Desenvolupament i aplicació de metodologies òmiques, analÃtiques i quimiomètriques en estudis ambientals
[cat] En aquesta Tesi s’han dissenyat, desenvolupat i aplicat noves metodologies quimiomètriques d’anà lisi multivariant de dades que permeten l’extracció d’informació quÃmica i bioquÃmica en estudis òmics i ambientals a partir de dades cromatogrà fiques. S’ha estudiat la influència de diferents factors externs (estadi de creixement, tipus de cultiu, varietat i tipus de mostra) sobre el cultiu del blat mitjançant la comparació de les metodologies ANOVA-PCA i ASCA. Aquestes eines quimiomètriques s’han aplicat sobre l’anà lisi dirigida dels perfils TIC LC-MS dels metabòlits secundaris (al·leloquÃmics) del blat. Aquestes eines han permès interpretar els canvis dels seus metabòlits en relació als factors externs considerats. La concentració dels metabòlits al·leloquÃmics DIMBOA-Glc, DIMBOA, HMBOA i MBOA ha canviat de forma sistemà tica durant el creixement de la planta del blat en relació als factors estadi de creixement, tipus de mostra i la seva interacció. S’ha desenvolupat una estratègia experimental no dirigida per a l’estudi i anà lisi d’experiments metabolòmics de mostres de llevat, utilitzant cromatografia lÃquida amb detecció per espectrometria de masses (LC-MS) i mètodes quimiomètrics d’anà lisi de les dades experimentals obtingudes. Aquesta estratègia ha permès avaluar l’efecte de condicions estressants sobre el cultiu del llevat (canvi de temperatura de cultiu i concentracions creixents de Cu(II)). S’han avaluat els canvis en les à rees dels perfils d’elució cromatogrà fica dels metabòlits resolts per MCR-ALS. Els mètodes quimiomètrics de selecció de variables VIP-PLS i SR-PLS han permès la discriminació entre mostres de llevat control i tractades. La identificació temptativa dels metabòlits s’ha fet a partir dels seus espectres de masses resolts per MCR-ALS. S’ha proposat una interpretació biològica dels canvis observats en el metaboloma com a conseqüència dels canvis en les condicions del seu cultiu (canvis de temperatura i concentracions creixents de Cu(II)). S’han observat modificacions en el metaboloma del llevat quan aquest s’ha cultivat a 42ºC (condicions estressants), i s’ha detectat l’inici procés d’estrès oxidatiu en el llevat quan aquest s’ha cultivat a concentracions (subletals) creixents de Cu(II). S’ha realitzat l’estudi de les correlacions existents entre les concentracions dels compostos orgà nics determinades per CG-MS acumulats als sediments marins (IODP- U1318) durant l’època del Miocè i la temperatura superficial del mar (SST) en el mateix perÃode de temps. S’ha aplicat el mètode VIP-PLSR als perfils cromatogrà fics TIC GC-MS de mostres de sediments marins per a la selecció de nous possibles marcadors paleoclimà tics dels canvis de temperatura en el perÃode de temps investigat. Aquest estudi ha permès la diferenciació entre les aportacions dels compostos del grup de lÃpids d’origen marà i terrestre. Finalment, s’han estudiat i comparat diferents mètodes quimiomètrics de selecció de variables, VIP-PLS i SR-PLS, en l’anà lisi de diversos conjunts de dades multivariants de naturalesa i complexitat diversa. En particular, aquest estudi s’ha aplicat a la selecció de variables en els següents casos: 1) selecció dels parà metres fisicoquÃmics de l’aigua que influeixen més en el seu gust; 2) selecció dels compostos orgà nics fòssils dels sediments marins, les concentracions dels quals (obtingudes per GC-MS) es correlacionaven més amb els canvis de temperatura superficial de l’aigua del mar en estudis paleoclimà tics; i 3) selecció dels gens, a partir d’anà lisi de xips d’ADN (microarrays), de l’organisme Daphnia magna exposat a dosis subletals de SSRI, en relació a la seva reproducció. S’ha demostrat que el mètode SR té una alta sensibilitat i especificitat. El mètode VIP també ha presentat alta sensibilitat i ha estat més rigorós per a la selecció de variables en els cromatogrames TIC, però ha demostrat baixa especificitat en la resta de conjunt de dades.[eng] In this Thesis we designed, developed and implemented new chemometric methodologies for data multivariate analysis to enable the extraction of chemical and biochemical information from chromatographic data in omic studies. We studied the influence of different external factors (growth stage, type of cultivation, tissue sample and variety) in wheat cultivation by comparing ANOVA-PCA and ASCA. These chemometric methods have been applied to TIC LC-MS profiles of secondary metabolites (allelochemicals) to interpret changes in relation to the considered factors. We developed an untargeted strategy for the study and analysis of metabolic yeast profiles using liquid chromatography coupled to mass spectrometry (LC-MS) and chemometric methods. This strategy allowed the evaluation of the effect of stressful conditions on yeast cultivation (temperature cultivation change and Cu(II) increasing concentrations). Changes in metabolites of the chromatographic elution profiles resolved by MCR-ALS were evaluated. PLS-DA method allowed the discrimination between treated and control yeast samples. Metabolites identification was performed from their mass spectra resolved by MCR-ALS. A biological interpretation of the observed changes in yeast metabolome was proposed as a result of cultivation condition changes. We studied the correlations between TIC GC-MS profiles of organic compounds accumulated in marine sediments (IODP-U1318) and sea surface temperatures (SST) during the early-middle Miocene. VIP-PLS method was applied to marine sediments chromatographic profiles for the selection of possible paleoclimatic markers of SST changes. Finally, we studied and compared VIP-PLS and SR-PLS methods for variable selection in the analysis of multivariate data sets of different nature and varying complexity: 1) physicochemical water parameters in relation to water taste; 2) fossil organic compounds (obtained by GC-MS) from marine sediments in relation to changes in SST; 3) Daphnia magna genes (obtained by DNA microarrays) exposed to sublethal doses of SSRI in relation to its reproduction
Authentication and quantitation of frauds in extra virgin olive oils based on HPLC-UV fingerprinting and multiariate calibration
HPLC-UV fingerprinting, obtained on a Zorbax Eclipse XDB-C8 reversed-phase column under gradient elution employing 0.1% formic acid aqueous solution and methanol as mobile phase, was applied to the analysis and characterization of olive oils. More than 130 edible oils, including monovarietal extra-virgin olive oils (EVOOs) and other vegetable oils, were analyzed. Principal component analysis results showed a noticeable discrimination between olive oils and other vegetable oils from raw HPLC-UV chromatographic profiles as data descriptors. In contrast, selected HPLC-UV chromatographic time-window segments were necessary to achieve discrimination among monovarietal EVOOs. Partial least square (PLS) regression was employed to tackle olive oil authentication of an Arbequina EVOO adulterated with a Picual EVOO, a refined olive oil, and a sunflower oil. Highly satisfactory results were obtained after PLS analysis, with overall errors in the quantitation of adulterations of an Arbequina EVOO (down to 2.5% adulterant) below 2.9%
HPLC-UV polyphenolic profiles in the classification of olive oils and other vegetable oils by principal component analysis
HPLC-UV was applied to the analysis and characterization of olive oils and other vegetable oils. A chromatographic separation on a Zorbax Eclipse XDB-C8 reversed-phase column was proposed under gradient elution, employing 0.1% formic acid aqueous solution and methanol as mobile phase, for the determination of 14 polyphenols and phenolic acids, allowing to obtain compositional profiles in less than 20 min. Acceptable sensitivity (LOD values down to 80 µg/L in the best of cases), linearity (r2 higher than 0.986), good run-to-run and day-to-day precisions (RSD values lower than 11.5%), and method trueness (relative errors lower than 6.8%) were obtained. The proposed HPLC-UV method was then applied to the analysis of 72 oil samples (47 olive oils and 27 vegetable oils including sunflower, soy, corn, and mixtures of them). Analytes were recovered by liquid-liquid extraction method employing ethanol:water 70:30 (v/v) solution and hexane as extracting and defatting solvents, respectively. HPLC-UV polyphenolic profiles using peak areas were then analysed by principal component analysis (PCA) to extract information of the most significant data contributing to the characterization and classification of olive oils against other vegetable oils, as well as among Arbequina and Picual olive oil varieties. PCA results showed a noticeable separation among olive oils and the other classes. Besides, a reasonable discrimination of olive oils as a function of fruit varieties was also encountered
Estudi del progrés d'una marea roja de Protoceratium reticulatum a la badia de Chipana (Xile) a través d'un cicle intensiu : efectes del fenomen de El Niño
Aquest projecte s'emmarca dins del projecte europeu CENSOR, que pretén fer una detecció, recopilació i comprensió dels efectes de El Niño i La Niña als ecosistemes marins costaners i als seus recursos. L'estudi es basa en l'ús de biomarcadors de producció fitoplanctònica en una zona de surgència (Iquique, Xile) per a conèixer el desenvolupament d'una marea roja tòxica de Protoceratium reticulatum, mitjançant l'anà lisi de les mostres corresponents a un cicle intensiu a la columna d'aigua. També s'estudia com aquesta pot influir en la disponibilitat d'aliment i en la comercialització dels organismes bentònics filtradors d'aquesta zona est del PacÃfic, i quins efectes potencials té el fenomen de El Niño sobre els blooms algals
Tooth crown size and dentine exposure in Australopithecus and early Homo: testing hypotheses of dietary related selective preasures
Preliminary analyses of total crown area and dentine exposure surfaces in a sample of hominid teeth seem to indicate that a significant correlation between tooth size and occlusal abrasion occurs, most probably related to dental occlusal function rather than to normal food mastication. Dental microwear on the buccal surfaces of teeth might be determined by foodstuffs abrasiveness whereas occlusal gross wear could be more dependent on either cultural practices with teeth or food processing. The effect of occlusal function of teeth on diet determination and ecological interpretation of hominid behaviour is dealt with, considering that habitat occupation and diet might affect differently and distinctly to tooth wear rate patterns
