    “Saia do Brasil Agora”: emigração brasileira como ação antecipatória

    In recent years, the number of Brazilians leaving the country has reached record levels, suggesting a new and unprecedented phase of emigration. In contrast to earlier periods of emigration from Brazil, this recent phase is primarily comprised of Brazilians who have left with their families with the intention of never returning to Brazil as residents. This article analyses this new movement through an ethnography of Brazilians living in Greater Vancouver, Canada, a region in which the Brazilian population quadrupled between 2006 and 2016. We argue that this migratory movement is a response to generalized and diffuse feelings of apprehension and of fear of the future in Brazil, that diverge from the  relatively comfortable standard of living and privileged position of those leaving Brazil. As a result, emigration emerges as a form of anticipatory action motivated by what we call “middle-class malaise.”Nos Ășltimos anos, o nĂșmero de brasileiros a deixarem o paĂ­s atingiu cifras recordes, o que parece indicar uma nova e sem precedentes fase emigratĂłria. Em contraste com os perĂ­odos anteriores da emigração brasileira, esta recente fase Ă© composta, principalmente, de brasileiros que partem em famĂ­lia, com a intenção de nĂŁo mais retornarem ao Brasil como residentes. O artigo analisa este novo movimento, por meio de uma etnografia dos brasileiros na Grande Vancouver, CanadĂĄ, regiĂŁo onde essa população triplicou entre 2006 e 2021. Argumentamos se tratar de um movimento migratĂłrio em resposta a sentimentos generalizados e difusos de apreensĂŁo e medo em relação ao futuro no Brasil, que destoam do padrĂŁo de vida confortĂĄvel e da posição relativamente privilegiada daqueles que estĂŁo se mudando.     Consumption patterns of sweet drinks in a population of Australian children and adolescents (2003–2008)

    <p>Abstract</p> <p>Background</p> <p>Intake of sweet drinks has previously been associated with the development of overweight and obesity among children and adolescents. The present study aimed to assess the consumption pattern of sweet drinks in a population of children and adolescents in Victoria, Australia.</p> <p>Methods</p> <p>Data on 1,604 children and adolescents (4–18 years) from the comparison groups of two quasi-experimental intervention studies from Victoria, Australia were analysed<it>.</it> Sweet drink consumption (soft drink and fruit juice/cordial) was assessed as one day’s intake and typical intake over the last week or month at two time points between 2003 and 2008 (mean time between measurement: 2.2 years).</p> <p>Results</p> <p>Assessed using dietary recalls, more than 70% of the children and adolescents consumed sweet drinks, with no difference between age groups (p = 0.28). The median intake among consumers was 500 ml and almost a third consumed more than 750 ml per day. More children and adolescents consumed fruit juice/cordial (69%) than soft drink (33%) (p < 0.0001) and in larger volumes (median intake fruit juice/cordial: 500 ml and soft drink: 375 ml). Secular changes in sweet drink consumption were observed with a lower proportion of children and adolescents consuming sweet drinks at time 2 compared to time 1 (significant for age group 8 to <10 years, p = 0.001).</p> <p>Conclusion</p> <p>The proportion of Australian children and adolescents from the state of Victoria consuming sweet drinks has been stable or decreasing, although a high proportion of this sample consumed sweet drinks, especially fruit juice/cordial at both time points.</p

    Your Trash Is Someone's Treasure The Politics of Value at a Michigan Landfill

    This article discusses scavenging and dumping as alternative approaches to deriving value from rubbish at a large Michigan landfill. Both practices are attuned to the indeterminacy and power of abandoned things, but in different ways. Whereas scavenging relies on acquiring familiarity with an object by getting to know its particular qualities, landfilling and other forms of mass disposal make discards fungible and manipulable by stripping them of their former identities. By way of examining the different ways in which people become invested in the politics of value at the landfill, whether as part of expressions of gender and class or for personal enjoyment, different comportments toward materiality are revealed to have underlying social and moral implications. In particular, it is argued that different approaches to the evaluation of rubbish involve competing understandings of human and material potential

    “Still good life”: On the value of reuse and distributive labor in “depleted” rural Maine

    This article explores the production of wealth through distributive labor in Maine\u27s secondhand economy. While reuse is often associated with economic disadvantage, our research complicates that perspective. The labor required to reclaim, repair, redistribute, and reuse secondhand goods provides much more than a means of living in places left behind by international capitalism, but the value generated by this work is persistently discounted by dominant economic logics. On the basis of semistructured interviews, participant observation, and statewide surveys with reuse market participants in Maine, we find that the relational value of reuse, produced through caring, flexible, distributive labor, is especially significant. We argue that paying attention to the practices, politics, and value of distribution is critical for understanding wealth in communities perceived to have been left behind by global capitalist systems, particularly as wage labor opportunities and natural resources grow increasingly scarce

    A dominant gain-of-function mutation in universal tyrosine kinase <i>SRC </i>causes thrombocytopenia, myelofibrosis, bleeding, and bone pathologies

    The Src family kinase (SFK)member SRC is amajor target in drug development because it is activated in many human cancers, yet deleterious SRC germline mutations have not been reported. We used genome sequencing and Human Phenotype Ontology patient coding to identify a gain-of-function mutation in SRC causing thrombocytopenia, myelofibrosis, bleeding, and bone pathologies in nine cases. Modeling of the E527K substitution predicts loss of SRC's self-inhibitory capacity, whichwe confirmedwith in vitro studies showing increased SRC kinase activity and enhanced Tyr419 phosphorylation in COS-7 cells overexpressing E527K SRC. The active form of SRC predominates in patients' platelets, resulting in enhanced overall tyrosine phosphorylation. Patientswith myelofibrosis have hypercellular bone marrow with trilineage dysplasia, and their stem cells grown in vitro form more myeloid and megakaryocyte (MK) colonies than control cells. These MKs generate platelets that are dysmorphic, low in number, highly variable in size, and have a paucity of a-granules. Overactive SRC in patient-derived MKs causes a reduction in proplatelet formation, which can be rescued by SRC kinase inhibition. Stem cells transduced with lentiviral E527K SRC formMKs with a similar defect and enhanced tyrosine phosphorylation levels. Patient-derived and E527K-transduced MKs show Y419 SRC- positive stained podosomes that induce altered actin organization. Expression of mutated src in zebrafish recapitulates patients' blood and bone phenotypes. Similar studies of platelets andMKs may reveal the mechanism underlying the severe bleeding frequently observed in cancer patients treated with next-generation SFK inhibitors. © 2016 by the American Association for the Advancement of Science; all rights reserved

    A gain-of-function variant in <i>DIAPH1 </i>causes dominant macrothrombocytopenia and hearing loss

    Macrothrombocytopenia (MTP) is a heterogeneous group of disorders characterized by enlarged and reduced numbers of circulating platelets, sometimes resulting in abnormal bleeding. In most MTP, this phenotype arises because of altered regulation of platelet formation from megakaryocytes (MK). We report the identification of DIAPH1, which encodes the Rho-effector diaphanous-related formin 1 (DIAPH1), as a candidate gene for MTP using exome sequencing, ontological phenotyping and similarity regression. We describe two unrelated pedigrees with MTP and sensorineural hearing loss that segregate with a DIAPH1 p.R1213* variant predicting partial truncation of the DIAPH1 diaphanous autoregulatory domain. The R1213* variant was associated with reduced proplatelet formation from cultured MKs, cell clustering and abnormal cortical filamentous actin. Similarly, in platelets there was increased filamentous actin and stable microtubules, indicating constitutive activation of DIAPH1. Over-expression of DIAPH1 R1213* in cells reproduced the cytoskeletal alterations found in platelets. Our description of a novel disorder of platelet formation and hearing loss extends the repertoire of DIAPH1-related disease and provides new insights into the autoregulation of DIAPH1 activity

    Expanded repertoire of RASGRP2 variants responsible for platelet dysfunction and severe bleeding.

    Heritable platelet function disorders (PFDs) are genetically heterogeneous and poorly characterized. Pathogenic variants in RASGRP2, which encodes calcium and diacylglycerol-regulated guanine exchange factor I (CalDAG-GEFI), have been reported previously in 3 pedigrees with bleeding and reduced platelet aggregation responses. To better define the phenotype associated with pathogenic RASGRP2 variants, we compared high-throughput sequencing and phenotype data from 2042 cases in pedigrees with unexplained bleeding or platelet disorders to data from 5422 controls. Eleven cases harbored 11 different, previously unreported RASGRP2 variants that were biallelic and likely pathogenic. The variants included 5 high-impact variants predicted to prevent CalDAG-GEFI expression and 6 missense variants affecting the CalDAG-GEFI CDC25 domain, which mediates Rap1 activation during platelet inside-out αIIbÎČ3 signaling. Cases with biallelic RASGRP2 variants had abnormal mucocutaneous, surgical, and dental bleeding from childhood, requiring ≄1 blood or platelet transfusion in 78% of cases. Platelets displayed reduced aggregation in response to adenosine 5'-diphosphate and epinephrine, but variable aggregation defects with other agonists. There were no other consistent clinical or laboratory features. These data enable definition of human CalDAG-GEFI deficiency as a nonsyndromic, recessive PFD associated with a moderate or severe bleeding phenotype and complex defects in platelet aggregation
