    A Systematic Analysis on DNA Methylation and the Expression of Both mRNA and microRNA in Bladder Cancer

    Background: DNA methylation aberration and microRNA (miRNA) deregulation have been observed in many types of cancers. A systematic study of methylome and transcriptome in bladder urothelial carcinoma has never been reported. Methodology/Principal Findings: The DNA methylation was profiled by modified methylation-specific digital karyotyping (MMSDK) and the expression of mRNAs and miRNAs was analyzed by digital gene expression (DGE) sequencing in tumors and matched normal adjacent tissues obtained from 9 bladder urothelial carcinoma patients. We found that a set of significantly enriched pathways disrupted in bladder urothelial carcinoma primarily related to "neurogenesis" and "cell differentiation" by integrated analysis of -omics data. Furthermore, we identified an intriguing collection of cancer-related genes that were deregulated at the levels of DNA methylation and mRNA expression, and we validated several of these genes (HIC1, SLIT2, RASAL1, and KRT17) by Bisulfite Sequencing PCR and Reverse Transcription qPCR in a panel of 33 bladder cancer samples. Conclusions/Significance: We characterized the profiles between methylome and transcriptome in bladder urothelial carcinoma, identified a set of significantly enriched key pathways, and screened four aberrantly methylated and expressed genes. Conclusively, our findings shed light on a new avenue for basic bladder cancer research

    A common variant near TGFBR3 is associated with primary open angle glaucoma

    Primary open angle glaucoma (POAG), a major cause of blindness worldwide, is a complex disease with a significant genetic contribution. We performed Exome Array (Illumina) analysis on 3504 POAG cases and 9746 controls with replication of the most significant findings in 9173 POAG cases and 26 780 controls across 18 collections of Asian, African and European descent. Apart from confirming strong evidence of association at CDKN2B-AS1 (rs2157719 [G], odds ratio [OR] = 0.71, P = 2.81 × 10−33), we observed one SNP showing significant association to POAG (CDC7–TGFBR3 rs1192415, ORG-allele = 1.13, Pmeta = 1.60 × 10−8). This particular SNP has previously been shown to be strongly associated with optic disc area and vertical cup-to-disc ratio, which are regarded as glaucoma-related quantitative traits. Our study now extends this by directly implicating it in POAG disease pathogenesis

The Single stream FRR(Fast Re-Route) solution has the advantages of implementing fast multicast protection and of avoiding double multicast bandwidth occupation in both normal and abnormal situations.</p

    ACAT2 suppresses the ubiquitination of YAP1 to enhance the proliferation and metastasis ability of gastric cancer via the upregulation of SETD7

    Abstract The contributions of aberrantly expressed metabolic enzymes to gastric cancer (GC) initiation and progression have been widely appreciated in recent years. Acetyl-CoA acetyltransferase 2 (ACAT2) is one member of the acetyl- CoA thiolase family. Previous studies demonstrated that ACAT2 either promotes or suppresses tumor progression in different conditions. However, the function and mechanisms of ACAT2 in GC remain unknown. We found that the expression of this enzyme was significantly increased in GC tissues compared with normal counterparts, which prompted us to further investigate the roles of this protein in GC biology. In vitro functional studies showed that ACAT2 knockdown markedly halted the proliferation and the motility of GC cells; these functions favoring malignant phenotypes of GC cells were further validated in animal experiments. Mechanistically, ACAT2 depletion significantly reduced the transcription of SETD7, which is a histone methyltransferase and plays critical roles in GC cells. We found that the pro-tumoral functions of ACAT2 were largely dependent on SETD7. Moreover, SETD7 decreased the ubiquitination level of Yes-associated protein 1 (YAP1), thereby protecting YAP1 from proteasome degradation. Increased YAP1 protein expression remarkably activated the YAP1/TAZ-TEAD1 signaling pathway, which further boosted the malignant phenotypes in GC cells. In conclusion, these findings highlight the pro-tumoral functions and molecular underpinnings of ACAT2 in GC cells, and suggest that ACAT2 could be a promising target in GC treatment

    Research on explosion characteristics and consequences of compressor room in compressor station of long-distance natural gas pipeline

    As the core place in the compressor station, the compressor room has the possibility of gas leakage and explosion during its operation. In this paper, an equal proportional physical model is established based on FLCAS software, the effects of different stoichiometric ratios on the explosion temperature, flame propagation velocity and overpressure in the compressor room are investigated. The results show that when the stoichiometric ratio is in the range of 0.8–1.4, the indoor temperature peak, flame propagation velocity and peak overpressure generated by the explosion in the compressor room all increase first and then decrease with the increase of the stoichiometric ratio. The outdoor temperature peak and the flame propagation velocity increase with the increase of the stoichiometric ratio in the range of 0.8–1.4, and the flame acceleration during the flame propagation process can effectively promote the increase of the explosion overpressure inside the flame surface. In addition, in order to avoid more serious consequences in the event of an explosion in the compressor room, a fire sprinkler system should be laid between the adjacent compressor room doors, and the doors of the adjacent compressor rooms should be installed in staggered positions to prevent the occurrence of secondary danger

    A short-term study of laparoscopic-dominant individualised levator ani resection in abdominoperineal resection: A retrospective investigation

    Objective: The objective is to investigate if laparoscopic-dominant abdominoperineal resection (LDAPR) with individualised levator ani resection inhibits local recurrence (LR) and prolongs survival as compared to laparoscopic abdominoperineal resection (APR). Materials and Methods: Rectal cancer surgery cases were retrospectively identified from September 2014 to December 2019. LDAPR-treated group (55 patients) and the APR-treated group (71 patients) were included in the study. The operation time, circumferential resection margin (CRM), intraoperative tumor surgery (ITP), post-operative complications, the 2-year overall survival (OS) and LR were compared in the two groups. Results: The CRM and ITP were significantly reduced in the LDAPR as compared to the APR group (3.6% vs. 16.9%, t = 5.522, P = 0.019; 3.6% vs. 14.1%, t = 3.926, P = 0.048). In terms of post-operative complications, the incidence of urinary retention in LDAPR was significantly reduced than the APR group (10.9% vs. 25.4%, χ2 = 4.139, P = 0.041). Similarly, perineal pain at 6 months or 1 year after surgery was significantly down-regulated in LDAPR than in the APR group (72.7% vs. 88.7%, χ2 = 5.320, P = 0.021; 18.2% vs. 43.2%, χ2 = 8.288, P = 0.004). However, there was no statistically significant difference in the post-operative complications between the LDAPR and APR groups. Finally, LDAPR led to a significantly improved 2-year OS and a reduced LR compared to APR. Conclusion: LDAPR reduces CMR, ITP and LR and simplified the perineum operation, subsequently protecting the pelvic autonomic nerves. Compared to the conventional APR, LDAPR is a promising procedure worth adopting for rectal cancer treatment

    Energy-efficient two-way full-duplex relay transmission strategy with SWIPT and direct links

    Abstract In this paper, we improve networks’ spectral efficiency (SE), extend networks’ lifetime, and maximize networks’ energy efficiency (EE) of two-way full-duplex (FD) relay networks. Firstly, to improve networks’ SE and to extend networks’ lifetime simultaneously, we design a two-way FD relay transmission strategy with simultaneous wireless information and power transfer and direct links (DLs). The designed transmission strategy can complete a bidirectional communication in only one time slot with the exists of DLs and the energy-constrained relay node. With the designed transmission strategy, we further give the characteristics of relay amplification factor, the analysis of the designed transmission strategy, and the EE analysis of traditional half-duplex two-way amplify-and-forward relaying. Secondly, to maximize networks’ EE, we present both the EE maximization problems and analyses of the designed transmission strategy with equal power allocation and optimal power allocation. To solve the EE maximization problems, we further propose the alternating optimal algorithm and give complexity analysis of the algorithm. Simulations show that our designed transmission strategy can improve the SE and EE of the networks
