Genotypic and Phenotypic Heterogeneity in Alicyclobacillus acidoterrestris: A Contribution to Species Characterization.
Alicyclobacillus acidoterrestris is the main cause of most spoilage problems in fruit juices
and acidic products. Since soil borne species often contaminate fruit juices and do not need
strict extreme requirements for survival, it is a great concern to investigate whether and how
soil species could evolve from their ecological niches in microbial community to new environments as fruit juices. In this study, 23 isolates of thermo-acidophilic, spore-forming bacteria from soil were characterized by cultural and molecular methods. In addition, 2 strains
isolated from a spoilage incident in pear juice were typed. Strains phenotyping showed that
they could be grouped into 3 different clusters, and some isolates showed identical or quite
similar patterns. Analyzing pH and temperature ranges for growth, the majority of strains
were able to grow at values described for many species of Alicyclobacillus. Qualitative utilization of lysine, arginine and indole production from tryptophan revealed, for the first time,
deamination of lysine and decarboxylation of arginine. Resistance to 5% NaCl as well as
the ability to hydrolyze starch and gelatin, nitrate reduction, catalase and oxidase activities
confirmed literature evidences. Examining of 16S rRNA, showed that isolates were divided
into three blocks represented by effectively soil species and strains that are moving from
soil to other possible growing source characterized by parameters that could strongly influence bacterial survival. RAPD PCR technique evidenced a great variability in banding patterns and, although it was not possible to obtain genotypically well-distinguished groups, it
was feasible to appreciate genetic similarity between some strains. In conclusion, the investigation of a microbial community entails a combination of metagenomic and classic culturedependent approaches to expand our knowledge about Alicyclobacillus and to look for new
subspecies
Masked unbiased principles for parameter selection in variational image restoration under Poisson noise
In this paper we address the problem of automatically selecting the regularization parameter in variational models for the restoration of images corrupted by Poisson noise. More specifically, we first review relevant existing unmasked selection criteria which fully exploit the acquired data by considering all pixels in the selection procedure. Then, based on an idea originally proposed by Carlavan and Blanc-Feraud to effectively deal with dark backgrounds and/or low photon-counting regimes, we introduce and discuss the masked versions—some of them already existing—of the considered unmasked selection principles formulated by simply discarding the pixels measuring zero photons. However, we prove that such a blind masking strategy yields a bias in the resulting principles that can be overcome by introducing a novel positive Poisson distribution correctly modeling the statistical properties of the undiscarded noisy data. Such distribution is at the core of newly proposed masked unbiased counterparts of the discussed strategies. All the unmasked, masked biased and masked unbiased principles are extensively compared on the restoration of different images in a wide range of photon-counting regimes. Our tests allow to conclude that the novel masked unbiased selection strategies, on average, compare favorably with unmasked and masked biased counterparts
Membrane-seeded autologous chondrocytes: cell viability and characterization at surgery
The implantation of chondrocytes, seeded on matrices such as hyaluronic acid or collagen membranes, is a method that is being widely used for the treatment of chondral defects. The aim of the present study was to evaluate the distribution, viability and phenotype expression of the cells seeded on a collagen membrane just at the time of the implantation. Twelve patients who were suffering from articular cartilage lesions were treated by the MACI(®) procedure. The residual part of each membrane was tested by colorimetric assay (MTT) and histochemical and ultrastructural analyses were carried out. In all of the samples a large number of viable cells, quite homogenously distributed, was detected. The cells expressed the markers of the differentiated hyaline chondrocytes. These data reassure in that the MACI procedure provides a suitable engineered tissue for cartilage repair, in line with the clinical evidences emerging in the literature

Comparison of five different RNA sources to examine the lactating bovine mammary gland transcriptome using RNA-Sequencing.
The objective of this study was to examine five different sources of RNA, namely mammary gland tissue (MGT), milk somatic cells (SC), laser microdissected mammary epithelial cells (LCMEC), milk fat globules (MFG) and antibody-captured milk mammary epithelial cells (mMEC) to analyze the bovine mammary gland transcriptome using RNA-Sequencing. Our results provide a comparison between different sampling methods (invasive and non-invasive) to define the transcriptome of mammary gland tissue and milk cells. This information will be of value to investigators in choosing the most appropriate sampling method for different research applications to study specific physiological states during lactation. One of the simplest procedures to study the transcriptome associated with milk appears to be the isolation of total RNA directly from SC or MFG released into milk during lactation. Our results indicate that the SC and MFG transcriptome are representative of MGT and LCMEC and can be used as effective and alternative samples to study mammary gland expression without the need to perform a tissue biopsy

We describe the multidisciplinary work of nephrological realities belonging to two different Regional Health Systems, Lombardia and Lazio. The interventional nephrologist is a specialist in nephrology with ultra-specialist know-how for vascular access for dialysis. He is the coordinator of a team of vascular accesses and applies a team work with the other interventional nephrologists, with the vascular surgeon and the interventional radiologist, with whom he decides the diagnostic-therapeutic procedure to perform the best possible vascular access for that individual patient
Rescue of mutant rhodopsin traffic by metformin-induced AMPK activation accelerates photoreceptor degeneration
Protein misfolding caused by inherited mutations leads to loss of protein function and potentially toxic ‘gain of function’, such as the dominant P23H rhodopsin mutation that causes retinitis pigmentosa (RP). Here, we tested whether the AMPK activator metformin could affect the P23H rhodopsin synthesis and folding. In cell models, metformin treatment improved P23H rhodopsin folding and traffic. In animal models of P23H RP, metformin treatment successfully enhanced P23H traffic to the rod outer segment, but this led to reduced photoreceptor function and increased photoreceptor cell death. The metformin-rescued P23H rhodopsin was still intrinsically unstable and led to increased structural instability of the rod outer segments. These data suggest that improving the traffic of misfolding rhodopsin mutants is unlikely to be a practical therapy, because of their intrinsic instability and long half-life in the outer segment, but also highlights the potential of altering translation through AMPK to improve protein function in other protein misfolding diseases

An integrated assessment of the Good Environmental Status of Mediterranean Marine Protected Areas

targets to achieve Good Environmental Status (GES) of marine ecosystems by implementing the Marine Strategy
Framework Directive (MSFD) across member states. We combined an extensive dataset across five Mediterranean
ecoregions including 26 Marine Protected Areas (MPAs), their reference unprotected areas, and a no-trawl case
study. Our aim was to assess if MPAs reach GES, if their effects are local or can be detected at ecoregion level or
up to a Mediterranean scale, and which are the ecosystem components driving GES achievement. This was
undertaken by using the analytical tool NEAT (Nested Environmental status Assessment Tool), which allows an
integrated assessment of the status of marine systems. We adopted an ecosystem approach by integrating data from several ecosystem components: the seagrass Posidonia oceanica, macroalgae, sea urchins and fish. Thresholds to define the GES were set by dedicated workshops and literature review.
In the Western Mediterranean, most MPAs are in good/high status, with P. oceanica and fish driving this result
within MPAs. However, GES is achieved only at a local level, and the Mediterranean Sea, as a whole, results in a
moderate environmental status. Macroalgal forests are overall in bad condition, confirming their status at risk.
The results are significantly affected by the assumption that discrete observations over small spatial scales are
representative of the total extension investigated. This calls for large-scale, dedicated assessments to realistically
detect environmental status changes under different conditions.
Understanding MPAs effectiveness in reaching GES is crucial to assess their role as sentinel observatories of
marine systems. MPAs and trawling bans can locally contribute to the attainment of GES and to the fulfillment of
the MSFD objectives. Building confidence in setting thresholds between GES and non-GES, investing in long-term
monitoring, increasing the spatial extent of sampling areas, rethinking and broadening the scope of complementary tools of protection (e.g., Natura 2000 Sites), are indicated as solutions to ameliorate the status of the
