    PRIORIZAÇÃO DOS CRITÉRIOS COMPETITIVOS DAS OPERAÇÕES BASEADO NOS RECURSOS ESTRATÉGICOS

    Na literatura da área de estratégia de operações, os critérios competitivos devem ser priorizados tendo como referência a importância desses para os clientes, o desempenho da comparativo aos concorrentes, os trade-offs existentes entre os critérios e a disponibilidades de recursos estratégicos. Entretanto, há uma lacuna na literatura de como fazer essa escolha. Nesse sentido foi realizada a revisão da literatura sobre os temas análise estratégica de operações e teoria baseada em recursos com vistas a contribuir para o entendimento de como os recursos estratégicos se associam com os critérios competitivos. Dessa revisão foi proposto um framework de análise. Esse framework foi pré-testado em duas grandes empresas de Canoas-RS. Durante essa etapa foram realizadas observações diretas nas empresas e entrevistas. Os resultados mostram que um recurso específico pode contribuir com vários critérios competitivos, não havendo uma relação específica entre um recurso estratégico e um critério competitivo em particular e que o framework pode ser utilizado para pesquisas futuras.-----------------------------------------------In the literature of the field of strategy of operations, the competitive criteria must be priorized having as reference the importance of those for the clients, the comparative performance for the competitors, the existing trade-offs between the criteria and the availability of strategic resources. Therefore, there is a lack in the literature in how to make this choice. In this sense, it was carried out a revision of the literature about the themes strategic analysis of operations and theory based on resources aiming to contribute for the understanding of how the strategic resources associate with the competitive criteria. From this revision was proposed a framework of analysis. This framework was pre-tested in two big companies in Canoas, State of Rio Grande do Sul. In the time of this stage, direct observations were carried out in the companies and in the interviews.     Microrheology reveals microscale viscosity gradients in planktonic systems

    Microbial activity in planktonic systems creates a dynamic and heterogeneous microscale seascape that harbors a diverse community of microorganisms and ecological interactions of global significance. In recent decades great effort has been put into understanding this complex system, particularly focusing on the role of chemical patchiness, while overlooking a physical parameter that governs microbial life and is affected by biological activity: viscosity. Here we reveal spatial heterogeneity of viscosity in planktonic systems by using microrheological techniques that allow measurement of viscosity at length scales relevant to microorganisms. We show the viscous nature and the spatial extent of the phycosphere, the region surrounding phytoplankton. In ~45% of the phytoplankton cells analyzed we detected increases in viscosity that extended up to 30 μm away from the cell with up to 40 times the viscosity of seawater. We show also how these gradients of viscosity can be amplified around a lysing phytoplankton cell as its viscous contents leak away. Finally, we report conservative estimates of viscosity inside marine aggregates, hotspots of microbial activity, more than an order of magnitude higher than in seawater. Since the diffusivities of dissolved molecules, particles and microorganisms are inversely related to viscosity, microheterogeneity in viscosity alters the microscale distribution of microorganisms and their resources, with pervasive implications for the functioning of the planktonic ecosystem.     Obtaining Valid Laboratory Data in Clinical Trials Conducted in Resource Diverse Settings: Lessons Learned from a Microbicide Phase III Clinical Trial

    BACKGROUND: Over the last decade several phase III microbicides trials have been conducted in developing countries. However, laboratories in resource constrained settings do not always have the experience, infrastructure, and the capacity to deliver laboratory data meeting the high standards of clinical trials. This paper describes the design and outcomes of a laboratory quality assurance program which was implemented during a phase III clinical trial evaluating the efficacy of the candidate microbicide Cellulose Sulfate 6% (CS) [1]. METHODOLOGY: In order to assess the effectiveness of CS for HIV and STI prevention, a phase III clinical trial was conducted in 5 sites: 3 in Africa and 2 in India. The trial sponsor identified an International Central Reference Laboratory (ICRL), responsible for the design and management of a quality assurance program, which would guarantee the reliability of laboratory data. The ICRL provided advice on the tests, assessed local laboratories, organized trainings, conducted supervision visits, performed re-tests, and prepared control panels. Local laboratories were provided with control panels for HIV rapid tests and Chlamydia trachomatis/Neisseria gonorrhoeae (CT/NG) amplification technique. Aliquots from respective control panels were tested by local laboratories and were compared with results obtained at the ICRL. RESULTS: Overall, good results were observed. However, discordances between the ICRL and site laboratories were identified for HIV and CT/NG results. One particular site experienced difficulties with HIV rapid testing shortly after study initiation. At all sites, DNA contamination was identified as a cause of invalid CT/NG results. Both problems were timely detected and solved. Through immediate feedback, guidance and repeated training of laboratory staff, additional inaccuracies were prevented. CONCLUSIONS: Quality control guidelines when applied in field laboratories ensured the reliability and validity of final study data. It is essential that sponsors provide adequate resources for implementation of such comprehensive technical assessment and monitoring systems.     A trade-off between thickness and length in the zebra finch sperm mid-piece

    The sperm mid-piece has traditionally been considered to be the engine that powers sperm. Larger mid-pieces have therefore been assumed to provide greater energetic capacity. However, in the zebra finch Taeniopygia guttata, a recent study showed a surprising negative relationship between mid-piece length and sperm energy content. Using a multi-dimensional approach to study mid-piece structure, we tested whether this unexpected relationship can be explained by a trade-off between mid-piece length and mid-piece thickness and/or cristae density inside the mitochondrial helix. We used selective plane illumination microscopy to study mid-piece structure from three-dimensional images of zebra finch sperm and used high-resolution transmission electron microscopy to quantify mitochondrial density. Contrary to the assumption that longer mid-pieces are larger and therefore produce or contain a greater amount of energy, our results indicate that the amount of mitochondrial material is consistent across mid-pieces of varying lengths, and longer mid-pieces are simply proportionately ‘thinner’

    Impact of Glucocorticoid Receptor Gene Polymorphisms on the Metabolic Profile of Adult Patients with the Classical Form of 21-Hydroxylase Deficiency

    Background: CAH patients have an increased risk of cardiovascular disease, and it remains unknown if lifelong glucocorticoid (GC) treatment is a contributing factor. In the general population, glucocorticoid receptor gene (NR3C1) polymorphisms are associated with an adverse metabolic profile. Our aim was to analyze the association between the NR3C1 polymorphisms and the metabolic profile of CAH patients. Methodology: Sixty-eight adult patients (34SV/34SW) with a mean age of 28.4 +/- 9 years received dexamethasone (mean 0.27 +/- 0.11 mg/day) to obtain normal androgen levels. SW patients also received fludrocortisone (50 mu g/day). Metabolic syndrome (MetS) was defined by the NCEP ATPIII criteria and obesity by BMI >= 30 kg/m(2). NR3C1 alleles were genotyped, and association analyses with phenotype were carried out with Chi-square, t-test and regression analysis. Results: Obesity and MetS were observed in 23.5% and 7.3% of patients, respectively, and were not correlated with GC doses and treatment duration. BMI was positively correlated with blood pressure (BP), triglycerides (TG), LDL-c levels and HOMA-IR and inversely correlated with HDL-c levels. BclI and A3669G variants were found in 26.4% and 9.6% of alleles, respectively. Heterozygotes for the BclI polymorphism presented with higher BMI (29 kg/m(2) +/- 5.3 vs. 26 kg/m(2) +/- 5.3, respectively) and waist circumference (89 cm +/- 12.7 vs. 81 cm +/- 13, respectively) compared to wild-type subjects. Hypertension was found in 12% of patients and heterozygotes for the BclI polymorphism presented higher systolic BP than wild type subjects. Low HDL-c and high TG levels were identified in 30% and 10% of patients, respectively, and were not associated with the NR3C1 polymorphisms. A3669G carriers and non-carriers did not differ. Conclusion: In addition to GC therapy, the BclI GR variant might play an important role in obesity susceptibility in CAH patients.     Microrheology with an anisotropic optical trap

    Microrheology with optical tweezers (MOT) measurements are usually performed using optical traps that are close to isotropic across the plane being imaged, but little is known about what happens when this is not the case. In this work, we investigate the effect of anisotropic optical traps on microrheology measurements. This is an interesting problem from a fundamental physics perspective, but it also has practical ramifications because in 3D all optical traps are anisotropic due to the difference in the intensity distribution of the trapping laser along axes parallel and perpendicular to the direction of beam propagation. We find that attempting viscosity measurements with highly anisotropic optical traps will return spurious results, unless the axis with maximum variance in bead position is identified. However, for anisotropic traps with two axes of symmetry such as traps with an elliptical cross section, the analytical approach introduced in this work allows us to explore a wider range of time scales than those accessible with symmetric traps. We have also identified a threshold level of anisotropy in optical trap strength of ~ 30 %, below which conventional methods using a single arbitrary axis can still be used to extract valuable microrheological results. We envisage that the outcomes of this study will have important practical ramifications on how all MOT measurements should be conducted and analysed in future applications

    Sperm gatekeeping : 3D imaging reveals a constricted entrance to zebra finch sperm storage tubules

    Females across many internally fertilizing taxa store sperm, often in specialized storage organs in their reproductive tracts. In birds, several hundred sperm storage tubules exist in the utero-vaginal junction of the oviduct, and there is growing evidence that sperm storage in these tubules is selective. The mechanisms underlying female sperm storage in birds remain unknown because of our limited ability to make three-dimensional, live observations inside the large, muscular avian oviduct. Here, we describe a new application of fluorescence selective plane illumination microscopy to optically section oviduct tissue from zebra finch Taeniopygia guttata females label free by harnessing tissue autofluorescence. Our data provide the first description of the three-dimensional structure of sperm storage organs in any vertebrate to the best of our knowledge and reveal the presence of gate-like constricted openings that may play a role in sperm selection

    OptoRheo: Simultaneous in situ micro-mechanical sensing and 3D imaging of live cell cultures

    Biomechanical cues from the extracellular matrix (ECM) are essential for directing many cellular processes, from normal development and repair, to disease progression. To better understand cell-matrix interactions, we have developed a new instrument named ‘OptoRheo’ that combines light sheet fluorescence microscopy with particle tracking microrheology. OptoRheo lets us image cells in 3D as they proliferate over several days while simultaneously sensing the mechanical properties of the surrounding extracellular and pericellular matrix at a sub-cellular length scale. OptoRheo can be used in two operational modalities (with and without an optical trap) to extend the dynamic range of microrheology measurements. We corroborated this by characterising the ECM surrounding live breast cancer cells in two distinct culture systems, cell clusters in 3D hydrogels and spheroids in suspension culture. This cutting-edge instrument will transform the exploration of drug transport through complex cell culture matrices and optimise the design of the next-generation of disease models
