Climate-ready conservation objectives: a scoping study
AbstractAnticipated future climate change is very likely to have a wide range of different types of ecological impact on biodiversity across the whole of Australia. There is a high degree of confidence that these changes will be significant, affecting almost all species, ecosystems and landscapes. However, because of the complexity of ecological systems and the multiple ways climate change will affect them, the details of the future change are less certain for any given species or location. The nature of the changes means that the multiple ways biodiversity is experienced, used and valued by society will be affected in different ways. The likely changes present a significant challenge to any societal aspiration to preserve biodiversity in its current state, for example, to maintain a species in its current abundance and distribution. Preserving biodiversity ‘as is’ may have been feasible in a stationary climate (one that is variable but not changing), but this will not be possible with the widespread, pervasive and large ecological changes anticipated under significant levels of climate change. This makes the impacts of climate change quite unlike other threats to biodiversity, and they challenge, fundamentally, what it actually means to conserve biodiversity under climate change: what should the objectives of biodiversity conservation be under climate change? And what are the barriers to recalibrating conservation objectives?Based on key insights from the scientific literature on climate change and biodiversity, the project developed three adaptation propositions about managing biodiversity:Conservation strategies accommodate large amounts of ecological change and the likelihood of significant climate change–induced loss in biodiversity. Strategies remain relevant and feasible under a range of possible future trajectories of ecological change.Strategies seek to conserve the multiple different dimensions of biodiversity that are experienced and valued by society. Together these propositions summarise the challenge of future climate change for biodiversity conservation, and define a new way of framing conservation we called the ‘climate ready’ approach. In the near term, conservation strategies may be able to include some consideration of these propositions. However, under significant levels of climate change many of the current approaches to conservation will become increasingly difficult and ineffective (e.g. maintaining community types in their current locations). This challenge is fundamentally different from that posed by other threats to biodiversity, and the climate-ready approach is akin to a paradigm shift in conservation.The project used a review of 26 conservation strategy documents (spanning scales from international to local) and four case studies with conservation agencies to test and refine the climate-ready approach. The project found the approach to be robust and highly relevant; in the majority of situations, if adopted, it would lead to significant changes in the objectives and priorities of conservation. There were also many ‘green shoots’ of elements of the new approach in existing conservation practice. However, the project found there are currently substantial barriers to fully adopting a climate-ready approach. These include the need for: further development of ecological characterisation of ecosystem health and human activities in landscapesmuch better understanding of how society values different aspects of biodiversity, including ecosystems and landscapesdevelopment of policy tools to codify and implement new ecologically robust and socially endorsed objectives. Changes in subcellular localization reveal interactions between human cytomegalovirus terminase subunits
BACKGROUND: During herpesvirus replication, terminase packages viral DNA into capsids. The subunits of herpes simplex virus terminase, UL15, UL28, and UL33, assemble in the cytoplasm prior to nuclear import of the complex. METHODS: To detect similar interactions between human cytomegalovirus terminase subunits, the orthologous proteins UL89, UL56, and UL51 were expressed in HEK-293 T cells (via transfection) or insect cells (via baculovirus infection) and subcellular localizations were detected by cellular fractionation and confocal microscopy. RESULTS: In both cell types, UL56 and UL89 expressed alone were exclusively cytoplasmic, whereas UL51 was ~50% nuclear. Both UL89 and UL56 became ~50% nuclear when expressed together, as did UL56 when expressed with UL51. Nuclear localization of each protein was greatest when all three proteins were co-expressed. CONCLUSIONS: These results support inclusion of UL51 as an HCMV terminase subunit and suggest that nuclear import of human cytomegalovirus terminase may involve nuclear import signals that form cooperatively upon subunit associations
Cigarette smoking worsens systemic inflammation in persons with metabolic syndrome
Background
Emerging data suggests that the combination of smoking and metabolic syndrome (MetS) markedly increases cardiovascular disease risk well beyond that of either condition. In this study we assess if this interaction can be explained by an additive increase in the risk of systemic inflammation by MetS and cigarette smoking. Methods
We evaluated 5,503 healthy non-diabetic Brazilian subjects (mean age of 43 ± 10 years, 79% males). Participants were divided into sub-groups of smokers and non-smokers with or without MetS. High-sensitivity C reactive protein (hs-CRP) was measured to assess degree of underlying inflammation. Results
Overall (19%) had hs-CRP \u3e 3 mg/L. In adjusted regression analyses, compared to non-smokers, there was a 0.19 mg/L (95% CI: 0.05, 0.32) increase in hs-CRP among smokers in the entire population and 0.63 mg/L (95% CI: 0.26, 1.01) increase among smokers with MetS while there was no significant increase among smokers without MetS (β = 0.09 95% CI: -0.05, 0.24). In a fully adjusted logistic regression model, smokers compared to non-smokers were 55% more likely to have elevated hs-CRP in the entire population (OR 1.55, 95% CI: 1.25, 1.92) and more than twice as likely to have elevated hs-CRP if they had MetS ( OR 2.05, 95% CI: 1.40, 3.01) while the risk was non-significant among those without MetS (OR = 1.29, 95% CI: 0.98, 1.69). The study demonstrates an additive effect of cigarette smoking on the risk of systemic inflammation in MetS thus highlighting the need for determining smoking status among those with MetS and aggressively targeting smoking cessation in this population
Canonical Wnt signals combined with suppressed TGFβ/BMP pathways promote renewal of the native human colonic epithelium
Background: A defining characteristic of the human intestinal epithelium is that it is the most rapidly renewing tissue in the body. However, the processes underlying tissue renewal and the mechanisms that govern their coordination have proved difficult to study in the human gut. Objective: To investigate the regulation of stem cell-driven tissue renewal by canonical Wnt and TGFβ/bone morphogenetic protein (BMP) pathways in the native human colonic epithelium. Design: Intact human colonic crypts were isolated from mucosal tissue samples and placed into 3D culture conditions optimised for steady-state tissue renewal. High affinity mRNA in situ hybridisation and immunohistochemistry were complemented by functional genomic and bioimaging techniques. The effects of signalling pathway modulators on the status of intestinal stem cell biology, crypt cell proliferation, migration, differentiation and shedding were determined. Results: Native human colonic crypts exhibited distinct activation profiles for canonical Wnt, TGFβ and BMP pathways. A population of intestinal LGR5/OLFM4-positive stem/progenitor cells were interspersed between goblet-like cells within the crypt-base. Exogenous and crypt cell-autonomous canonical Wnt signals supported homeostatic intestinal stem/progenitor cell proliferation and were antagonised by TGFβ or BMP pathway activation. Reduced Wnt stimulation impeded crypt cell proliferation, but crypt cell migration and shedding from the crypt surface were unaffected and resulted in diminished crypts. Conclusions: Steady-state tissue renewal in the native human colonic epithelium is dependent on canonical Wnt signals combined with suppressed TGFβ/BMP pathways. Stem/progenitor cell proliferation is uncoupled from crypt cell migration and shedding, and is required to constantly replenish the crypt cell population
Compound Voids and Unproductive Entrepreneurship: The Rise of the “English Fever” in China
China has emerged as an economic power due, in part, to government policies that opened China to the world and created a modern consumer culture. One of these policies is the advancement of English-language education, including private providers, which has spawned the “English Fever” phenomenon. We use the unique context of the private English-language education industry in China to illustrate the concept of “compound (institutional) voids” and their relationship to unproductive entrepreneurship. Our contribution is in untangling some of the complexities related to institutional relationships, and in describing how compound voids increase the likelihood of rent-seeking behavior.Ye
Increasing Accessibility for Community Participants at Academic Conferences
Background: To decrease the gulf between academic and community perspectives, there has been an increased call among stakeholder groups to collaborate in solving complex urban health problems. Despite these recent shifts, however, community partners face barriers to participate in conferences, ultimately limiting exchange of ideas and uptake of research data. This paper reports on the evaluation of the strategy used to engage community participants at the 4th International Conference on Urban Health (ICUH) held in Toronto, Canada, in October, 2005.
Method: We surveyed participants (n = 98) and conducted follow-up interviews (n = 23) to assess factors that facilitated attendance at ICUH 2005 as well as the impact of the conference on their work.
Results: Community registrants were drawn by accessible fee structures, scholarship opportunities, and preconference workshops relevant to their interests. Both community and academic registrants were drawn by the presence of a separate conference stream showcasing high-quality and rigorous community-based participatory research (CBPR). The conference provided valuable opportunities for networking with other community-based researchers by facilitating the development of relationships between community registrants and researchers, increased the profile and legitimacy of CBPR, and reinforced the value of community input in research. It also provided opportunities for capacity building—knowledge sharing and heightened awareness of CBPR.
Conclusion: The 4th ICUH had a significant impact on community registrants and provided valuable opportunities to bridge academic and community divides. These data support the need for comprehensive strategies for community engagement at health conferences
Action sports athletes as entrepreneurs: Female professional wakeboarders, sponsorship, and branding
This teaching case explores the sport of wakeboarding to examine how its female practitioners need to become entrepreneurial in order to make a living from the sport. Students ponder the financial pressures of becoming a professional female wakeboarder, and how to sustain momentum once one turns professional. The significance and inter-relationship of sponsorship and self-branding/promotion figure prominently. Costs of competing are high in the best of circumstances, and nearly insurmountable without corporate backing. Obtaining such support requires a healthy degree of marketing oneself. The female riders offer a number of anecdotes and recollections providing salience to issues concerning sustainability and gender inequity. Male riders may earn up to four times more on tour than female riders and there is pressure to adapt to the “boys’ club” to advance one's career. Earning a living through involvement in wakeboarding is difficult for women. Models of entrepreneurism are provided to guide student discussion in developing strategies to overcome issues for female wakeboarders making the sport financially attractive for female competitors.Ye
The effect of high-frequency rTMS of the left dorsolateral prefrontal cortex on the resolution of response, semantic and task conflict in the colour-word Stroop task.
Previous work investigating the effect of rTMS of left Dorso-Lateral Prefrontal Cortex (DLPFC) on Stroop task performance reports no changes to the Stroop effect but reduced reaction times on both congruent and incongruent trials relative to sham stimulation; an effect attributed to an enhanced attentional (or task) set for colour classification. The present study tested this account by investigating whether, relative to vertex stimulation, rTMS of the left DLPFC modifies task conflict, a form of conflict that arises when task sets for colour classification and word reading compete, given that this particular type of conflict would be reduced by an enhanced task set for colour classification. Furthermore, the present study included measures of other forms of conflict present in the Stroop task (response and semantic conflict), the potential effects on which would have been hidden in previous studies employing only incongruent and congruent stimuli. Our data showed that left DLPFC stimulation had no effect on the magnitude of task conflict, nor did it affect response,
semantic or overall conflict (where the null is supported by sensitive Bayes Factors in most cases). However, consistent with previous research left DLPFC stimulation had the general effect of reducing reaction times. We therefore show for the first time that relative to real vertex stimulation left DLPFC stimulation does not modify Stroop interference.
