    Stressful life events correlate with depression symptoms

    Stressful life events, such as abuse, divorce, or spending time in jail have been known to cause psychological and physical symptoms (Billings, 1982). Previous research has examined the association between the amount of stressful life events and the onset of depression(Kendler, 1999). The present study aimed to investigate the relationship between stress and depression in a sample of college students. As a part of a larger study, participants (N = 223) completed online questionnaires that measured their exposure to life stressors on the Life Stress Checklist - Revised (Wolfe and Kimerling, 1997) and depression symptoms on the Beck Depression Inventory (Beck et al., 1996). There was a small/weak positive correlation between the number of life stressors and depressive symptoms, r(221) = .26, p \u3c .01. Future directions from this study could include investigating age and sex differences in relation to stress and depression to see if there is a stronger correlation

    Multifunctional and durable graphene-based composite sponge doped with antimonene nanosheets

    The development of flexible multifunctional composites is an important topic in the fields of materials engineering, electronics, aerospace and biomedicine. However, there are still major challenges to achieve a variety of functions to meet the requirement for the application. Herein, a flexible multifunctional porous composite is successfully prepared by fabricating both modified graphene and antimonene into a melamine sponge. Compared with the graphene composite sponge, the addition of antimonene improved its electrochemical and sensing performances. The specific capacitance of antimonene/graphene composite sponge was significantly increased, while the capacitance retention rate was 83% under 20,000 charge–discharge cycles. The pressure sensitivity of the prepared flexible multifunctional device assembled was 44% higher than that of the graphene composite sponge. A power supply-integrated sensing system was assembled for monitoring human motion signals. The experimental results show that this system is a promising monitoring device with broad potentials in the fields of biosensing

    Bayesian non-negative factor analysis for reconstructing transcription factor mediated regulatory networks

    <p>Abstract</p> <p>Background</p> <p>Transcriptional regulation by transcription factor (TF) controls the time and abundance of mRNA transcription. Due to the limitation of current proteomics technologies, large scale measurements of protein level activities of TFs is usually infeasible, making computational reconstruction of transcriptional regulatory network a difficult task.</p> <p>Results</p> <p>We proposed here a novel Bayesian non-negative factor model for TF mediated regulatory networks. Particularly, the non-negative TF activities and sample clustering effect are modeled as the factors from a Dirichlet process mixture of rectified Gaussian distributions, and the sparse regulatory coefficients are modeled as the loadings from a sparse distribution that constrains its sparsity using knowledge from database; meantime, a Gibbs sampling solution was developed to infer the underlying network structure and the unknown TF activities simultaneously. The developed approach has been applied to simulated system and breast cancer gene expression data. Result shows that, the proposed method was able to systematically uncover TF mediated transcriptional regulatory network structure, the regulatory coefficients, the TF protein level activities and the sample clustering effect. The regulation target prediction result is highly coordinated with the prior knowledge, and sample clustering result shows superior performance over previous molecular based clustering method.</p> <p>Conclusions</p> <p>The results demonstrated the validity and effectiveness of the proposed approach in reconstructing transcriptional networks mediated by TFs through simulated systems and real data.</p

    A facile approach to fabricate highly sensitive, flexible strain sensor based on elastomeric/graphene platelet composite film

    This work developed a facile approach to fabricate highly sensitive and flexible polyurethane/graphene platelets composite film for wearable strain sensor. The composite film was fabricated via layer-by-layer laminating method which is simple and cost-effective; it exhibited outstanding electrical conductivity of 1430 ± 50 S/cm and high sensitivity to strain (the gauge factor is up to 150). In the sensor application test, the flexible strain sensor achieves real-time monitoring accurately for five bio-signals such as pulse movement, finger movement, and cheek movement giving a great potential as wearable-sensing device. In addition, the developed strain sensor shows response to pressure and temperature in a certain region. A multifaceted comparison between reported flexible strain sensors and our strain sensor was made highlighting the advantages of the current work in terms of (1) high sensitivity (gauge factor) and flexibility, (2) facile approach of fabrication, and (3) accurate monitoring for body motions

    PON2 Deficiency Leads to Increased Susceptibility to Diet-Induced Obesity.

    (1) Background: Paraoxonase 2 (PON2) is a ubiquitously expressed protein localized to endoplasmic reticulum and mitochondria. Previous studies have shown that PON2 exhibits anti-oxidant and anti-inflammatory functions, and PON2-deficient (PON2-def) mice are more susceptible to atherosclerosis. Furthermore, PON2 deficiency leads to impaired mitochondrial function. (2) Methods: In this study, we examined the susceptibility of PON2-def mice to diet-induced obesity. (3) Results: After feeding of an obesifying diet, the PON2-def mice exhibited significantly increased body weight due to increased fat mass weight as compared to the wild-type (WT) mice. The increased adiposity was due, in part, to increased adipocyte hypertrophy. PON2-def mice had increased fasting insulin levels and impaired glucose tolerance after diet-induced obesity. PON2-def mice had decreased oxygen consumption and energy expenditure. Furthermore, the oxygen consumption rate of subcutaneous fat pads from PON2-def mice was lower compared to WT mice. Gene expression analysis of the subcutaneous fat pads revealed decreased expression levels of markers for beige adipocytes in PON2-def mice. (4) Conclusions: We concluded that altered systemic energy balance, perhaps due to decreased beige adipocytes and mitochondrial dysfunction in white adipose tissue of PON2-def mice, leads to increased obesity in these mice

    Investigative baseline reference on the status of pork pH, shear force, colour, drip and cooking loss in RYR1 mutation free, commercial 3-way crosses in Malaysia

    This paper attempts to provide findings of an investigative study on the baseline status of the pork quality in Malaysia. With consumer preferences changing towards the selection of good quality meat for consumption, there is a need to establish an investigative reference for the operators in the industry to gauge the performance of their animals and pork quality. This is also important to increase the competitiveness among producers to continuously improve the pork quality available to consumers. In this study, 30 commercial three-way crossed female pigs were randomly selected from government accredited abattoirs from east and west Malaysia and longisimus dorsi were collected for the determination of pH, drip loss, cooking loss, shear force and colour. All animals were screened for the RYR1 gene and the results were then compiled with statistical analysis to obtain an investigative baseline pork quality data in Malaysia. The average pork quality obtained from this study falls within the category of Red, Soft and Exudative (RSE), with an average ultimate pH of 5.83, drip loss more than 5% and L* values at 45.94. We have proposed an investigative baseline meat quality data for Malaysian pork from the average commercial pork quality data obtained. The proposed investigative pork quality baseline data in Malaysian is comparable in terms of studies done in other established countries and/or with international standards and falls within the RSE category of acceptable quality. It provides an investigative benchmark for researchers and end-producers to judge the quality of pork in an objective manner, both for consumption and for export purpose. Moreover, continuous selection against the RYR1 gene has successfully removed the gene from the sample size above, but constant random monitoring is still advisable if farms aim to ensure the elimination of this gene from their herd

    Reserpine-Induced Reduction in Norepinephrine Transporter Function Requires Catecholamine Storage Vesicles

    Treatment of rats with reserpine, an inhibitor of the vesicular monoamine transporter (VMAT), depletes norepinephrine (NE) and regulates NE transporter (NET) expression. The present study examined the molecular mechanisms involved in regulation of the NET by reserpine using cultured cells. Exposure of rat PC12 cells to reserpine for a period as short as 5min decreased [ H]NE uptake capacity, an effect characterized by a robust decrease in the V of the transport of [ H]NE. As expected, reserpine did not displace the binding of [ H]nisoxetine from the NET in membrane homogenates. The potency of reserpine for reducing [ H]NE uptake was dramatically lower in SK-N-SH cells that have reduced storage capacity for catecholamines. Reserpine had no effect on [ H]NE uptake in HEK-293 cells transfected with the rat NET (293-hNET), cells that lack catecholamine storage vesicles. NET regulation by reserpine was independent of trafficking of the NET from the cell surface. Pre-exposure of cells to inhibitors of several intracellular signaling cascades known to regulate the NET, including Ca /Ca -calmodulin dependent kinase and protein kinases A, C and G, did not affect the ability of reserpine to reduce [ H]NE uptake. Treatment of PC12 cells with the catecholamine depleting agent, α-methyl-p-tyrosine, increased [ H]NE uptake and eliminated the inhibitory effects of reserpine on [ H]NE uptake. Reserpine non-competitively inhibits NET activity through a Ca -independent process that requires catecholamine storage vesicles, revealing a novel pharmacological method to modify NET function. Further characterization of the molecular nature of reserpine\u27s action could lead to the development of alternative therapeutic strategies for treating disorders known to be benefitted by treatment with traditional competitive NET inhibitors

    The effectiveness of a smartphone intervention targeting suicidal ideation in young adults : randomized controlled trial examining the influence of loneliness

    Background: Loneliness is commonly reported by young people and has been shown to contribute to the rapid onset and escalation of depression and suicidal ideation during adolescence. Lonely people may also be particularly susceptible to disengaging from treatment early given the likelihood of their more complex clinical profiles leading to cognitive fatigue. While a smartphone intervention (LifeBuoy) has been shown to effectively reduce suicidal ideation in young adults, poor engagement is a well-documented issue for this therapeutic modality and has been shown to result in poorer treatment outcomes. Objective: This study aims to determine whether loneliness affects how young people experiencing suicidal ideation engage with and benefit from a therapeutic smartphone intervention (LifeBuoy). Methods: A total of 455 community-based Australian young adults (aged 18-25 years) experiencing recent suicidal ideation were randomized to use a dialectical behavioral therapy–based smartphone intervention (LifeBuoy) or an attention-matched control app (LifeBuoy-C) for 6 weeks. Participants completed measures of suicidal ideation, depression, anxiety, and loneliness at baseline (T0), post intervention (T1), and 3 months post intervention (T2). Piecewise linear mixed models were used to examine whether loneliness levels moderated the effect of LifeBuoy and LifeBuoy-C on suicidal ideation and depression across time (T0 to T1; T1 to T2). This statistical method was then used to examine whether app engagement (number of modules completed) influenced the relationship between baseline loneliness and suicidal ideation and depression across time. Results: Loneliness was positively associated with higher levels of overall suicidal ideation (B=0.75, 95% CI 0.08-1.42; P=.03) and depression (B=0.88, 95% CI 0.45-1.32; P<.001), regardless of time point or allocated condition. However, loneliness did not affect suicidal ideation scores across time (time 1: B=1.10, 95% CI –0.25 to 2.46; P=.11; time 2: B=0.43, 95% CI –1.25 to 2.12; P=.61) and depression scores across time (time 1: B=0.00, 95% CI –0.67 to 0.66; P=.99; time 2: B=0.41, 95% CI –0.37 to 1.18; P=.30) in either condition. Similarly, engagement with the LifeBuoy app was not found to moderate the impact of loneliness on suicidal ideation (B=0.00, 95% CI –0.17 to 0.18; P=.98) or depression (B=–0.08, 95% CI –0.19 to 0.03; P=.14). Conclusions: Loneliness was not found to affect young adults’ engagement with a smartphone intervention (LifeBuoy) nor any clinical benefits derived from the intervention. LifeBuoy, in its current form, can effectively engage and treat individuals regardless of how lonely they may be
