    Updated constraints on amplitude and tilt of the tensor primordial spectrum

    We have taken a comprehensive approach to update the limits on the tensor-to-scalar ratio (rr) and the tensor spectral index (ntn_t), using 10 datasets from the BICEP/Keck Array 2015 and 2018, Planck releases 3 and 4, and LIGO-Virgo-KAGRA Collaboration. By fitting the complete Λ\LambdaCDM+rr+ntn_t model with two different approaches for the tensor sector, we have not only established which method is the most reliable, but have also achieved the strongest constraint on the tensor-to-scalar ratio in current literature: r<0.028r<0.028 and −1.37<nt<0.42-1.37 < n_t < 0.42 at 95% confidence level. Furthermore, our examination of the common signal detected by the NANOGrav Collaboration further confirms that a simple power-law cannot reconcile the constraints from different datasets if the NANOGrav detection is due to a primordial inflationary gravitational wave background, as previously shown in the literature.Comment: Version coherent with published one.     Bioengineered Surfaces for Real-Time Label-Free Detection of Cancer Cells

    Biosensing technology is an advancing field that benefits from the properties of biological processes combined to functional materials. Recently, biosensors have emerged as essential tools in biomedical applications, offering advantages over conventional clinical techniques for diagnosis and therapy. Optical biosensors provide fast, selective, direct, and cost-effective analyses allowing label-free and real-time tests. They have also shown exceptional potential for integration in lab-on-a-chip (LOC) devices. The major challenge in the biosensor field is to achieve a fully operative LOC platform that can be used in any place at any time. The choice of an appropriate strategy to immobilize the biological element on the sensor surface becomes the key factor to obtain an applicable analytical tool. In this chapter, after a brief description of the main biofunctionalization procedures on silicon devices, two silicon-based chips that present an (i) IgG antibody or (ii) an Id-peptide as molecular probe, directed against the B-cell receptor of lymphoma cancer cells, will be presented. From a comparison in detecting cells, the Id-peptide device was able to detect lymphoma cells also at low cell concentrations (8.5 × 10−3 cells/ÎŒm2) and in the presence of a large amount of non-specific cells. This recognition strategy could represent a proof-of-concept for an innovative tool for the targeting of patient-specific neoplastic B cells during the minimal residual disease; in addition, it represents an encouraging starting point for the construction of a lab-on-a-chip system for the specific recognition of neoplastic cells in biological fluids enabling the follow-up of the changes of cancer cells number in patients, highly demanded for therapy monitoring applications

    Purpose: The aim of this study was to evaluate the relationships between health literacy, unrealistic optimism, and adherence to glycometabolic disease management related to erectile dysfunction (ED) in male patients with type 2 diabetes (T2D) or preDM.Materials and Methods: This prospective observational study enroled 167 consecutive patients with T2D and ED. All patients underwent the following examinations: (a) medical history collection; (b) Body Mass Index (BMI) determination; (c) hormonal and biochemical assessment; (d) duration of T2D, complications and treatment; (e) International Index of Erectile Function-5 questionnaire to assess ED; and (f) validated questionnaire to evaluate health literacy, unrealistic optimism, and treatment adherence.Results: Overall, mean age was 62.5 +/- 9.4 years (range: 20-75) and mean BMI was 28.4 +/- 4.8 kg/m(2) (range: 18.4-46.6). The mean IIEF-5 score was 15.4 +/- 5.2 (range: 5-25). The majority of patients showed high health literacy. However, low health literacy was found in patients with higher IIEF-5 scores and high BMI. Unrealistic optimism was low in most patients. Higher adherence to treatment was found in patients who reported regular physical activity, who followed a diet, and in patients with a family history of T2D. Regarding anti-diabetic treatment, patients treated with insulin showed higher health literacy than patients not treated with other medications, whereas higher adherence was found in patients using SGLT2-i.Conclusions: This study highlighted the close relationship between metabolic compensation, BMI, ED, and psychological attitudes, including health literacy and unrealistic optimism

    Cellular Interaction of Human Eukaryotic Elongation Factor 1A Isoforms

    Besides its canonical role in protein synthesis, the eukaryotic translation elongation factor 1A (eEF1A) is also involved in many other cellular processes such as cell survival and apoptosis. We showed that eEF1A phosphorylation by C-Raf in vitro occurred only in the presence of eEF1A1 and eEF1A2, thus suggesting that both isoforms interacted in cancer cells (heterodimer formation). This hypothesis was recently investigated in COS-7 cells where fluorescent recombinant eEF1A isoforms colocalized at the level of cytoplasm with a FRET signal more intense at plasma membrane level. Here, we addressed our attention in highlighting and confirming this interaction in a different cell line, HEK 293, normally expressing eEF1A1 but lacking the eEF1A2 isoform. To this end, His-tagged eEF1A2 was expressed in HEK 293 cells and found to colocalize with endogenous eEF1A1 in the cytoplasm, also at the level of cellular membranes. Moreover, FRET analysis showed, in this case, the appearance of a stronger signal mainly at the level of the plasma membrane. These results confirmed what was previously observed in COS-7 cells and strongly reinforced the interaction among eEF1A isoforms. Moreover, the formation of eEF1A heterodimer in cancer cells could also be important for cytoskeleton rearrangements rather than for phosphorylation, most likely occurring during cell survival and apoptosis

    Repeated Sprint Ability in Young Basketball Players (Part 2): The Chronic Effects of Multidirection and of One Change of Direction Are Comparable in Terms of Physiological and Performance Responses

    The aim of this study was to examine the effects of a 5-week training program, consisting of repeated 30-m sprints, on two repeated sprint ability (RSA) test formats: one with one change of direction (RSA) and the other with multiple changes of direction (RSM). Thirty-six young male and female basketball players (age 16.1 ± 0.9 years), divided into two experimental groups, were tested for RSA, RSM, squat jump, counter-movement jump, and the Yo-Yo Intermittent Recovery-Level-1 (Yo-Yo IR1) test, before and after a 4-week training program and 1 week of tapering. One group performed 30-m sprints with one change of direction (RSA group, RSAG), whereas the other group performed multidirectional 30-m sprints (RSM group, RSMG). Both groups improved in all scores in the post-intervention measurements (P < 0.05), except for the fatigue index in the RSM test. However, when comparing the two groups, similar effects were found for almost all parameters of the tests applied, except for RPE in the RSA test, which had a greater decrease in the RSAG (from 8.7 to 5.9) than in the RSMG (from 8.5 to 6.6, P = 0.021). We can conclude that repeated 30-m sprints, either with one change of direction or multidirectional, induce similar physiological and performance responses in young basketball players, but have a different psycho-physiological impact

    Diatomite is a natural porous silica material of sedimentary origin, formed by remains of diatom skeletons called “frustules.” The abundance in many areas of the world and the peculiar physico-chemical properties made diatomite an intriguing material for several applications ranging from food production to pharmaceutics. However, diatomite is a material still rarely used in biomedical applications. In this chapter, the properties of diatom frustules reduced to nanoparticles, with an average diameter less than 350 nm, as potential drug vectors are described. Their biocompatibility, cellular uptake, and capability to transport molecules inside cancer cells are discussed. Preliminary studies of in vivo toxicity are also presented.Peer reviewe

    Repeated Sprint Ability in Young Basketball Players: Multi-direction vs. One-Change of Direction (Part 1)

    The aim of the present study was to examine the reliability of a novel multi-direction repeated sprint ability (RSA) test [RSM; 10 7 (6 7 5-m)] compared with a RSA with one change of direction [10 7 (2 7 15-m)], and the relationship of the RSM and RSA with Yo-Yo intermittent recovery test level 1 (Yo-Yo IR1) and jump performances [squat jump (SJ) and counter-movement-jump (CMJ)]. Thirty-six (male, n = 14, female n = 22) young basketball players (age 16.0 \ub1 0.9 yrs) performed the RSM, RSA, Yo-Yo IR1, SJ, and CMJ, and were re-tested only for RSM and RSA after 1 week. The absolute error of reliability (standard error of the measurement) was lower than 0.212 and 0.617-s for the time variables of the RSA and RSM test, respectively. Performance in the RSA and RSM test significantly correlated with CMJ and SJ. The best time, worst time, and total time of the RSA and RSM test were negatively correlated with Yo-Yo IR1 distance. Based on these findings, consistent with previously published studies, it was concluded that the novel RSM test was valid and reliable
