Nanoneedles Induce Targeted siRNAÂ Silencing of p16 in the Human Corneal Endothelium
Nanoneedles can target nucleic acid transfection to primary cells at tissue interfaces with high efficiency and minimal perturbation. The corneal endothelium is an ideal target for nanoneedle-mediated RNA interference therapy aimed at enhancing its proliferative capacity, necessary for tissue regeneration. This work develops a strategy for siRNA nanoninjection to the human corneal endothelium. Nanoneedles can deliver p16-targeting siRNA to primary human corneal endothelial cells in vitro without toxicity. The nanoinjection of siRNA induces p16 silencing and increases cell proliferation, as monitored by ki67 expression. Furthermore, siRNA nanoinjection targeting the human corneal endothelium is nontoxic ex vivo, and silences p16 in transfected cells. These data indicate that nanoinjection can support targeted RNA interference therapy for the treatment of endothelial corneal dysfunction

FTO gene-lifestyle interactions on serum adiponectin concentrations and central obesity in a Turkish population
The aim of the study was to investigate whether lifestyle factors modify the association fat mass and obesity-associated (FTO) gene single nucleotide polymorphisms (SNPs) and obesity in a Turkish population. The study included 400 unrelated individuals, aged 24-50 years recruited in a hospital setting. Dietary intake and physical activity were assessed using 24-hour dietary recall and self-report questionnaire, respectively. A genetic risk score (GRS) was developed using FTO SNPs, rs9939609 and rs10163409. Body mass index and fat mass index were significantly associated with FTO SNP rs9939609 (P=0.001 and P=0.002, respectively) and GRS (P=0.002 and P=0.003, respectively). The interactions between SNP rs9939609 and physical activity on adiponectin concentrations, and SNP rs10163409 and dietary protein intake on increased waist circumference were statistically significant (Pinteraction=0.027 and Pinteraction=0.044, respectively). This study demonstrated that the association between FTO SNPs and central obesity might be modified by lifestyle factors in this Turkish population
The probiotics in dentistry: a narrative review
The total number of microbes that colonize the human body is far greater than the number of cells that make it up. In recent years, it has been shown that bacteria play an essential role in the body; in fact, they are essential for the maturation of the intestine, the development and control of the immune system, the development of the brain, the metabolism of macronutrients, the synthesis of vitamins, and the energy balance. Bacteria play an essential role in defense of their territory against the entry of other bacteria that may be pathogenic to health. Metchnikoff, about a century ago, invented probiotics, assuming that the use of certain bacteria could be beneficial to maintaining health. Bacteria colonize our body from birth and breastfeeding, using the bacterial flora of the mother by accessing newborns through the mouth. Antibiotic therapies in pregnancy or cesarean section prevent this flow of probiotics to infants and open the way for very important diseases, such as diabetes and obesity. The alterations of oral bacterial flora are responsible for numerous diseases of the oral cavity and the idea of the use of probiotics is leading the way to new therapeutic perspectives
FTO rs9939609 influence on adipose tissue localization in the Italian population
OBJECTIVE:
Among the genes involved in obesity, the Fat mass and obesity-associated gene (FTO) is certainly one of the most known and the relation between FTO rs9939609 and BMI is highly discussed; nevertheless, data about its influence on body composition are limited.
MATERIALS AND METHODS:
We carried out a study on a sample of 1066 Italian subjects, whose body composition and FTO rs9939609 were analyzed.
RESULTS:
We found significant relations between FTO with arm (p=0.01), abdomen (p=0.00), and trunk circumferences (p=0.00), BMI (p=0.01), FM% (p=0.00), and android FM% (p=0.01), whereas no relations were found between FTO and both gynoid fat and lean mass.
CONCLUSIONS:
To conclude, the relation between FTO and BMI is confirmed and is related specifically with android FM%. These results indicated that FTO rs9939609 may be a genetic etiological factor for obesity. Indeed, the specificity for the android FM% would indicate FTO as an etiological factor in the development of cardiovascular diseases
