    Tato diplomová práce je zaměřena na zhodnocení výkonnosti a současného stavu firmy Diadex, s.r.o. Za pomoci specifických analýz a nástrojů, popsaných v teoretické části, analyzuje prostředí, ve kterém společnost působí, a odhaluje pozitiva a negativa, která významně ovlivňují firemní činnost. Na základě výsledků jednotlivých finančních i nefinančních ukazatelů poukazuje na silné a slabé stránky podniku. Vybrané metody by poté v závěru práce měly poukázat na oblasti, které vyžadují zvýšenou pozornost, a popřípadě navrhnout řešení pro změnu a zlepšení dané oblasti či celkové situace.This master’s thesis is focused on evaluation of performance and current state of the company Diadex Ltd. With the help of specific analysis and tools, described in the theoretical part, this thesis describes the environment, in which the company operates, and reveals pros and cons, that significantly affect corporate activities. Based on the results of financial and non-financial indicators, this work exposes the strengths and weaknesses. In conclusion, the selected methods should refer to the areas, which require increased attention, and possibly propose a solution for change and improvement in particular area or overall situation.

    Tato bakalářská práce je zaměřena na zjištění stavu a vývoje firmy Liolios Christos – Rychlostav. Za pomoci různých analýz popisuje prostředí, ve kterém působí, a odhaluje nedostatky, které významně ovlivňují firemní činnost. Na základě výsledků jednotlivých ukazatelů poukazuje na silné a slabé stránky podniku. Na závěr obsahuje návrhy přípustných řešení, které vedou k odstranění nebo zmírnění zjištěných nedostatků a ke zlepšení celkové situace.This bachelor thesis focuses on findings regarding the status and development of the firm Liolios Christos – Rychlostav. Using different analyses, this work describes the environment in which it operates and reveals deficiencies that significantly influence businesses activity. Based on the results of individual indicators, this thesis points to strengths and weaknesses. The conclusion includes proposals of acceptable solutions, which lead to the removal or mitigation of identified deficiencies to improve the overall situation.

    Utilization of the discrete differential evolution for optimization in multidimensional point clouds

    The Differential Evolution (DE) is a widely used bioinspired optimization algorithm developed by Storn and Price. It is popular for its simplicity and robustness. This algorithm was primarily designed for real-valued problems and continuous functions, but several modified versions optimizing both integer and discrete-valued problems have been developed. The discrete-coded DE has been mostly used for combinatorial problems in a set of enumerative variants. However, the DE has a great potential in the spatial data analysis and pattern recognition. This paper formulates the problem as a search of a combination of distinct vertices which meet the specified conditions. It proposes a novel approach called the Multidimensional Discrete Differential Evolution (MDDE) applying the principle of the discrete-coded DE in discrete point clouds (PCs). The paper examines the local searching abilities of the MDDE and its convergence to the global optimum in the PCs. The multidimensional discrete vertices cannot be simply ordered to get a convenient course of the discrete data, which is crucial for good convergence of a population. A novel mutation operator utilizing linear ordering of spatial data based on the space filling curves is introduced. The algorithm is tested on several spatial datasets and optimization problems. The experiments show that the MDDE is an efficient and fast method for discrete optimizations in the multidimensional point clouds.Web of Scienceart. no. 632953

    Tumour Necrosis Factor Inhibitor monotherapy versus combination therapy for the treatment of psoriatic arthritis: combined analysis of European biologics databases

    OBJECTIVE To investigate whether tumor necrosis factor inhibitor (TNFi) combination therapy with conventional synthetic disease-modifying antirheumatic drugs (csDMARD) is more effective for psoriatic arthritis (PsA) and/or improves TNFi drug survival compared to TNFi monotherapy. METHODS Five PsA biologics cohorts were investigated between 2000 and 2015: the ATTRA registry (Czech Republic); the Swiss Clinical Quality Management PsA registry; the Hellenic Registry of Biologics Therapies (Greece); the University of Bari PsA biologics database (Italy); and the Bath PsA cohort (UK). Drug persistence was analyzed using Kaplan-Meier and equality of survival using log-rank tests. Comparative effectiveness was investigated using logistic regression with propensity scores. Separate analyses were performed on (1) the combined Italian/Swiss cohorts for change in rate of Disease Activity Score in 28 joints (DAS28); and (2) the combined Italian, Swiss, and Bath cohorts for change in rate of Health Assessment Questionnaire (HAQ). RESULTS In total, 2294 patients were eligible for the drug survival analysis. In the Swiss (P = 0.002), Greek (P = 0.021), and Bath (P = 0.014) databases, patients starting TNFi in combination with methotrexate had longer drug survival compared to monotherapy, while in Italy the monotherapy group persisted longer (P = 0.030). In eligible patients from the combined Italian/Swiss dataset (n = 1056), there was no significant difference between treatment arms in rate of change of DAS28. Similarly, when also including the Bath cohort (n = 1205), there was no significant difference in rate of change of HAQ. CONCLUSION Combination therapy of a TNFi with a csDMARD does not appear to affect improvement of disease activity or HAQ versus TNFi monotherapy, but it may improve TNFi drug survival

    Obsahem diplomové práce je alternativní návrh nosné konstrukce jednokolejného ocelového železničního mostu o jednom poli. Most se nachází v km 44,179 na trati Jaroměř - Dvůr Králové. Překračovanou překážkou je silnice I/33. Teoretické rozpětí je 42 m. Hlavní nosná konstrukce je Langerův trám. Je tvořena trámem, obloukem, závěsy, mostovkou vyztuženou podélnými a příčnými výztuhami. Nosné prvky jsou z oceli S355J2+N, S235J2+N, S355NL. Návrh jednotlivých částí je prováděn podle platných norem ČSN EN.This master thesis content is alternative design of one span single-railed steel bridge. The bridge is located in km 44,179 on the track Jaroměř - Dvůr Králové. Crossing barrier is roadway I/33. The teoretical distance between supports is 42 m. Main bearing structure is designed as Langer beam. Structure consist of beams, arcs, suspenders, bridge deck with longitudinal and transversal bracings. Elements of the structure are made out of steel S355J2+N, S235J2+N, S355NL. The bridge is designed according to current standards ČSN EN.

    In my bachelor thesis, I present a design and an evaluation of a bearing steel structure of a train wash. It is located in the industrial zone of Lípa, a village located near Zlín. With a floorplan of 8 meters x 96 meters , it is divided into two dilatation units of 48 meters. The roof ridge is 10.71 meters high. Its basic bearing element is a steel frame connected by joints with the columns anchored in the foundations. The frame holds a steel rooftree, upon which the roof lies. Its spatial stiffness is ensured by a transverse wall and roof stiffeners. The washing frame has its own bearing structure so it in now way affects the designed structure at all. Each of the two front walls has a gate in it. A statistical calculation of the main bearing elements, including details and a drawing documentation, is included

    Obsahem bakalářské práce je návrh a posouzení nosné ocelové konstrukce myčky na vlaky. Objekt je umístěn v průmyslové zóně obce Lípa nacházejícího se nedaleko krajského města Zlína. Objekt má půdorysný tvar 8 x 96 m. Je rozdělen na dva dilatační celky po 48 m. Výška hřebene střechy dosahuje 10,71 m. Základním nosným prvkem je ocelový rám kloubové spojený ve vrcholu se sloupy vetknutými do základu. Na rámu jsou uloženy ocelové vaznice, na kterých leží střešní plášť. Prostorovou tuhost zajišťují příčná stěnová a střešní ztužidla. Mycí rám má vlastní nosnou konstrukci. Nevyvozuje tedy žádné účinky na navrhovanou konstrukci. Na čelních stěnách se nacházejí z obou stran vrata.
