The effect of pro-ecological procedures and insect foraging on the total content of phenol compounds in winter wheat
In laboratory conditions, the effect of pro-ecological procedures (application of effective microorganisms and Asahi SL biostimulator) and foraging by insects [cereal leaf beetle (Oulema melanopa L.) and bird cherry-oat aphid (Rhopalosiphum padi (L.)] on the total content of phenolic compounds in winter wheat, was studied. Correlations between the total content of phenolic compounds (determined by the Folin-Ciocalteau colorimetric method) expressed as the amount of pyrogallol in wheat plants: undamaged, damaged by O. melanopa, damaged by R. padi, the length of feeding scar left by cereal leaf beetle and the number of pricks made by actively feeding insects of bird cherry-oat aphid were analysed. The wheat was treated by EM inoculant and a biostimulator. The mode of application of the preparations used had a significant effect on level the total phenolic compounds in the undamaged wheat and the wheat exposed to foraging by the above-mentioned insects. The plants not exposed to insects foraging contained greater amounts of phenolic compounds than those exposed to the insects. Wpływ ogólnej zawartości związków fenolowych w wybranych roślinach kapustowatych na żerowanie i rozwój Dolycoris baccarum i Carpocoris fuscispinus (Pentatomidae, Hemiptera)
Background. The plants from the Brassicaceae family are the basic food product practically all over the world. In the growing season these plants can be attacked by insects. We studied the response of two phytophagous species from the family Pentatomidae to little-known but interesting plants from the family Brassicaceae. Material and methods. In laboratory conditions, the effect of the total content of phenolic compounds (Folin-Ciocalteau colorimetric method) in a few plants from the family Brassicaceae (Thlaspi arvense L., Descurainia sophia (L.) Webb ex Prantl, Berteroa incana (L.) D.C. and Diplotaxis tenuifolia (L.) D.C.) on the feeding and development parameters of two herbivores from the family Pentatomidae (Hemiptera): Dolycoris baccarum L. and Carpocoris fuscispinus (Boh.) was studied. Results. From among the above four plant species, B. incana contain the highest total amounts of phenolic compounds of 1.87% DM. D. baccarum much prefer feeding on D. sophia (53.3%) and the other one (C. fuscispinus ) on T. arvense. The eggs need on average 11 days for embryonal phase of development. There is a strong correlation between the total content of phenolic compounds and the total pricks of plant tissues for D. baccarum (r = -0.7812) and for C. fuscispinus eggs hatching (r = 0.9299). Conclusion. Among the two Pentatomidae insects, C. fuscispinus much prefer feeding and oviposition on all four plant species than D. baccarum.Rośliny kapustowate są podstawowym produktem żywnościowym praktycznie na całym świecie. Podczas wzrostu w sezonie wegetacyjnym mogą być atakowane przez owady. W warunkach laboratoryjnych badano wpływ zawartości sumy związków fenolowych (metoda kolorymetryczna Folin-Ciocalteau) w wybranych roślinach z rodziny Brassicaceae (T. arvense, D. sophia, B. incana i D. tenuifolia) na żerowanie i rozwój dwóch owadów z rodziny Pentatomidae (Hemiptera): D. baccarum i C. fuscispinus. Stwierdzono, że największą ilość polifenoli zawierał ekstrakt z części nadziemnych B. incana (1,87% SM). D. baccarum zdecydowanie preferowały żerowanie na D. sophia (53,3%), zaś C. fuscispinus na T. arvense. Jaja owadów potrzebowały średnio ok. 11 dni na przejście okresu rozwoju embrionalnego. Stwierdzono silną ujemną korelację pomiędzy zawartością sumy związków fenolowych a sumą nakłuć tkanek roślinnych przez D. baccarum (r = -0,7812) oraz z wylęgiem larw L1 dla C. fuscispinus (r = 0,9299). Wykazano, że spośród dwóch pluskwiaków z rodziny tarczówkowatych, C. fuscispinus chętniej żeruje i składa jaja na badanych gatunkach roślin niż D. baccarum
Reakcja Rhopalosiphum padi L. (Aphididae, Hemiptera) na zawartość flawonoidów w pszenicy ozimej traktowanej biostymulatorem i pożytecznymi mikroorganizmami
Background. The bird cherry-oat aphid (Rhopalosiphum padi L.) is one of the most important pests of cereal plants. Limiting its number by using pro-ecological methods such as biostimulants does not result in environmental contamination. Material and methods. Winter wheat plants were grown under laboratory conditions and treated with the following preparations: EM Naturalnie Aktywny, which is based on beneficial microorganisms, and/or the Asahi SL biostimulant. In the BBCH 32 phase some plants were harvested and the percentage flavonoid content, calculated as quercetin, was determined, while the remaining plants were used to assess the level of feeding and development of the bird cherry-oat aphid. Results. Wheat plants treated with biostimulants containing beneficial microorganisms contained on average 0.641% dry matter flavonoids. The pre-reproductive period of bird cherry-oat aphids was 8 days. Daily aphid fecundity was slightly more than three larvae per day. The length of the reproductive period of bird cherry-oat aphids on wheat plants ranged from 8 to 9 days. After 5 days of feeding, 2 adult aphids caused on average 15% damage to the plant surface. Correlation coefficients between the flavonoid content in wheat plants and development parameters and the extent of damage caused by bird cherry-oat aphids were insignificant. Conclusion. The use of a biostimulant and beneficial microorganisms in wheat cultivation was found to be not statistically significant, but a reduction in the feeding and development of R. padi was observed.Mszyca czeremchowo-zbożowa (Rhopalosiphum padi L.) należy do ważniejszych szkodników roślin zbożowych. Ograniczanie jej liczebności poprzez stosowanie proekologicznych metod, jak biostymulanty, nie powoduje zanieczyszczania środowiska. W laboratorium wysiewano ziarno pszenicy ozimej. Następnie w odpowiedniej fazie rozwojowej rośliny poddano aplikacji szczepionką EM Naturalnie Aktywny, opartej na pożytecznych mikroorganizmach, i/lub biostymulatorem Asahi SL. W fazie BBCH 32 część nadziemną połowy roślin ścięto i ustalono procentową zawartość flawonoidów w przeliczeniu na kwercetynę, a pozostałe posłużyły do badań oceniających poziom żerowania i rozwoju mszycy czeremchowo-zbożowej. Rośliny pszenicy traktowane szczepionką EM Naturalnie Aktywny i/lub biostymulatorem Asahi SL zawierały średnio 0,641% flawonoidów w suchej masie. Okres przedreprodukcyjny mszycy czeremchowo-zbożowej wyniósł 8 dni. Jej płodność dzienna wyniosła nieco ponad trzy larwy dziennie. Długość okresu reprodukcyjnego mszycy czeremchowo-zbożowej na roślinach pszenicy kształtowała się w przedziale od 8 do 9 dni. 2 imago mszyc przez 5 dni żerowania spowodowały 15% uszkodzeń powierzchni roślin. Współczynniki korelacji pomiędzy zawartością flawonoidów w roślinach pszenicy a parametrami rozwoju i żerowania mszycy czeremchowo-zbożowej były nieistotne. Stosowanie biostymulatora i pożytecznych mikroorganizmów w uprawie pszenicy skutkowało zauważalnym ograniczeniem żerowania i rozwoju R. padi. Nie wykazano jednak istotnego statystycznie wpływu tych preparatów na płodność, okres przedreprodukcyjny oraz reprodukcyjny mszycy
Właściwości suchego wyciągu z Nepeta cataria var. citriodora zwiększające zdolność przenikania
This paper presents the research on permeation enhancing properties of Nepeta cataria
var. citriodora (catnip) dry extract in comparison to oleanolic acid and ursolic acid. Progesterone
was chosen as a model substance for permeation test. The hydrogels made
of hydroxypropylmethylcellulose with progesterone, enhancers and ethanol were applied
in the study. The in vitro progesterone penetration test was based on the method
proposed by Fürst, using artificial lipophilic membranes which were made of colloxylin
and dodecanol. Statistical analyses showed an increase in penetration of progesterone
caused by catnip dry extract in comparison to ursolic acid and blank sample. HPLC assay
was applied to study the effect of enhancers on progesterone physicochemical properties.
The solubility of progesterone was tested in solvent systems corresponding to
liquid phases of gels. The statistical increase in progesterone solubility was observed
in the presence of dry extract in comparison to the result from ursolic acid-containing
sample. The partition coefficient of progesterone was evaluated by standard procedures.
The statistically significant reduction of log P values for progesterone was determined in
the presence of catnip dry extract.W pracy oceniono wpływ wyciągu suchego z Nepeta cataria var. citriodora (kocimiętki cytrynowej)
oraz kwasu oleanolowego i ursolowego (promotory wchłaniania) na penetrację
substancji czynnej. W badaniach wykorzystano hydrożele wykonane z hydroksypropylmetylcelulozy,
progesteronu (substancja modelowa), promotorów wchłaniania i etanolu. Badania
in vitro penetracji progesteronu były oparte o metodę zaproponowaną przez Fürsta,
wykorzystującą sztuczne lipofilowe błony wykonane z koloksyliny i dodekanolu. Analiza
statystyczna wykazała zwiększoną penetrację progesteronu spowodowaną obecnością
wyciągu suchego w porównaniu z kwasem ursolowym i próbą odniesienia. Metodę HPLC wykorzystano do zbadania wpływu promotorów na właściwości fizyko-chemiczne progesteronu.
Rozpuszczalność progesteronu była badana w układzie rozpuszczalników odpowiadającej
fazie płynnej żeli. Statystycznie istotny wzrost rozpuszczalności progesteronu
zaobserwowano w obecności wyciągu suchego w porównaniu z próbą zawierającą kwas
ursolowy. Współczynnik podziału określono za pomocą standartowych procedur. Statystycznie
istotny spadek wartości log P progesteronu zaobserwowano w obecności wyciągu
suchego z kocimiętki
Simultaneous UHPLC/DAD/( plus /-)HESI-MS/MS Analysis of Phenolic Acids and Nepetalactones in Methanol Extracts of Nepeta Species: A Possible Application in Chemotaxonomic Studies
Introduction - Nepeta species contain a variety of secondary
metabolites, including iridoid monoterpenes - nepetalactones and
phenolic acids - that are considered the main bioactive constituents.
This work represents the first attempt to comparatively explore
variations in these two major groups of secondary metabolites within the
genus.
Objective - To develop an efficient analytical methodology for
simultaneous analysis of nepetalactones and phenolic acids in methanol
extracts of selected Nepeta species, and to evaluate its potential
application in chemotaxonomic studies.
Material and methods - A UHPLC combined with linear-trap quadrupole
(LTQ) orbitrap MS method was used to characterise chemical diversity and
complexity of phenolics among 12 selected Nepeta species. A targeted
metabolomic approach using UHPLC coupled to a diode array detector (DAD)
and combined with (+/-) heated electrospray ionisation (HESI) MS/MS was
developed and validated for quantitative analysis of six hydroxycinnamic
acid derivatives and four nepetalactones.
Results - Phenolic profiling provided a valuable database of bioactive
compounds in the plant group studied, including phenolic acids
(hydroxybenzoic and hydroxycinnamic acids) and flavonoids (flavones,
flavonols and flavanones). Principal component analysis and cluster
analysis suggested the applicability of 10 targeted compounds as
chemomarkers for chemotaxonomic studies. Pearson's correlation analysis
revealed significant positive correlations between metabolites involved
in different biosynthetic pathways (phenylpropanoid or monoterpenoid).
Conclusion - The described targeted metabolomic approach proved to be
highly beneficial in designing a phytochemical overview of the genus
Nepeta, and might have applications in further clarification of
phylogenetic relations. Furthermore, it has the potential to be
implemented in a routine quality control of plant material and herbal
