other regulatory issues, as well as the search for more cost-effective processes, led to the need to develop more efficient downstream processes for biopharmaceuticals.[1] In that sense, new monitoring and product characterization methods, which can be applied at all stages of downstream processing, are needed. Here is reported a valuable platform for the downstream processing and monitoring of the in vivo production of site-specifically functionalized enveloped Influenza VLPs. This strategy involves a two-step procedure that consists of residue-specific replacement of methionine by an analog (azidohomolanine) that enabled for post-expression functionalization with a fluorophore.[2] Importantly, this platform does not impact VLP production or purification processes, and allows functionalization without deleterious effect on hemagglutinin biological function. As a proof of concept a complete downstream processing was performed, including clarification, capture and polishing steps. A flow cytometry analysis (FACS) step was added to achieve a refined discrimination and separation between VLPs and baculovirus - the major impurity of the process.[3] This was further confirmed using atomic force microscopy (AFM). This tool allowed to accurately monitor our product, achieve higher product recovery yields and higher impurity removal levels. The versatile system presented here is broadly applicable to the production of functionalized enveloped VLPs, for vaccine design, targeted drug delivery and molecular imaging. A LINGUAGEM ARTÍSTICA DO ARTESANATO: ARTESANATO COMO PEÇA AUTORAL; COM CONCEITO, SIMBOLISMO E MATERIAIS CULTURAIS

The research proposes a sensitive look at the artistic language of crafts as an authorial piece; with concept, symbolism, and regional cultural materials. The guiding problem is how to add value to handicrafts, not limited to the product's functional aspects. The methodology used, through bibliographical research, opts for crossing the meanings of authors who address the theme. The dialogue takes place considering the historicity, the cultural dynamics rooted in the communities, and the spaces of production, fruition, and connections. The aim is to recreate a space for reflection, where one can bet on the richness of the artistic processes present in handicrafts, finding their place in the world of arts and the creative economy, as well as their relevance as intangible heritage of their territories, in the desire for a place that gives hypotheses beyond conceptual limits.A pesquisa propõe um olhar sensível para a linguagem artística do artesanato como peça autoral; com conceito, simbolismo e materiais culturais regionais. O problema norteador está em como agregar valor ao artesanato, não se limitando aos aspectos funcionais do produto. A metodologia utilizada, essencialmente por meio de pesquisa bibliográfica, opta por cruzamento de acepções de autores que abordam o tema. O diálogo se dá considerando a historicidade, a dinâmica cultural enraizada nas comunidades e os espaços de produção, fruição e conexões. Deseja-se recriar um espaço de reflexão, onde se possa apostar na riqueza dos processos artísticos presentes no artesanato, encontrar seu lugar no mundo das artes e da economia criativa, bem como, sua relevância como patrimônio imaterial de seus territórios, no desejo de um lugar que dê hipóteses para além dos limites conceituais
