Import 23/07/2015Tato diplomová práce se zabývá leteckou přepravou nebezpečného zboží v rámci firmy DHL Express. V úvodu jsou popsány počátky firmy DHL Express, všeobecná filozofie a legislativa. V další části je definována mezinárodní klasifikace nebezpečného zboží do příslušných tříd a dále postupy pro bezpečnou leteckou přepravu těchto látek v DHL Express dle platné legislativy. Na základě klasifikace a známých postupů, byla vytvořena příručka popisující konkrétní postupy pro případ nehody za účasti nebezpečných látek. Postupy jsou vytvořeny pro jednotlivé třídy nebo skupiny nebezpečných látek pro případ požáru nebo úniku těchto nebezpečných látek.The thesis deals with an air transport of dangerous goods in DHL Express company. The first part describes the beginnings of the company, its philosophy and legislation. The next part talks about dangerous goods international classification generally and safe air transport procedures in DHL Express regarding these dangerous goods according to applicable regulations. A manual describing specific procedures in case of accidents related to dangerous goods has been formed, based on the classification and already known procedures. Possibilities of SQL database design in MATLAB and realization of database structure for storing measurement results

Tato práce se zabývá možností vyžití databázových systémů SQL s programem MATLAB. Úkolem bylo provést průzkum na trhu v oblasti SQL databází, porovnat různé systémy a jejich implementaci. Cílem praktické části je potom vytvořit nástroje, které umožní pracovat s SQL databázemi prostřednictvím programu MATLAB, bez použití placeného Database Toolbox rozšíření. A tyto nástroje potom demonstrovat na ukázkovém příkladu.This thesis is describing the usage of SQL database systems with MATLAB. The objective is to do market research considering the use of SQL databases, compare those systems and their implementation. The aim is to create tools that would allow the SQL database to be operated using MATLAB without the usage of a paid Database Toolbox. The last goal is to showcase these tools.

Monitoring of the influence of indigenous culture of yeasts on the fermentation process of making wine

Tato diplomová práce se zabývá identifikací vinných kvasinek izolovaných ze zakvašeného vinného moštu. Kvasinky byly izolovány z moštu odrůdy Rulandského modrého, které bylo pěstováno a vyráběno podle poţadavků kladených na ekologické a integrované zemědělství. Odebrané vzorky byly zpracovány v laboratoři, kde byly získány čisté kultury jednotlivých kvasinek. Z těchto byla pomocí komerčního kitu vyizolována DNA, která byla pouţita k další analýze. Pomocí polymerázové řetězové reakce a za pouţití primerů ITS1 a ITS4 došlo k amplifikaci specifického úseku oblasti 5.8S-ITS rDNA. Získané PCR produkty byly poté elektroforeticky detekovány v agarózovém gelu a po následné purifikaci byly podrobeny restrikční analýze za pouţití tří restrikčních endonukleáz: HaeIII, HinfI a HhaI. DNA byla tímto rozštěpena na fragmenty specifické pro daný druh kvasinky a ty byly opět detekovány pomocí agarózové elektroforézy. Byla srovnána podobnost těchto izolátů za pouţití programu BioNumerics a výsledkem je dendrogram genetické podobnosti izolovaných kvasinek. Byla také provedena základní chemická analýza vzorků moštu.This thesis deals with the identification of wine yeasts isolated from the grape must using PCR-RFLP method. The yeasts were isolated from Pinot Noir grape variety must. Grapes were grown and produced in accordance with the requirements placed on organic and integrated farming. Samples were processed in the laboratory, where pure cultures of individual yeast were obtained. A commercial kit was used for yeast DNA isolation. Obtained DNA was used for further analysis. Using the polymerase chain reaction and the primers ITS1 and ITS4 a specific segment of 5.8S rDNA-ITS region was amplified. The PCR products were then detected by electrophoresis in an agarose gel, and after a subsequent purification, three restriction enzymes: HaeIII, HinfI and HhaI were subjected to restriction analysis. The DNA was digested to fragments specific for yeast species and they were detected by agarose electrophoresis. Similarity of these isolates was compared using BioNumerics program and the result is dendrogram of genetic similarity of isolated yeast. The basic chemical analysis of samples must was also performed.

Comparison of cultural change of the Kyrgyz in Afghanistan and Kyrgyzstan

Creation of the national state in Kyrgyzstan had significant influence to national awareness of the Kyrgyz. It was aroused here an interest in the history of own nation, there were presented historical constructs to the population about the ancientness and certain exclusivity of the Kyrgyz ethnic group, what many inhabitants adopted. For all Kyrgyz in all investigated areas is common relatively high historical awareness of membership to the Kyrgyz nation – this perception is not bound to the language. All Kyrgyz living beyond the Kyrgyzstan border perceive very similarly the creation of national state. It is a symbol of their national identity for them, to which they are looking up with a certain respect. Also the Afghani Kyrgyz that usually do not know the Kyrgyzstan state symbols express the wishes to visit Kyrgyzstan, a country, where they can feel as at home. More important that the proper area is the own existence of the Kyrgyz state that, already by its name, offers a certain proximity to the Kyrgyz living outside its borders
