    Assessing the effect of β-glucan diets on innate immune response of tilapia macrophages against trichlorfon exposure: an in vitro study

    [EN] The widespread use of pesticides in some areas where fish species such as tilapia are farmed may cause damage to the environment and affect commercial fish and therefore, human health. Water leaching with the pesticide trichlorfon, during the fumigation season in the field, can affect water quality in fish farms and consequently affect fish health. At the same time, the use of immunomodulatory compounds such as β-glucan supplied in the diet has become widespread in fish farms as it has been shown that improves the overall immune response. The present research examines the immunomodulatory impacts observed in macrophages of Nile tilapia (Oreochromis niloticus) after being fed a diet supplemented with β-glucan for 15 days, followed by their in vitro exposure to trichlorfon, an organophosphate pesticide, at concentrations of 100 and 500 µg mL−1 for 24 h. The results showed that β-glucan diet improved the viability of cells exposed to trichlorfon and their antioxidant capacity. However, β-glucan did not counteract the effects of the pesticide as for the ability to protect against bacterial infection.     Imidacloprid-induced oxidative stress in honey bees and the antioxidant action of caffeine

    International audienceAbstractThe use of pesticides on crops contributes to the decline of bee populations, and in this sense, bioactive nutrients have been studied to counteract this effect. We suppose that caffeine might be one of these nutrients. We exposed honey bees (Apis mellifera L.) to 0.7 or 2.0 ng/mL imidacloprid, 5.0 μg/mL caffeine in syrup, or 5.0 μg/mL caffeine in syrup plus 0.7 or 2.0 ng/mL imidacloprid. After 72 h, the oxidative status and the food intake were verified. Imidacloprid increased glutathione peroxidase and catalase activities. Caffeine alone or with 2.0 ng/mL imidacloprid also stimulated the activity of glutathione peroxidase but did not alter the effect of the insecticide on the catalase activity. A significant reduction in the concentration of the thiol group of proteins was observed in the two imidacloprid-fed groups, and the addition of caffeine protected these groups. Imidacloprid increased the malondialdehyde concentration while the addition of caffeine partially decreased this effect. Food intake was higher for bees treated with 2.0 ng/mL imidacloprid. Our results show that imidacloprid increased the food intake resulting in oxidative damage, which was partially reversed by caffeine. From these findings, it is inferred that caffeine treatments can be used to mitigate the sublethal effects of this insecticide on honey bees

    Conceptions about the Use of Cannabis among Medical Students from Public Universities

    Background: Substance abuse is a public health concern given its high prevalence worldwide. The early onset of such abuse predicts greater severity of addiction, morbidity, and use of multiple drugs. The use of psychoactive substances among Brazilian university students is frequent and cannabis stands out as the most consumed illicit drug. This study aimed to assess the prevalence of cannabis use among medical students from public universities in the state of São Paulo, correlating it with socioeconomic data, perceptions, and conceptions about use, triggering factors and possible implications in academic performance.Methods: The data were collected using an anonymous online survey, which was sent to students attending public medical universities in the state of São Paulo in the year 2020.Findings: The survey was answered by 225 participants. Among all participants, 147 (65.3%) reported at least one episode of cannabis use during their study in university. 91 (61.9%) reported the first use before entering university, while 56 (38.1%) used it for the first time during the university years. The frequent group included 41 (27.9%) people and the sporadic group included 106 (72.1%) people.Conclusion: The present study indicated that the medical students in public universities in the state of São Paulo have higher cannabis use rates compared to the general Brazilian population and to other medical students worldwide. The users are aware of the possible damages caused by cannabis use, but this does not stop them from smoking

    World Press Photo 2012: the discursive construction of the Arab Spring

    O presente artigo pretende analisar a forma como o fenómeno da Primavera Árabe foi retratado nas fotografias vencedoras do concurso World Press Photo em 2012. As fotografias jornalísticas, embora se apresentem como índices do real, condicionam frequentemente a percepção dos indivíduos e influenciam as suas práticas sociais. Também as fotografias vencedoras do World Press Photo, um dos concursos mais prestigiados de fotojornalismo, em 2012 não fogem ao construtivismo discursivo que molda a representação e a percepção dos acontecimentos. A partir da análise crítica do discurso, nomeadamente dos instrumentos teóricos da semiótica barthesiana e da semiótica social de Gunther Kress e Theo van Leeuwen, procurámos interpretar as 38 fotografias vencedoras do World Press Photo 2012, a fim de reflectirmos sobre o modo como contribuíram para a compreensão da Revolta Árabe e como despertaram o nosso interesse e a nossa imaginação para o desenrolar do conflito.ABSTRACT:This article aims to analyze how the Arab Spring phenomenon was represented in the award-winning photographs of the World Press Photo contest in 2012. Although news photographs are usually seen as indexes of the real, they often limit the perception of individuals and influence their social practices.     Protective Role of Taurine against Arsenic-Induced Mitochondria-Dependent Hepatic Apoptosis via the Inhibition of PKCδ-JNK Pathway

    BACKGROUND: Oxidative stress-mediated hepatotoxic effect of arsenic (As) is mainly due to the depletion of glutathione (GSH) in liver. Taurine, on the other hand, enhances intracellular production of GSH. Little is known about the mechanism of the beneficial role of taurine in As-induced hepatic pathophysiology. Therefore, in the present study we investigated its beneficial role in As-induced hepatic cell death via mitochondria-mediated pathway. METHODOLOGY/PRINCIPAL FINDINGS: Rats were exposed to NaAsO(2) (2 mg/kg body weight for 6 months) and the hepatic tissue was used for oxidative stress measurements. In addition, the pathophysiologic effect of NaAsO(2) (10 microM) on hepatocytes was evaluated by determining cell viability, mitochondrial membrane potential and ROS generation. As caused mitochondrial injury by increased oxidative stress and reciprocal regulation of Bcl-2, Bcl-xL/Bad, Bax, Bim in association with increased level of Apaf-1, activation of caspase 9/3, cleavage of PARP protein and ultimately led to apoptotic cell death. In addition, As markedly increased JNK and p38 phosphorylation with minimal disturbance of ERK. Pre-exposure of hepatocytes to a JNK inhibitor SP600125 prevented As-induced caspase-3 activation, ROS production and loss in cell viability. Pre-exposure of hepatocytes to a p38 inhibitor SB2035, on the other hand, had practically no effect on these events. Besides, As activated PKCdelta and pre-treatment of hepatocytes with its inhibitor, rottlerin, suppressed the activation of JNK indicating that PKCdelta is involved in As-induced JNK activation and mitochondrial dependent apoptosis. Oral administration of taurine (50 mg/kg body weight for 2 weeks) both pre and post to NaAsO(2) exposure or incubation of the hepatocytes with taurine (25 mM) were found to be effective in counteracting As-induced oxidative stress and apoptosis. CONCLUSIONS/SIGNIFICANCE: Results indicate that taurine treatment improved As-induced hepatic damages by inhibiting PKCdelta-JNK signalling pathways. Therefore taurine supplementation could provide a new approach for the reduction of hepatic complication due to arsenic poisoning

    Inhibitors of trypanosoma cruzi Sir2 related protein 1 as potential drugs against Chagas disease.

    Chagas disease remains one of the most neglected diseases in the world despite being the most important parasitic disease in Latin America. The characteristic chronic manifestation of chagasic cardiomyopathy is the region's leading cause of heart-related illness, causing significant mortality and morbidity. Due to the limited available therapeutic options, new drugs are urgently needed to control the disease. Sirtuins, also called Silent information regulator 2 (Sir2) proteins have long been suggested as interesting targets to treat different diseases, including parasitic infections. Recent studies on Trypanosoma cruzi sirtuins have hinted at the possibility to exploit these enzymes as a possible drug targets. In the present work, the T. cruzi Sir2 related protein 1 (TcSir2rp1) is genetically validated as a drug target and biochemically characterized for its NAD+-dependent deacetylase activity and its inhibition by the classic sirtuin inhibitor nicotinamide, as well as by bisnaphthalimidopropyl (BNIP) derivatives, a class of parasite sirtuin inhibitors. BNIPs ability to inhibit TcSir2rp1, and anti-parasitic activity against T. cruzi amastigotes in vitro were investigated. The compound BNIP Spermidine (BNIPSpd) (9), was found to be the most potent inhibitor of TcSir2rp1. Moreover, this compound showed altered trypanocidal activity against TcSir2rp1 overexpressing epimastigotes and anti-parasitic activity similar to the reference drug benznidazole against the medically important amastigotes, while having the highest selectivity index amongst the compounds tested. Unfortunately, BNIPSpd failed to treat a mouse model of Chagas disease, possibly due to its pharmacokinetic profile. Medicinal chemistry modifications of the compound, as well as alternative formulations may improve activity and pharmacokinetics in the future. Additionally, an initial TcSIR2rp1 model in complex with p53 peptide substrate was obtained from low resolution X-ray data (3.5 Å) to gain insight into the potential specificity of the interaction with the BNIP compounds. In conclusion, the search for TcSir2rp1 specific inhibitors may represent a valuable strategy for drug discovery against T. cruzi
