    A study of Pc-5 ULF oscillations

    International audienceA study of Pc-5 magnetic pulsations using data from the Combined Release and Radiation Effects Satellite (CRRES) was carried out. Three-component dynamic magnetic field spectrograms have been used to survey ULF pulsation activity for the approximate fourteen month lifetime of CRRES. Two-hour panels of dynamic spectra were examined to find events which fall into two basic categories: 1) toroidal modes (fundamental and harmonic resonances) and 2) poloidal modes, which include compressional oscillations. The occurence rates were determined as a function of L value and local time. The main result is a comparable probability of occurence of toroidal mode oscillations on the dawn and dusk sides of the magnetosphere inside geosynchronous orbit, while poloidal mode oscillations occur predominantly along the dusk side, consistent with high azimuthal mode number excitation by ring current ions. Pc-5 pulsations following Storm Sudden Commencements (SSCs) were examined separately. The spatial distribution of modes for the SSC events was consistent with the statistical study for the lifetime of CRRES. The toroidal fundamental (and harmonic) resonances are the dominant mode seen on the dawn-side of the magnetosphere following SSCs. Power is mixed in all three components. In the 21 dusk side SSC events there were only a few examples of purely compressional (two) or radial (one) power in the CRRES study, a few more examples of purely toroidal modes (six), with all three components predominant in about half (ten) of the events. The compositions of the electrolytes were elaborated, and their electroplating characteristics were determined. It was shown that the effect of TRIS on the electrode process is due to its adsorption by zinc electrodes

    Crop wild relatives in the protected areas of the Republic of Bashkortostan

    Background. Comprehensive research into crop wild relatives (CWR) is essential to ensure food security. Protected areas serve as the main reserves for CWR in situ conservation and their population genetics studies. There are four federal protected areas in the Republic of Bashkortostan: the Bashkiria National Park, the Shulgan-Tash State Biosphere Nature Reserve, the South Ural and Bashkir State Nature Reserves.Materials and methods. The floristic study of CWR was conducted using the standard route method. The methods of comparative floristics were applied in the CWR analysis,. The economic value of plant species was assessed according to the ranking developed at the N.I. Vavilov All-Russian Institute of Plant Genetic Resources.Results. In total, out of 259 species of CWR growing in Bashkortostan, 186 (72%) occur in the federal protected areas, with the highest number in the Bashkiria National Park (148 spp.). Species belonging to the Poaceae, Fabaceae, Rosaceae and Lamiaceae families and the Poa L., Trifolium L., Lathyrus L., Allium L. genera are predominant. CWR are mainly confined to the forest meadow, forest margin, steppe and riverside communities. Alpine and raised bog CWR species occur only in the South-Ural Nature Reserve. There are 13 regional endemics in the studied territories and 34 species on their range boundaries. Of the 23 CWR species listed in the regional Red Book, seven are conserved in the surveyed areas: Allium obliquum L., Crambe tataria Sebeok, Linum uralense Juz., Melilotoides platycarpos (L.) Soják, Oxycoccus microcarpus Turcz. ex Rupr., Rubus arcticus L., and R. humulifolius C.A. Mey.Conclusion. Monitoring of the rare CWR species populations is required in the surveyed protected areas. Eight CWR species are recommended for further population genetics studies: Amygdalus nana L., Cerasus fruticosa Pall., Allium rubens Schrad. ex Willd., A. obliquum L., A. schoenoprasum L., Lathyrus gmelinii Fritsch, L. litvinovii Iljin, and L. sylvestris L

    Cytochalasin B-induced membrane vesicles convey angiogenic activity of parental cells

    © Gomzikova et al. Naturally occurring extracellular vesicles (EVs) play essential roles in intracellular communication and delivery of bioactive molecules. Therefore it has been suggested that EVs could be used for delivery of therapeutics. However, to date the therapeutic application of EVs has been limited by number of factors, including limited yield and full understanding of their biological activities. To address these issues, we analyzed the morphology, molecular composition, fusion capacity and biological activity of Cytochalasin B-induced membrane vesicles (CIMVs). The size of these vesicles was comparable to that of naturally occurring EVs. In addition, we have shown that CIMVs from human SH-SY5Y cells contain elevated levels of VEGF as compared to the parental cells, and stimulate angiogenesis in vitro and in vivo

    Cyclin A1 and P450 aromatase promote metastatic homing and growth of stem-like prostate cancer cells in the bone marrow

    Bone metastasis is a leading cause of morbidity and mortality in prostate cancer (PCa). While cancer stem-like cells have been implicated as a cell of origin for PCa metastases, the pathways which enable metastatic development at distal sites remain largely unknown. In this study, we illuminate pathways relevant to bone metastasis in this disease. We observed that cyclin A1 (CCNA1) protein expression was relatively higher in PCa metastatic lesions in lymph node, lung, and bone/bone marrow. In both primary and metastatic tissues, cyclin A1 expression was also correlated with aromatase (CYP19A1), a key enzyme that directly regulates the local balance of androgens to estrogens. Cyclin A1 overexpression in the stem-like ALDHhigh subpopulation of PC3M cells, one model of PCa, enabled bone marrow integration and metastatic growth. Further, cells obtained from bone marrow metastatic lesions displayed self-renewal capability in colony forming assays. In the bone marrow, Cyclin A1 and aromatase enhanced local bone marrow-releasing factors, including androgen receptor, estrogen and matrix metalloproteinase MMP9 and promoted hte metastatic growth of PCa cells. Moreover, ALDHhigh tumor cells expressing elevated levels of aromatase stimulated tumor/host estrogen production and acquired a growth advantage in the presence of host bone marrow cells. Overall, these findings suggest that local production of steroids and MMPs in the bone marrow may provide a suitable microenvironment for ALDHhigh PCa cells to establish metastatic growths, offering new approaches to therapeutically target bone metastases

    Targeted suppression of AR-V7 using PIP5K1α inhibitor overcomes enzalutamide resistance in prostate cancer cells

    One mechanism of resistance of prostate cancer (PCa) to enzalutamide (MDV3100) treatment is the increased expression of AR variants lacking the ligand binding-domain, the best characterized of which is AR-V7. We have previously reported that Phosphatidylinositol-4-phosphate 5-kinase alpha (PIP5Ka), is a lipid kinase that links to CDK1 and AR pathways. The discovery of PIP5Ka inhibitor highlight the potential of PIP5K1α as a drug target in PCa. In this study, we show that AR-V7 expression positively correlates with PIP5K1α in tumor specimens from PCa patients. Overexpression of AR-V7 increases PIP5K1α, promotes rapid growth of PCa in xenograft mice, whereas inhibition of PIP5K1α by its inhibitor ISA-2011B suppresses the growth and invasiveness of xenograft tumors overexpressing AR-V7. PIP5K1α is a key co-factor for both AR-V7 and AR, which are present as protein-protein complexes predominantly in the nucleus of PCa cells. In addition, PIP5K1α and CDK1 influence AR-V7 expression also through AKT-associated mechanism dependent on PTEN-status. ISA-2011B disrupts protein stabilization of AR-V7 which is dependent on PIP5K1α, leading to suppression of invasive growth of AR-V7-high tumors in xenograft mice. Our study suggests that combination of enzalutamide and PIP5K1α may have a significant impact on refining therapeutic strategies to circumvent resistance to antiandrogen therapies
