    Austrian Academy Corpus Digital Resources and Textual Studies

    The Austrian Academy Corpus (AAC) is a newly founded institution based at the Austrian Academy of Sciences in Vienna. It was designed to set up a text corpus and to conduct research in the field of electronic text corpora. The electronic text collections established at the AAC so far and its future projects will mainly focus on electronic representations not only of literary texts, literary magazines, journals and newspapers but also on a carefully considered selection of texts from many cultural and social domains. The aim of the proposed paper is to investigate the potential of digital resources for textual studies in various fields of the humanities. The paper will consider the advance of new systems of digital representation and its implications for the study of language, literature and cultural history. The paper will show the range of interests pursued in the AAC research group.     Loss of the Actin Remodeler Eps8 Causes Intestinal Defects and Improved Metabolic Status in Mice

    In a variety of organisms, including mammals, caloric restriction improves metabolic status and lowers the incidence of chronic-degenerative diseases, ultimately leading to increased lifespan. Here we show that knockout mice for Eps8, a regulator of actin dynamics, display reduced body weight, partial resistance to age- or diet-induced obesity, and overall improved metabolic status. Alteration in the liver gene expression profile, in behavior and metabolism point to a calorie restriction-like phenotype in Eps8 knockout mice. Additionally, and consistent with a calorie restricted metabolism, Eps8 knockout mice show increased lifespan. The metabolic alterations in Eps8 knockout mice correlated with a significant reduction in intestinal fat absorption presumably caused by a 25% reduction in intestinal microvilli length. Our findings implicate actin dynamics as a novel variable in the determination of longevity. Additionally, our observations suggest that subtle differences in energy balance can, over time, significantly affect bodyweight and metabolic status in mice

    Pleiotropic effects in Eya3 knockout mice

    <p>Abstract</p> <p>Background</p> <p>In <it>Drosophila</it>, mutations in the gene <it>eyes absent </it>(<it>eya</it>) lead to severe defects in eye development. The functions of its mammalian orthologs <it>Eya1-4 </it>are only partially understood and no mouse model exists for <it>Eya3</it>. Therefore, we characterized the phenotype of a new <it>Eya3 </it>knockout mouse mutant.</p> <p>Results</p> <p>Expression analysis of <it>Eya3 </it>by <it>in-situ </it>hybridizations and β-Gal-staining of <it>Eya3 </it>mutant mice revealed abundant expression of the gene throughout development, e.g. in brain, eyes, heart, somites and limbs suggesting pleiotropic effects of the mutated gene. A similar complex expression pattern was observed also in zebrafish embryos.</p> <p>The phenotype of young adult <it>Eya3 </it>mouse mutants was systematically analyzed within the German Mouse Clinic. There was no obvious defect in the eyes, ears and kidneys of <it>Eya3 </it>mutant mice. Homozygous mutants displayed decreased bone mineral content and shorter body length. In the lung, the tidal volume at rest was decreased, and electrocardiography showed increased JT- and PQ intervals as well as decreased QRS amplitude. Behavioral analysis of the mutants demonstrated a mild increase in exploratory behavior, but decreased locomotor activity and reduced muscle strength. Analysis of differential gene expression revealed 110 regulated genes in heart and brain. Using real-time PCR, we confirmed <it>Nup155 </it>being down regulated in both organs.</p> <p>Conclusion</p> <p>The loss of <it>Eya3 </it>in the mouse has no apparent effect on eye development. The wide-spread expression of <it>Eya3 </it>in mouse and zebrafish embryos is in contrast to the restricted expression pattern in <it>Xenopus </it>embryos. The loss of <it>Eya3 </it>in mice leads to a broad spectrum of minor physiological changes. Among them, the mutant mice move less than the wild-type mice and, together with the effects on respiratory, muscle and heart function, the mutation might lead to more severe effects when the mice become older. Therefore, future investigations of <it>Eya3 </it>function should focus on aging mice.</p

    IGF-1 and Bone: New Discoveries From Mouse Models

    Insulin-like growth factor-1 (IGF-1) plays a central role in cellular growth, differentiation, survival, and cell cycle progression. It is expressed early during development and its effects are mediated through binding to a tyrosine kinase receptor, the insulin-like growth factor-1 receptor (IGF-1R). In the circulation, the IGFs bind to IGF-binding proteins (IGFBPs), which determine their bioavailability and regulate the interaction between the IGFs and IGF-1R. Studies in animal models and in humans have established critical roles for IGFs in skeletal growth and development. In this review we present new and old findings from mouse models of the IGF system and discuss their clinical relevance to normal and pathological skeletal physiology. © 2010 American Society for Bone and Mineral Research

    Production Resource Flexibility Assessment: Case Studies in the Automotive Industry

    The automotive industry is characterized by a high degree of uncertainty. Companies are facing the challenge of producing different systems simultaneously. Additionally, the global quantity of electric vehicles is also expected to increase significantly. This results in the following capability to remain competitive: Effective and efficient adaptions of production systems to model variations and volume increases. While flexible production is identified as the most promising concept, defining the actual flexibility level of included production resources is essential for its proper realization. A literature review on existing flexibility assessment approaches revealed their emphasis on high-level enablers and limited practical applicability in the automotive industry. In contrast, focusing the assessment on single workstations supports the selection of appropriate production resources. Therefore, a simple and structured standard procedure for a production resource flexibility assessment was developed. This theoretical construct was subsequently complemented with practical insights through its application on two real-life case studies within one automotive engineering company. Summarizing and discussing the findings in combination with a conclusion completed this paper

    Agile Powertrain Development: Considerations to Incorporate Agile Principles

    Pressure to increase effectiveness and efficiency drives the product development process. The software industry has been using agile development approaches due to their advantages when coping with uncertainties and simultaneously increasing value. Therefore, the following work deals with the considerations of agile powertrain development. In order to identify important conditions for the incorporation of agile principles into powertrain development, semistructured interviews were conducted with experts from an engineering company. Having identified agile development challenges, advantages, and principles, the next step is comparing agile principles with the current principles of conventional powertrain development. Furthermore, a basic understanding of the powertrain development process regarding the incorporation of agile principles is established, the main attributes of powertrain development are investigated, and the key disciplines involved are identified. Finally, a model is created to assess whether the application of an agile or conventional development approach is more suitable
