    Physicochemical Quality, Antioxidant Capacity and Nutritional Value in Tuberous Roots of Some Wild Dahlia Species

    The aim of this research was to evaluate the physicochemical quality, antioxidant capacity and nutritional value in tuberous roots of some wild dahlia species. The experiment was carried out in the Department of Plant Science of the Autonomous University Chapingo, Mexico. Plants were established in a randomized complete block design with five replications. The total soluble solids (TSS), titratable acidity (TA), vitamin C (VC), total phenols (TP), antioxidant capacity (AC), inulin and its proximate composition were evaluated. Among the materials analyzed, the most outstanding wild species were Dahlia campanulata, D. coccinea and D. brevis, where D. campanulata stood out for its concentration of VC (0.05 mg 100 g-1), AC (1.88 mg VCEAC g-1), inulin, DM and TC (72.25, 24.38 and 88.37%, respectively), however, the inulin content was similar to D. coccinea (66.17%), which was also outstanding with respect to the content of TP (1.74 mg GA g-1). Likewise, D. brevis presented the highest values of RFi (5.49%) and A (78.42%). According to our results, the tuberous roots of wild dahlia species can be used as food, as well as being a source of selection of traits of nutraceutic interest for genetic improvement

    Alzheimer-Like Cell Alterations after Vanadium Pentoxide Inhalation

    Vanadium (V), a widely distributed transition metal, has been considered toxic, which depends on the valence of the compound. V pentoxide (V2O5) is considered the most harmful. Its long-term exposure produces neurotoxicity. Mice exposed to inhaled V2O5 displayed less tubulin+ in testicular cells and dendritic spines loss, cell death, and CA1 neuropil modifications, considered as the result of V interaction with the cytoskeleton, which made us suppose that V2O5 inhalation could initiate CA1 cell alterations comparable to what happen in the brains of Alzheimer disease (AD) patients. This study intends to demonstrate pyramidal CA1 cytoskeletal changes in rats which inhaled V2O5. Twenty rats were exposed to V2O5 0.02 M one hour, three times a week for several months. Our findings showed that V2O5-exposed rats had cell death that reached 56,57% after six months; we also observed collapsed strong argyrophilic nuclei and characteristic flame-shaped somas in all V2O5-exposed animals hippocampus CA1 compared to controls. We also found somatodendritic deformations. Neurite’s cytoskeleton exhibited visible thickening and nodosities and prominent dendritic spine loss. Our results demonstrate that V2O5 induces AD-like cell death with evident cytoskeletal and synaptic alterations

    Diversidad genética y distribución regional de cepas de Mycobacterium bovis del ganado en México

    The molecular fingerprints of 878 isolates of Mycobacterium bovis from cattle, mostly dairy cattle, collected from cattle between 2009 and 2010 in different regions of Mexico were obtained by spoligotyping. Seventy-two percent (72 %) of the spoligotypes fell into nine clusters, and 27 % of the isolates fell into only two spoligotypes; 149 were orphan spoligotypes. The two predominant spoligotypes, arbitrarily designated as SP1 and SP2, were found in almost all States in Mexico, especially in central Mexico, where a concentration of dairy cattle is known. In spite of the wide distribution of spoligotypes observed, some show high regional preference, especially those in geographically distant regions. Only a few spoligotypes show patterns completely different from those shown by the most frequent spoligotypes, suggesting strange sources of infection or the formation of new genetic lines derived from non-lethal mutations. Most States with predominantly high dairy cattle populations showed similar spoligotypes, suggesting exchange of animals between regions. Some spoligotypes are common to dairy and beef cattle, suggesting transmission between populations, most probably due to the movement of dairy cattle to non-dairy regions.Se obtuvieron patrones moleculares (espoligotipos) de 878 aislados de Mycobacterium bovis de ganado de diferentes regiones de México entre los años 2009 y 2010. Setenta y dos por ciento (72 %) de los espoligotipos cayeron en nueve grupos y 27 % de los aislados dentro de sólo dos espoligotipos; 149 fueron espoligotipos individuales. Los dos espoligotipos predominantes, arbitrariamente identificados como SP1 y SP2, se distribuyen en la mayor parte del territorio nacional, en especial en la zona centro de México en ganado especializado en producción de leche. A pesar de la amplia distribución geográfica de los espoligotipos de mayor frecuencia, algunos muestran cierta localización, en especial los encontrados en zonas geográficas distantes, como es el caso de Chihuahua y Baja California. Aunque pocos, algunos espoligotipos muestran patrones moleculares distintos a los mostrados por los espoligotipos de mayor frecuencia, sugiriendo fuentes de infección desconocida. La mayoría de los Estados con ganadería predominantemente lechera muestran espoligotipos comunes, lo que sugiere intercambio regional frecuente de ganado. Algunos espoligotipos son comunes en ganado para leche y ganado para carne, lo que sugiere transmisión entre estas dos poblaciones; se desconoce, sin embargo, si los animales de carne infectados provienen de explotaciones extensivas o si son de engordas ubicadas dentro de la explotación lechera. Se propone la tipificación rutinaria de aislados de M. bovis obtenidos en todos los laboratorios de diagnóstico y mejorar la captura de información epidemiológica de los casos, para hacer mejores conclusiones epidemiológicas de la distribución espacial de las cepas de este agente en el territorio nacional

    Resumen: Objetivo: Determinar las características epidemiológicas de adultos con insuficiencia cardiaca aguda admitidos en un hospital universitario. Métodos: Estudio de cohorte retrospectivo, descriptivo. Revisión de historias clínicas de mayores de 18 años hospitalizados entre julio y diciembre de 2015 en un hospital de Medellín, Colombia. Resultados: Se incluyeron 361 pacientes con insuficiencia cardíaca aguda. 193 (53,4%) fueron mujeres, cuya mediana de edad fue 76 años. 183 (50,6%) tenían fracción de eyección (FEVI) reducida, 19 (5,2%) FEVI intermedia y 148 (40,9%) FEVI preservada. El tratamiento farmacológico previo al ingreso incluía IECA/ARA2 en 253 (70%) pacientes, betabloqueador en 212 (58,7%) y espironolactona en 92 (25,4%). La principal causa de falla cardíaca aguda fue la presencia de taqui-bradiarritmias (17,5%), seguida de infección (17,2%) y exacerbación de neumopatía crónica (16,3%). La clasificación clínica de la descompensación fue Stevenson B en 335 (92,7%) pacientes, Stevenson C en 20 (5,5%) y Stevenson L en 6 (1,6%). La mediana de hospitalización fue 6 días (4-9 días). 30 pacientes (8,3%) fallecieron, 50% por infecciones asociadas a la descompensación cardiaca y 20% por síndrome coronario agudo. Conclusión: Se hallaron similitudes con estudios internacionales, pero mayor mortalidad vinculada principalmente a infección como factor precipitante de descompensación. Se caracterizaron factores desencadenantes y etiología, datos útiles en la práctica clínica. Hubo alta carga de comorbilidades, cuya descompensación impactó de manera significativa en la agudización de la falla cardiaca. El subgrupo con fracción de eyección intermedia presentó particularidades que ameritan mayor caracterización. Abstract: Objective: To determine the epidemiological characteristics of adults with acute heart failure admitted to a University Hospital. Method: A retrospective, descriptive cohort study conducted by reviewing the medical notes of patients over 18 years-old and admitted between July and December 2015 to a hospital in Medellin, Colombia. Results: The study included 361 patients with acute heart failure, with a mean age of 76 years, and of whom 193 (53.4%) were women. A reduced ejection fraction (LVEF) was observed in 183 (50.6%) patients, 19 (5.2%) with an intermediate LVEF, and 148 (40.9%) with normal LEVF. The pharmacological treatment prior to admission included angiotensin-converting-enzyme (ACE) inhibitors / angiotensin II receptor antagonists (ARA2) in 253 (70%) patients, a beta-blocker in 212 (58.7%), and spironolactone in 92 (25.4%). The main cause of acute heart failure was the presence of tachy-brady-arrhythmias (17.5%), followed by infection (17.2%), and exacerbation of chronic pulmonary disease (16.3%). The clinical classification of the decompensation was Stevenson B in 335 (92.7%) patients, Stevenson C in 20 (5.5%), and Stevenson L in 6 (1.6%). The mean admission time was 6 (4-9) days. There were 30 (8.3%) deaths, 50% due to infections associated with cardiac decompensation and 20% due to acute coronary syndrome. Conclusion: Similarities were found with international studies, but there was a higher mortality mainly linked to infection as a decompensation precipitating factor. The triggering factors and aetiology are presented, which are useful data in clinical practice. There was a high level of comorbidities, and their decompensation had a significant impact on the exacerbation of heart failure. The sub-group with the intermediate ejection fraction had features that require further characterisation.     Human Amniotic Membrane Mesenchymal Stem Cell-Synthesized PGE<sub>2</sub> Exerts an Immunomodulatory Effect on Neutrophil Extracellular Trap in a PAD-4-Dependent Pathway through EP2 and EP4

    Human amniotic membrane mesenchymal stem cells (hAM-MSC) secrete a myriad of components with immunosuppressive activities. In the present research, we aimed to describe the effect of prostaglandin E2 (PGE2) secreted by hAM-MSCs on neutrophil extracellular trap (NET) release and to characterize the role of its receptors (EP2/EP4) in PAD-4 and NFκB activity in neutrophils. Human peripheral blood neutrophils were ionomycin-stimulated in the presence of hAM-MSC conditioned medium (CM) treated or not with the selective PGE2 inhibitor MF-63, PGE2, EP2/EP4 agonists, and the selective PAD-4 inhibitor GSK-484. NET release, PAD-4, and NFκB activation were analyzed. Ionomycin induced NET release, which was inhibited in the presence of hAM-MSC-CM, while CM from hAM-MSCs treated with MF-63 prevented NET release inhibition. PGE2 and EP2/EP4 agonists, and GSK-484 inhibited NET release. EP2/EP4 agonists and GSK-484 inhibited H3-citrullination but did not affect PAD-4 protein expression. Finally, PGE2 and EP2/EP4 agonists and GSK-484 increased NFκB phosphorylation. Taken together, these results suggest that hAM-MSC exert their immunomodulatory activities through PGE2, inhibiting NET release in a PAD-4-dependent pathway.
